Tropomyosin receptor kinase A (TrkA/NTRK1) is a high-affinity receptor for nerve growth factor (NGF), a potent pain mediator. NGF/TrkA signaling elevates synovial sensory neuronal distributions in the joints and causes osteoarthritis (OA) pain. We investigated the mechanisms of pain transmission as to whether peripheral sensory neurons are linked to the cellular plasticity in the dorsal root ganglia (DRG) and are critical for OA hyperalgesia. Sensory neuron-specific deletion of TrkA was achieved by tamoxifen injection in 4-week-old TrkAfl/fl;NaV1.8CreERT2 (Ntrk1 fl/fl;Scn10aCreERT2) mice. OA was induced by partial medial meniscectomy (PMM) in 12-week-old mice, and OA-pain-related behavior was analyzed for 12 weeks followed by comprehensive histopathological examinations. OA-associated joint pain was markedly improved without cartilage protection in sensory-neuron-specific conditional TrkA knock-out (cKO) mice. Alleviated hyperalgesia was associated with suppression of the NGF/TrkA pathway and reduced angiogenesis in fibroblast-like synovial cells. Elevated pain transmitters in the DRG of OA-induced mice were significantly diminished in sensory-neuron-specific TrkA cKO and global TrkA cKO mice. Spinal glial activity and brain-derived neurotropic factor (BDNF) were significantly increased in OA-induced mice but were substantially eliminated by sensory-neuron-specific deletion. Our results suggest that augmentation of NGF/TrkA signaling in the joint synovium and the peripheral sensory neurons facilitate pro-nociception and centralized pain sensitization.  相似文献

Seventy-two pearl millet genotypes were water stressed at panicle development and grain filling stages. Neither grain yields, yield components, protein percent nor total protein per unit area were affected by water deficit during panicle development but protein content per grain was increased. When plants were water stressed during grain filling, grain yield, grains per unit area and 1000 grain weight were reduced, but grain protein percentage increased. Total protein per unit area was reduced primarily due to lower grain yield. The protein content per grain was unaffected by stress, suggesting that the apparent increase in protein percentage is due to reduced carbohydrate accumulation under stress.  相似文献

Java-MaC: A Run-Time Assurance Approach for Java Programs   总被引:2,自引:1,他引:2  
We describe Java-MaC, a prototype implementation of the Monitoring and Checking (MaC) architecture for Java programs. The MaC architecture provides assurance that the target program is running correctly with respect to a formal requirements specification by monitoring and checking the execution of the target program at run-time. MaC bridges the gap between formal verification, which ensures the correctness of a design rather than an implementation, and testing, which does not provide formal guarantees about the correctness of the system.Use of formal requirement specifications in run-time monitoring and checking is the salient aspect of the MaC architecture. MaC is a lightweight formal method solution which works as a viable complement to the current heavyweight formal methods. In addition, analysis processes of the architecture including instrumentation of the target program, monitoring, and checking are performed fully automatically without human direction, which increases the accuracy of the analysis. Another important feature of the architecture is the clear separation between monitoring implementation-dependent low-level behaviors and checking high-level behaviors, which allows the reuse of a high-level requirement specification even when the target program implementation changes. Furthermore, this separation makes the architecture modular and allows the flexibility of incorporating third party tools into the architecture. Virtualized datacenters and clouds are being increasingly considered for traditional High-Performance Computing (HPC) workloads that have typically targeted Grids and conventional HPC platforms. However, maximizing energy efficiency and utilization of datacenter resources, and minimizing undesired thermal behavior while ensuring application performance and other Quality of Service (QoS) guarantees for HPC applications requires careful consideration of important and extremely challenging tradeoffs. Virtual Machine (VM) migration is one of the most common techniques used to alleviate thermal anomalies (i.e., hotspots) in cloud datacenter servers as it reduces load and, hence, the server utilization. In this article, the benefits of using other techniques such as voltage scaling and pinning (traditionally used for reducing energy consumption) for thermal management over VM migrations are studied in detail. As no single technique is the most efficient to meet temperature/performance optimization goals in all situations, an autonomic approach that performs energy-efficient thermal management while ensuring the QoS delivered to the users is proposed. To address the problem of VM allocation that arises during VM migrations, an innovative application-centric energy-aware strategy for Virtual Machine (VM) allocation is proposed. The proposed strategy ensures high resource utilization and energy efficiency through VM consolidation while satisfying application QoS by exploiting knowledge obtained through application profiling along multiple dimensions (CPU, memory, and network bandwidth utilization). To support our arguments, we present the results obtained from an experimental evaluation on real hardware using HPC workloads under different scenarios.  相似文献

