Novel antiviral nanotherapeutics, which may inactivate the virus and block it from entering host cells, represent an important challenge to face viral global health emergencies around the world. Using a combination of bioorthogonal copper-catalyzed 1,3-dipolar alkyne/azide cycloaddition (CuAAC) and photoinitiated thiol–ene coupling, monofunctional and bifunctional peptidodendrimer conjugates were obtained. The conjugates are biocompatible and demonstrate no toxicity to cells at biologically relevant concentrations. Furthermore, the orthogonal addition of multiple copies of two different antiviral peptides on the surface of a single dendrimer allowed the resulting bioconjugates to inhibit Herpes simplex virus type 1 at both the early and the late stages of the infection process. The presented work builds on further improving this attractive design to obtain a new class of therapeutics.  相似文献

Background: Proprotein convertase subtilisin kexin type 9 inhibitors (PCSK9i) lower LDL-cholesterol and slow atherosclerosis preventing cardiovascular events. While it is known that circulating PCSK9 enhances platelet activation (PA) and that PCSK9i reduce it, the underlying mechanism is not still clarified. Methods: In a multicenter before–after study in 80 heterozygous familial hypercholesterolemia (HeFH) patients on treatment with maximum tolerated statin dose ± ezetimibe, PA, soluble-NOX2-derived peptide (sNOX2-dp), and oxidized-LDL (ox-LDL) were measured before and after six months of PCSK9i treatment. In vitro study investigates the effects of plasma from HeFH patients before and after PCK9i on PA in washed platelets (wPLTs) from healthy subjects. Results: Compared to baseline, PCSK9i reduced the serum levels of LDL-c, ox-LDL, Thromboxane (Tx) B2, sNOX2-dp, and PCSK9 (p < 0.001). The decrease of TxB2 correlates with that of ox-LDL, while ox-LDL reduction correlated with PCSK9 and sNOX2-dp delta. In vitro study demonstrated that wPLTs resuspended in plasma from HeFH after PCSK9i treatment induced lower PA and sNOX2-dp release than those obtained using plasma before PCSK9i treatment. This reduction was vanished by adding ox-LDL. ox-LDL-induced PA was blunted by CD36, LOX1, and NOX2 inhibition. Conclusions: PCSK9i treatment reduces PA modulating NOX2 activity and in turn ox-LDL formation in HeFH patients.  相似文献

The surface texturing of ceramics is generally performed through acid-based etching and machining; however, laser texturing may be considered as a more precise, reproducible and eco-friendly process. Furthermore, laser ablation may be used to produce complex patterns on ceramic surfaces, thus offering new surface engineering opportunities. The studies so far conducted on this topic have mainly been application-driven, and since a wide variety of lasers have been used for surface texturing, it is difficult to have a comprehensive understanding of this technique applied to ceramics and ceramic composite materials. Laser texturing requires a great deal of knowledge of the material and the laser source parameters to optimise the process in order to obtain the expected results. It is therefore important to expand the research on the laser texturing of ceramics and CMCs in order to build a relevant amount of literature that can be used to identify the most appropriate parameters for each application. This review provides an overview of most of the technological aspects considered relevant for the laser surface texturing of ceramics and CMCs, and includes the fundamentals of laser-material interactions and a summary of the used equipment and parameters. Furthermore, most of the techniques related to the modifications of surface material induced by a laser are critically reviewed, and the new horizons that are opening up, in the context of the modification of surfaces to improve the performances of materials for several applications, are discussed.  相似文献

Food safety demands calls for new approaches to better understand the safe and hygienic practices of specific sectors in the food supply chain. In the present study, sauerkraut growers were investigated to explore their understanding of food safety and hygiene practices. A combination of quantitative (questionnaire prepared for the study) and qualitative (ten in-depth interviews) methods was used in the study so that topics could be discussed more completely and in greater depth. The results indicated that sauerkraut growers possess a rather good level of knowledge concerning food safety, although some gaps were discovered (e.g., record keeping and the feeling that recording data was an unnecessary burden, inadequate acquaintance with health status requirements, insufficient knowledge of the cabbage production process of food safety professionals, etc.). Some differences were found when comparing the results of the qualitative and quantitative studies. The qualitative analysis highlighted some contradictions (e.g., an attitude toward the production process: “We do not need the HACCP system”; a feeling concerning product safety: “Why the need for data recording?”) and differences between the sauerkraut growers and a professional understanding of good practice guidelines for sauerkraut production. All other determinations concerning food safety knowledge and practices are presented and discussed. The findings can be used as a guide for reorganizing current education programs aimed to improve safe and hygiene practices at the farm level.  相似文献

Climate change challenges water managers and researchers to find sustainable management solutions, in order to avoid undesirable impacts on water resources, environment and water-dependent sectors. Needed are projections into the future for the main driving forces, the resulting pressures on water resources, and quantification of the impacts. Modeling studies can play an important role in investigating, quantifying, and communicating possible impacts of climate change, with account of uncertainty of the results. However, climate change related impacts and a need for adaptation still play a minor role in current river basin management plans that have to comply e.g. with the EU Water Framework Directive (WFD). One important reason is that climate impact assessment is generally done in research institutes, while management plans are designed by practitioners working in national and regional environmental agencies and water supply companies. Knowledge transfer from science to practice and visa versa is often missing. In the present study, we propose a methodology and a case study for model-supported decision making in the water sector applicable to a participatory water resources planning process. The methodology is applied in a case study investigating climate change impacts on water resources. The case study area is the German State of Saxony-Anhalt, where the task was to develop a climate change impact assessment including possible adaptation measures as basis for a federal adaptation directive.  相似文献

The paper by Haewon McJeon and coauthors seeks to estimate the benefits (measured as avoided climate policy costs) of different carbon-free and low-carbon technologies in the context of climate stabilization. The paper does not include a full cost-benefit analysis of these technologies nor does it evaluate what would be needed to bring these technologies about (e.g., in an optimal R&D investment portfolio). Nonetheless, in its main conclusions, the paper provides a sensible analysis and adds to the expanding literature on this extremely relevant topic by exploring a new dimension, that of the interactions among technologies and their combined impact.  相似文献

Reliability of water distribution networks (WDNs) has received much attention in recent years due to progressive aging of infrastructures and climate change. Several reliability indicators, focusing on hydraulic aspects rather than water quality, have been proposed in literature. Reliability is generally assessed resorting to well established methods coupling hydraulic simulations and stochastic techniques that describe the WDNs hydraulic performance and component availability respectively. Two main algorithms are employed to simulate WDNs: the demand driven approach (DDA) that disregards the physical relationship between actual water demand and nodal pressure, and the pressure driven approach (PDA) that explicitly incorporates it. In this paper, we show how the choice of hydraulic solver may affect reliability indicators. We modify existing quantitative indicators at nodal and network level, and define novel indicators to consider water quality aspects. These indicators are evaluated for three example WDNs; discrepancies between results obtained with the two approaches depend on network size, feeding scheme and skeletonization. Results suggest to use with caution the DDA for reliability assessment at both local and global level.  相似文献
