Background: Molecular mechanisms of depression remain unclear. The brain metabolome after antidepressant therapy is poorly understood and had not been performed for different routes of drug administration before the present study. Rats were exposed to chronic ultrasound stress and treated with intranasal and intraperitoneal clomipramine. We then analyzed 28 metabolites in the frontal cortex and hippocampus. Methods: Rats’ behavior was identified in such tests: social interaction, sucrose preference, forced swim, and Morris water maze. Metabolic analysis was performed with liquid chromatography. Results: After ultrasound stress pronounced depressive-like behavior, clomipramine had an equally antidepressant effect after intranasal and intraperitoneal administration on behavior. Ultrasound stress contributed to changes of the metabolomic pathways associated with pathophysiology of depression. Clomipramine affected global metabolome in frontal cortex and hippocampus in a different way that depended on the route of administration. Intranasal route was associated with more significant changes of metabolites composition in the frontal cortex compared to the control and ultrasound groups while the intraperitoneal route corresponded with more profound changes in hippocampal metabolome compared to other groups. Since far metabolic processes in the brain can change in many ways depending on different routes of administration, the antidepressant therapy should also be evaluated from this point of view.  相似文献

In this study, we designed and developed novel poly(ε-caprolactone) (PCL)-based biomaterials, for use as bone scaffolds, through modification with both biphasic calcium phosphate (BCP), to impart bioactive/bioresorbable properties, and with silver nitrate, to provide antibacterial protection against Staphylococcus aureus, a microorganism involved in prosthetic joint infections (PJIs). Field emission scanning electron microscopy (FESEM) showed that the samples were characterized by square-shaped macropores, and energy dispersive X-ray spectroscopy analysis confirmed the presence of PCL and BCP phases, while inductively coupled plasma–mass spectrometry (ICP–MS) established the release of Ag+ in the medium (~0.15–0.8 wt% of initial Ag content). Adhesion assays revealed a significant (p < 0.0001) reduction in both adherent and planktonic staphylococci on the Ag-functionalized biomaterials, and the presence of an inhibition halo confirmed Ag release from enriched samples. To assess the potential outcome in promoting bone integration, preliminary tests on sarcoma osteogenic-2 (Saos-2) cells indicated PCL and BCP/PCL biocompatibility, but a reduction in viability was observed for Ag-added biomaterials. Due to their combined biodegrading and antimicrobial properties, the silver-enriched BCP/PCL-based scaffolds showed good potential for engineering of bone tissue and for reducing PJIs as a microbial anti-adhesive tool used in the delivery of targeted antimicrobial molecules, even if the amount of silver needs to be tuned to improve osteointegration.  相似文献

Rutin, one of the major flavonoids found in an assortment of plants, was reported to act as a sun protection factor booster with high anti-UVA defense, antioxidant, antiaging, and anticellulite, by improvement of the cutaneous microcirculation. This research work aimed at evaluating the rutin in vitro release from semisolid systems, in vertical diffusion cells, containing urea, isopropanol and propylene glycol, associated or not, according to the factorial design with two levels with center point. Urea (alone and in association with isopropanol and propylene glycol) and isopropanol (alone and in association with propylene glycol) influenced significant and negatively rutin liberation in diverse parameters: flux (microg/cm(2).h); apparent permeability coefficient (cm/h); rutin amount released (microg/cm(2)); and liberation enhancement factor. In accordance with the results, the presence of propylene glycol 5.0% (wt/wt) presented statistically favorable to promote rutin release from this semisolid system with flux = 105.12 +/- 8.59 microg/cm(2).h; apparent permeability coefficient = 7.01 +/- 0.572 cm/h; rutin amount released = 648.80 +/- 53.01 microg/cm(2); and liberation enhancement factor = 1.21 +/- 0.07.  相似文献

