Lipoxin A4 (LXA4) is a biologically active compound produced from arachidonic acid via interactions of lipoxygenases. Incubation of LXA4 either with human neutrophils or with the neutrophil microsomes leads to formation of a polar compound on a reverse-phase high-performance liquid chromatography. We have identified the metabolite as 20-hydroxy-LXA4, a novel metabolite of arachidonic acid, on the basis of ultraviolet spectrometry and gas chromatography-mass spectrometry. The LXA4 omega-hydroxylation requires both molecular oxygen and NADPH, and is inhibited by carbon monoxide, by antibodies raised against NADPH-cytochrome P-450 reductase, or competitively by leukotriene B4 (LTB4) and LTB5, substrates of LTB4 omega-hydroxylase. These findings indicate that the formation of 20-hydroxy-LXA4 is catalyzed by a neutrophil cytochrome P-450, the LTB4 omega-hydroxylase.  相似文献

To examine the expression of topoisomerase I and topoisomerase II in primary lung cancer specimens at mRNA level, we carried out Northern blot analysis. As for topoisomerase I expression, there was no remarkable difference between lung cancer specimens and non-cancerous lung tissues. On the other hand, we could detect topoisomerase II mRNA in almost all lung cancer specimens, but not in non-cancerous tissues. By Southern blot analysis, we could not detect large deletion nor rearrangement in DNA level. These results suggest that the expression of topoisomerase II is highly increased in lung cancer at mRNA level and drugs against topoisomerase II might be more tumor-specific than those against topoisomerase I.  相似文献

Transfer rates of copper and hydrogen ion through a cation exchange membrane were measured experimentally for the systems of Cu/H-SO4 and Cu/H-NO3. Transfer rates of anions in the membrane were also measured, and they were negligible compared to those of the cations. Cupric ion and hydrogen ion transfer in a ratio of 1:2 and in opposite directions. The self-diffusion coefficient of cupric ion in the membrane is obtained from the transfer rate of copper in high concentrations of sulfuric or nitric acid solutions using the equilibria data obtained in our previous work. The coefficient of the hydrogen ion is similarly obtained.  相似文献

Full orbit-following calculation is performed for the final orbit of the lost alpha particles, showing some orbits escaping from the last closed flux surface could be detected by a concealed lost ion probe (CLIP) installed under the shadow of the original first wall surface. While both passing and trapped orbits hit the same wall panel, detecting a trapped orbit by the CLIP is easier than detecting passing orbits. Whether the final orbit is detected or not is determined by the position of the reflection point. The CLIP successfully detects the trapped orbits, which are reflected before they hit to a first wall. Then the pitch angles of the orbits at the CLIP are close to and smaller than 90°. Optimization of the position of the CLIP in terms of broader detection window is investigated.  相似文献

A tangential Fast Ion Charge eXchange Spectroscopy is newly applied on a Large Helical Device (LHD) for co/countercirculating fast ions, which are produced by high energy tangential negative-ion based neutral beam injection. With this new observation geometry, both the tangential-neutral beam (NB) and a low-energy radial-NB based on positive ions can be utilized as probe beams of the measurement. We have successfully observed Doppler-shifted H-alpha lights due to the charge exchange process between the probing NB and circulating hydrogen ions of around 100 keV in LHD plasmas.  相似文献

Keggin ions (PW12O403− (PW12), SiW12O404− (SiW12), H2W12O406− (H2W12)) and TiO2 hybrid thin films were prepared using the layer-by-layer method. Their photocatalytic activities were investigated using gaseous 2-propanol decomposition. All films were transparent in the visible wavelength range. For 2-propanol decomposition, H2W12 was the most effective for the combination with TiO2 despite having the smallest TiO2 deposition amount. The photocatalytic activity of the PW12–TiO2 hybrid film was increased 2.3 times by visible light with UV illumination. This increase was less remarkable for hybrid films of other Keggin ions, suggesting that the visible light excitation of reduced PW12 plays an important role in the enhancement of 2-propanol decomposition.  相似文献

