The dramatic and apparently curative effect of penicillin for the treatment of acute syphilis led to follow-up studies for only comparatively brief periods, and the acceptance of the long-term benefit of penicillin has rested on uncontrolled clinical impressions. More certainty about the efficacy of penicillin was sought by a follow-up review of 251 patients treated between March 1944 and December 1950 under the Penicillin Study of the Office of Scientific Research and Development (OSRD) and continued under U. S. Public Health Service after World War II. Eighty-eight patients were interviewed and examined. Telephone conversation or correspondence was had with 43 subjects; an additional nine are known to be living but did not respond to letters. Thirty-two patients died greater than or equal to 20 years after treatment, and 21 patients died within less than 20 years of treatment. Fifty-eight patients could not be found. Treatment failures were documented. Syphilis was not shown to be the cause of disability or death, except for a patient with meningovascular syphilis who died soon after initial treatment. Disabilities recorded and deaths documented revealed only diseases common to any middle-aged population. The outcomes of 17 pregnancies of women treated for acute syphilis were documented. Blood samples obtained from the 88 subjects examined were tested at the Center for Disease Control (Atlanta, Ga.); the results are recorded and discussed. Methods for locating the patients are described, and the psychosocial findings for the 88 patients interviewed are presented. The study has confirmed the clinical impressions of the therapeutic effectiveness of penicillin, which have been accepted for greater than 30 years. 相似文献

A radioimmunoassay procedure for the determination of PGE1, PGE2, and PGF2alpha is presented. The procedure involves the pre-precipitation of each prostaglandin specific antiserum with the precipitating antisera (ARGG), and the use of these antisera mixtures in assaying for PGE1, PGE2, and PGF2alpha. Applicability of the methods to unextracted plasma, serum and myocardial homogenate has been demonstrated through tests of specificity, recovery, reproducability and parallelism. A mathematical correction for cross-reactivity between PGE1 and PGE2, and their opposing antisera is given. To demonstrate the utility of the methodology in differentiation of experimental variables, prostaglandin concentrations in unincubated serum, incubated serum, and the rate of prostaglandin production in serum of dogs are given. 相似文献

One challenge for automotive hybrid traction application is the use of high power IGBT modules that can withstand high ambient temperatures, from 90 °C to 120 °C, for reliability purpose. The paper presents ageing tests of 600 V–200 A IGBT modules subjected to power cycling with 60 °C junction temperature swings at 90 °C ambient temperature. Failure modes are described and obtained results on the module characteristics are detailed. Especially, physical degradations are described not only at the package level, like solder attach delaminations, but also at the chip level, with a shift on electrical characteristics such as threshold voltage. Finally, numerical investigations are performed in order to assess the thermal and thermo-mechanical constraints on silicon dies during power cycling and also to estimate the effect of ambient temperature on the mechanical stresses. 相似文献

Low dissociation or reverse rate constants of single-step or multistep complex formation equilibria are usually obtained with reduced precision from standard stopped-flow binding experiments by determination of the intercept of the concentration dependence of k(obs). Large and fast concentration jumps, based on two different step-motor-driven mixing setups, are performed with 60-300-fold dilutions that allow the precise, convenient, and independent determination of dissociation rate constants in the range of approximately 0.1-100 s(-1) in a single stopped-flow dissociation experiment. A theoretical basis is developed for the design and for the evaluation of such dilution experiments by considering the rebinding occurring during dissociation. The kinetics of three chemical systems are investigated, the binding of Mg2+ to 8-hydroxyquinoline as well as of Ca2+ and K+ to the cryptand [2.2.2], by carrying out standard stopped-flow binding as well as dissociation experiments employing various dilution factors. The maximum-likelihood (ML) expectation-maximization (EM) [ML-EM] algorithm is being widely used for image reconstruction in positron emission tomography. The algorithm is strictly valid if the data are Poisson distributed. However, it is also often applied to processed sinograms that do not meet this requirement. This may sometimes lead to suboptimal results: streak artifacts appear and the algorithm converges toward a lower likelihood value. As a remedy, we propose two simple pixel-by-pixel methods [noise equivalent counts (NEC)-scaling and NEC-shifting] in order to transform arbitrary sinogram noise into noise which is approximately Poisson distributed (the first and second moments of the distribution match those of the Poisson distribution). The convergence speed associated with both transformation methods is compared, and the NEC-scaling method is validated with both simulations and clinical data. These new methods extend the ML-EM algorithm to a general purpose nonnegative reconstruction algorithm. 相似文献