Acrylonitrile–butadiene–styrene (ABS)–clay composite and intercalated nanocomposites were prepared by melt processing, using Na‐montmorillonite (MMT), several chemically different organically modified MMT (OMMT) and Na‐laponite clays. The polymer–clay hybrids were characterized by WAXD, TEM, DSC, TGA, tensile, and impact tests. Intercalated nanocomposites are formed with organoclays, a composite is obtained with unmodified MMT, and the nanocomposite based on synthetic laponite is almost exfoliated. An unintercalated nanocomposite is formed by one of the organically modified clays, with similar overall stack dispersion as compared to the intercalated nanocomposites. Tg of ABS is unaffected by incorporation of the silicate filler in its matrix upto 4 wt % loading for different aspect ratios and organic modifications. A significant improvement in the onset of thermal decomposition (40–44°C at 4 wt % organoclay) is seen. The Young's modulus shows improvement, the elongation‐at‐break shows reduction, and the tensile strength shows improvement. Notched and unnotched impact strength of the intercalated MMT nanocomposites is lower as compared to that of ABS matrix. However, laponite and overexchanged organomontmorillonite clay lead to improvement in ductility. For the MMT clays, the Young's modulus (E) correlates with the intercalation change in organoclay interlayer separation (Δd001) as influenced by the chemistry of the modifier. Although ABS‐laponite composites are exfoliated, the intercalated OMMT‐based nanocomposites show greater improvement in modulus. © 2008 Wiley Periodicals, Inc. J Appl Polym Sci, 2008  相似文献

The role of secondary compounds (SC) in deterring herbivores and pathogens from vegetative parts of plants is well established, whereas their role in plant reproductive organs such as floral nectar is unclear. The present study aimed to reveal the response of free-flying honeybees to naturally occurring concentrations of four SC in floral nectar. We selected nicotine, anabasine, caffeine, and amygdalin, all of which are found in nectar of various plants. In repeated paired-choice experiments, we offered 20% sucrose solution as control along with test solutions of 20% sucrose with various concentrations of the above SC. Except for anabasine, naturally occurring concentrations of SC did not have a deterring effect. Furthermore, low concentrations of nicotine and caffeine elicited a significant feeding preference. SC can, therefore, be regarded as postingestive stimulants to pollinators, indicating that the psychoactive alkaloids in nectar may be a part of their mutualistic reward. Further studies are needed to test our hypothesis that psychoactive alkaloids in nectar impose dependence or addiction effects on pollinators.  相似文献

Electric vehicles (EVs) are becoming increasingly popular because of their low operating costs. However, the anticipated increase in EV usage presents opportunities as well as challenges for utilities. This paper models EV charging behavior of active consumers who strategize their EV charging schedule to minimize the charging cost. The electric utility employs a temporal and spatially varying real-time pricing scheme that minimizes peak-to-average load ratio by influencing charging behavior of multiple active consumers. These active consumers decide their optimal charging strategy based on real-time electricity prices in order to minimize their EV charging cost. A model predictive control (MPC)-based optimal pricing scheme for electric utility and optimal charging strategy for active consumers is proposed. Effects of the proposed approach on load profile is verified using simulations.  相似文献