This paper reports results of full scale testing to further explore potential GCL damage mechanisms in earth dam retrofit applications in seismically active areas; in particular, to a) investigate whether shear displacements could reduce the magnitude of GCL panel overlap during earthquake shaking; b) explore the influence of gravel particles on GCL thickness at localised point of contact; and c) observe the consequences of an accidental exposure of an uncovered GCL to short duration rainfall in terms of moisture content and effects during subsequent compaction. The results of these experiments indicate that even under severe shaking no movements were detected at the GCL panel overlap. Whereas gravel particles were observed to locally reduce the thickness of the GCL to 2.2 mm, no plowing of the particle into the GCL occurred due to a lack of shear displacement at the interface, resulting in no localised internal erosion through the barrier. Furthermore, hydration of GCL panels during construction due to surface wetting was observed to result in a state of hydration less than its post-construction state. These results indicate that although each of the three GCL damage mechanisms cannot be ruled out to ever be relevant in practice, the performance of the GCL retrofitted earth dam tested was satisfactory under even severe Level 2 earthquake shaking, and suggests that the retrofitting of small earth dams with GCLs is a promising strategy to improve their static and seismic resistance.  相似文献

Human–robot interaction during general service tasks in home or retail environment has been proven challenging, partly because (1) robots lack high-level context-based cognition and (2) humans cannot intuit the perception state of robots as they can for other humans. To solve these two problems, we present a complete robot system that has been given the highest evaluation score at the Customer Interaction Task of the Future Convenience Store Challenge at the World Robot Summit 2018, which implements several key technologies: (1) a hierarchical spatial concepts formation for general robot task planning and (2) a mixed reality interface to enable users to intuitively visualize the current state of the robot perception and naturally interact with it. The results obtained during the competition indicate that the proposed system allows both non-expert operators and end users to achieve human–robot interactions in customer service environments. Furthermore, we describe a detailed scenario including employee operation and customer interaction which serves as a set of requirements for service robots and a road map for development. The system integration and task scenario described in this paper should be helpful for groups facing customer interaction challenges and looking for a successfully deployed base to build on.  相似文献

Integrin activation and sebsequent ligand binding to it are regulated by intracellular mechanisms called inside-out signaling, which are not fully understood and are accompanied by dynamic structural changes of the integrin molecule itself. A monoclonal antibody recognizing a conformation-dependent epitope on human beta1 integrin was produced and characterized in detail. This antibody, AG89, reacted with human integrin beta1 chain regardless of the alpha subunit. AG89 can recognize resting state beta1 integrin on the cells, but the reactivity is increased approximately 2-fold upon integrin activation by activating anti-beta1 antibodies and approximately 3-fold by Mn2+. Furthermore, occupation of the ligand-binding pocket by a soluble ligand (RGD peptide for alpha(v)beta1 and CS-1 peptide for alpha4beta1) resulted in maximum binding of AG89, indicating that the epitope for AG89 is exposed during the conformational changes of beta1 integrin upon activation/ligation. Epitope mapping by using interspecies chimeric beta1 revealed that the epitope for AG89 lies within residues 426-587, which corresponds to the cysteine-rich repeat structure located in the middle of the beta1 chain. The fact that binding of AG89 itself could activate the resting beta1 integrin indicates that exposure of the AG89 epitope in the membrane-proximal stalk-like domain and "opening" of the ligand-binding pocket at the outermost domain are physically linked. We propose that the integrin "signaling" is mediated by this direct physical transduction of conformational information along the integrin molecule.  相似文献

Rationally manipulating the functional substituents plays a crucial role in tuning the luminescence and lasing properties of organic gain media. Herein, a cyanophenyl-moiety, which exhibits relatively weaker electron affinity, is connected to 2,6-dicarbonitrile diphenyl-1λ5-phosphinine (DCNP) via para-linking. Resultantly, the appreciated locally-excited characteristics ensuring a large oscillator strength and high radiative rate can be reserved in DCNP-4-(4-cyanophenyl) (DCNP-pCN). Interestingly, the weak charge-transfer state from the relative donor (D)/acceptor (A) interplay enables small singlet-triplet splitting (ΔEST ≈ 0.45 eV). Thus the triplets generated on DCNP-pCN can be efficiently scavenged by 4,4'-bis[(N-carbazole)styryl]biphenyl (BSBCz), which is used as the host with a lower-lying triplet energy level for DCNP-pCN. Moreover, benefitting from the mediation between the conjugated length extension and weak D/A interplay, the emission spectrum cannot be largely shifted, which can effectively suppress the overlap between the lasing emission of DCNP-pCN and the excited-state absorption of BSBCz, thereby avoiding detrimental singlet-triplet annihilation. Thus, high-quality distributed feedback lasings with ≈2.0 μJ cm−2 thresholds are achieved, and the organic light-emitting diodes exhibit external quantum efficiency exceeding 2.0% without efficiency rolloff under high current injection, indicating the potential for electrical-pumping organic lasings.  相似文献