In situ air sparging (IAS) has been used since the mid-1980s, but few carefully designed field studies have been performed to evaluate its effectiveness. In this study, 27 discrete monitoring points (MPs) were installed at a gasoline-contaminated site to investigate the efficacy of IAS. Each MP was instrumented with a pressure transducer and a Technalithics dissolved oxygen (DO) probe, and located so they could be used to characterize subsurface changes in total head and DO with depth, distance and orientation around a central injection well. Because the blower over-heated and automatically shut down after approximately 30 min and short-circuiting of air into two MPs occurred within 2 min, the study was designed as three sets of three 30-min trials. Longer trials would not have yielded different nor more insightful results. A volume of soil was not oxygenated during any injection. Instead, air traveled directly to at least four of seven different MPs during eight of the nine trials, probably as a result of an air bubble forming beneath a confining layer. The order of air arrival at the MPs varied during the first few trials, but once a preferential pathway was established, it did not collapse between trials and provided the shortest distance to the vadose zone during subsequent trials. Oxygen uptake rates estimated for MPs that received air during any trial exceeded the consumption rates of the Technalithics DO probes, and indicate that the probes could be used for estimating oxygen transfer during system operation or for oxygen uptake measurements during shut-down tests.The data from the monitoring system indicate that IAS is infeasible for remediation of soil and groundwater at this site due to its low horizontal hydraulic conductivity. BACKGROUND: The vitamin C and polyphenol content of apples constitute quality and nutritional parameters of great interest for consumers, especially in terms of health. They are commonly measured using laborious reference methods. The purpose of this study was to evaluate the potential of near‐infrared (NIR) spectroscopy as a rapid and non‐destructive method to determine the sugar, vitamin C and total polyphenol content in apples. RESULTS: The quality parameters of more than 150 apple genotypes were analyzed using NIR and reference methods. The results obtained using the least squares support vector machine regression method showed good to very good prediction performance. Low standard error of prediction values, in addition to relatively high ratio to prediction (RPD) values, demonstrated the precision of the models, especially for polyphenol and sugar content. High RPD values (5.1 and 4.3 for polyphenol and sugar, respectively) indicated that an accurate classification of apples based on their content could be achieved. CONCLUSION: NIR spectroscopy coupled with the multivariate calibration technique could be used to accurately measure the quality parameters of apples. In addition, in the case of breeding programs, NIR spectroscopy can be considered an interesting tool for sorting varieties according to a range of concentrations. Transition metal catalysts widely used in refineries are provided as oxides and require pre-reduction to become activated. The auto-reduction of a NiO/Al2O3 catalyst with acetic acid (HAc) followed by HAc steam reforming was investigated in a packed bed reactor. Effects of temperature and molar steam to carbon ratio (S/C) on reduction kinetics and catalyst performance were analysed. Results showed that a steady steam reforming regime along with complete NiO reduction could be obtained after a coexistence stage of reduction and reforming. A 2D nucleation and nuclei growth model fitted the NiO auto-reduction. The maximum reduction rate constant was attained at S/C = 2. Steam reforming activity of the auto-reduced catalyst was just below that of the H2-reduced catalyst, probably attributed to denser carbon filament formation and larger loss of active Ni. Despite this, a H2 yield of 76.4% of the equilibrium value and HAc conversion of 88.97% were achieved at 750 °C and S/C = 3. 相似文献

Anatase TiO2 nanoparticles have been successfully synthesized at 130 °C for 2 days via ionic liquid-assisted hydrothermal method. The obtained products are characterized using various techniques. The X-ray diffraction data reveal that the nanoparticles are anatase TiO2. FTIR spectrum shows that the presence of ionic liquid and indicates Ti–O–Ti peak at around 398 cm?1, and the bands at 1500 and 1600 cm?1 indicates C–H in-plane vibrations and stretching of imidazolium ring. Raman spectroscopy show bands at 142, 393, 513, and 636 cm?1 reveal crystalline anatase phase. UV–Vis spectroscopy shows the λ max at 355 nm corresponding to a band gap of 3.49 eV. TEM images reveal that the average diameters of anatase TiO2 nanoparticles are in the range 50–100 nm. Anatase TiO2 exhibited excellent photocatalysis for the degradation of organic dye. 相似文献

Single particle tracking (SPT) in biological systems is a quickly growing field. Many new technologies are being developed providing new tracking capabilities, which also lead to higher demands and expectations for SPT. Following a single biomolecule as it performs its function provides quantitative mechanistic information that cannot be obtained in classical ensemble methods. From the 3D trajectory, information is available over the diffusional behavior of the particle and precise position information can also be used to elucidate interactions of the tracked particle with its surroundings. Thus, three-dimensional (3D) SPT is a very valuable tool for investigating cellular processes. This review presents recent progress in 3D SPT, from image-based techniques toward more sophisticated feedback approaches. We focus mainly on the feedback technique known as orbital tracking. We present here a modified version of the original orbital tracking in which the intensities from two z-planes are simultaneously measured allowing a concomitant wide-field imaging. The system can track single particles with a precision down to 5 nm in the x-y plane and 7 nm in the axial direction. The capabilities of the system are demonstrated using single virus tracing to follow the infection pathway of Prototype Foamy Virus in living cells. 相似文献