CaTiO3 powders were synthesized using both a polymeric precursor method (CTref) and a microwave-assisted hydrothermal (CTHTMW) method in order to compare the chemical and physical properties of the perovskite-based material as a function of the synthesis method. To this end, X-ray diffraction, Raman spectroscopy, inductively coupled plasma atomic emission spectroscopy and experimental Ti and Ca K-edge X-ray absorption near-edge structure spectroscopy, as well as measurements of photoluminescence (PL) emission, were used to characterize the typical bottom-up process of the CaTiO3 perovskite phase at different times. Detailed Rietveld refinements show a random polycrystalline distortion in the powder structure, which can be associated with the tilting (α angle < O–Ti–O) between adjacent TiO6 octahedra (intermediate range) for CTHTMW samples and an intrinsic TiO6 distortion (short range) in relation to the polymeric precursor CTref sample. These properties were further investigated by first-principles calculations based on the density functional theory at the B3LYP level. The relationship between this tilting on the PL profile is highlighted and discussed. Thus, a structural model derived from both experimental results and theoretical simulations reveals a close relationship between this tilting and the presence of intermediate energy states within the band gap which are mainly responsible for PL emissions.  相似文献

This study concerns the partial dehydration of cherry tomatoes (Lycopersicon esculentum, var. Shiren) to obtain a product with 25% initial water content. Two kinds of dried tomatoes were obtained using a forced air oven at 40, 60 and 80 °C for different lengths of treatment. The first type was dehydrated after immersion of the fresh tomatoes in an aqueous solution of citric acid, sodium and calcium chloride (10:10:24 g/l); the second was obtained with no pre-treatment. The products were characterised by measuring their CIE L*a*b* colour parameters and levels of l-ascorbic acid, lycopene and β-carotene to evaluate thermal damage during processing under the different conditions. Moreover, water activity and the formation of 5-hydroxymethylfurfural were also determined as an index of sugar heat degradation. Treatment with a dipping solution protected both the nutritional and chemical qualities of the partially dried cherry tomatoes. Temperature was directly related to browning, ascorbic acid loss and HMF formation, while no clear influence could be found for carotenoid degradation.  相似文献

The aim of this study was to determine whether the intrinsic mechanism of apoptosis is involved in the death of germ cells in Robertsonian (Rb) heterozygous adult male mice. Testes from 5-month-old Rb heterozygous CD1 x Milano II mice were obtained and compared with those from homozygous CD1 (2n=40) and Milano II (2n=24) mice. For histological evaluation of apoptosis, TUNEL labelling and immunohistochemistry were used to localise Bax and cytochrome c. Expression of calbindin D(28k) (CB), an anti-apoptotic molecule, was also analysed by immunohistochemistry and immunoblotting. Testicular ultrastructure was visualised by electron microscopy. Morphology and cell associations were abnormal in the Rb heterozygous seminiferous epithelium. An intense apoptotic process was observed in tubules at stage XII, mainly in metaphase spermatocytes. Metaphase spermatocytes also showed Bax and cytochrome c redistributions. Mitochondria relocated close to the paranuclear region of spermatocytes. CB was mainly expressed in metaphase spermatocytes, but also in pachytene spermatocytes, spermatids and Sertoli cells at stage XII. The co-localisation of CB and TUNEL labelling was very limited. Sixty per cent of metaphase spermatocytes were apoptotic and calbindin negative, while 40% were calbindin positive without signs of apoptosis. Ten per cent of the Bax- and cytochrome c-positive cells were also calbindin positive. These data suggest that apoptosis of the germ cells in heterozygous mice occurs, at least in part, through a mitochondrial-dependent mechanism. Calbindin overexpression might prevent or reduce the apoptosis of germ cells caused by Rb heterozygosity, which could partially explain the subfertility of these mice.  相似文献

The molybdenum(VI)‐catalysed oxybromination of ethynylbenzene was performed with hydrogen peroxide as oxidant and potassium bromide as source of bromine, in a two‐phase water/solvent system, where solvent was either dichloromethane or an ionic liquid. The selectivity was toward 1,2‐dibromostyrene when performed in water/dichloromethane. In ionic liquids better yields and shorter reaction times were obtained that, together with the complete ethynylbenzene conversion and the preferred formation of α,α‐dibromoacetophenone, make the reaction synthetically useful.  相似文献
