The skeleton is potential endogenous source of lead during pregnancy and lactation. We have undertaken a longitudinal investigation into the mobilization of lead from the human maternal skeleton to determine whether lead is mobilized from the maternal skeleton during pregnancy and lactation, and if so, when and how much is released. Subjects in the study were migrants to Australia (n=15) whose skeletal lead isotopic composition (endogenous lead) was different to that prevailing in the Australian environment (exogenous lead). This migrant cohort was compared with 6 multi-generational Australian controls. Biological and environmental samples were taken pre-pregnancy where possible, throughout pregnancy and postpartum for at least 6 months. Newly-born infants of the migrant and Australian mothers were monitored for 6 months. Blood lead concentrations for the migrant mothers ranged from 1.5 to 20 microg/dl (geometric mean 2.8) and for Australian mothers ranged from 1.9 to 4.3 microg/dl (geometric mean 2.9). There was minimal change in lead isotopic composition of the Australian pregnant controls although there were increases of approximately 40% in blood lead concentration in 3 of 6 cases during the postpartum period and from 0 to 12% in the other 3. In the migrant pregnant subjects, the geometric mean skeletal lead contribution to blood lead using the isotopic composition was approximately 33% (range 10-88%) for 14 subjects using a revised estimate for exogenous lead. Skeletal contribution to blood lead during the postpartum period was significantly greater than during pregnancy (P<0.001). The skeletal contributions to blood lead are higher and the changes are more consistent in those subjects who conceived within 100 days of arrival in Australia compared with those who conceived longer than 100 days. In the migrant subjects, changes in blood lead concentration during pregnancy and postpartum varied from subject to subject with an overall 20% increase; the increases during the postpartum period were greater than during pregnancy (P<0.001). It was estimated that the amount of maternal skeletal lead mobilized during pregnancy and transferred to the infant via cord blood averaged approximately 79%. The increased skeletal contribution to blood lead is attributed to a low daily calcium intake of approximately 500 mgCa/day, a condition which was present in both migrant and Australian subjects. An ongoing clinical trial is providing a new cohort with calcium supplements. A summary of other aspects of the study is included and covers: additional flux released from the skeleton during pregnancy and postpartum; XRF bone lead results; urinary excretion of lead during pregnancy and postpartum; dietary contribution to blood lead in female adults and children; comparison of rates of exchange of lead in blood of newly-born infants and mothers; relationships of lead in breast milk to lead in blood, urine and diet of the infant and mother; changes in blood lead after cessation of breastfeeding; urinary lead isotopes during pregnancy and postpartum indicate no preferential partitioning of endogenous lead into plasma; a comparison of some aspects of the nonhuman primate and human pregnancy studies.  相似文献

Many chemicals released into the environment without toxicological risks have the capacities to disrupt the function of endocrine systems. These endocrine disruptors disturb normal endocrine mechanisms and have been observed in nearly all classes of vertebrates. The aim of this research is to develop a comprehensive model to study endocrine disruption using the amphibian Xenopus laevis. The assessment of estrogenic potencies of endocrine disruptors includes several levels of investigation: (I) binding to liver estrogen receptor, (II) estrogenic activity in vitro by inducing vitellogenin synthesis in primary cultured hepatocytes, and (III) in vivo effects on sexual development caused by exposure of larvae. The present paper is focused on the first part by establishing a radioreceptorassay for [3H]17 beta-estradiol ([3H]E2) binding using liver cytosol fraction. In order to get optimum binding conditions we performed kinetic, saturation, and competitive displacement experiments. Association of [3H]E2 to estrogen receptor revealed that maximum specific binding is achieved between 18 and 48 h of incubation. Scatchard analyses of saturation experiments resulted in a homogenous saturable population of estrogen receptors having no significant differences of binding parameters between both sexes. The values of Kd (dissociation constant) in males and females were 22.4 +/- 6.0 and 15.0 +/- 2.8 nM (mean +/- S.E.M.; n = 5), respectively, while corresponding Bmax (maximum binding capacity) revealed 89 +/- 46 and 136 +/- 46 fmol [3H]E2/mg protein. The specificity of estrogen receptors as shown by competitive displacement experiments demonstrated receptors being highly specific just for estrogens, but not for other endogenous steroids having the following ranking of binding affinities: E2 > estrone > dehydroepiandrosterone > aldosterone > or = testosterone > or = corticosterone > or = progesterone. The affinity ranking of environmental chemicals compared to E2 was: E2 > tetrachlorbiphenyl > diethylphthalate > 2,2-bis-(4-hydroxyphenyl)-propan (bisphenol A) > or = 4-nonylphenol > or = 3-t-butyl-4-hydroxyanisole > or = 4-octylphenol > dichlor-diphenyl-trichlor-ethan (4,4'-DDT). Analyses of five sewage effluents for displacement of [3H]E2 binding resulted in three samples displacing more than 50% of specific binding at their original concentration. Taken together the established radioreceptorassay for [3H]E2 binding in Xenopus laevis liver cytosol is useful to screen estrogen receptor binding of pure compounds or complex mixtures of them, which is the prerequisite for causing either estrogenic or antiestrogenic effects.  相似文献

Ten full-scale test fires were conducted in a chamber simulating a three person ship accommodation quarter. The test fires used three different ventilation conditions, two types of bunks and furnishings with either polyurethane foam or chloroprene foam. The chamber was instrumented to follow the development of the fire.The furnishings with polyurethane foam were readily ignited and produced an intense fire within 2 to 4 minutes. The furnishings with chloroprene foam ignited but burned slowly with a small flame or in smoldering combustion.The ventilation conditions had a significant effect on the development and intensity of the fire as the test fires were burning under oxygen-limiting conditions.The solid pan bunks retarded the early development of the fire compared to open bunks for polyurethane furnishings. However, once the mattresses on the three bunks were ignited the intensity of the test fires was similar for both types of bunks.  相似文献

Synthesis of ettringite from acid wastewaters of the aluminium anodising industry has been studied as a possible route of reducing the emissions to the environment, recovering at the same time resource materials as a useful marketable mineral. Wastewaters of different concentrations have been subjected to the process of synthesis suspending calcium oxide and calcium aluminate powders at different time and pH conditions. High caustic alkalinity (pH approximately 12) and low sulphate concentrations (<0.1 M) are the most suitable conditions to synthesise ettringite. The mineral characterisation has been performed using X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA) and differential thermal analysis (DTA), proving the high purity of the pursued solid product when hydrated in the appropriate sodium hydroxide concentrations. In such conditions, around 90% of the aluminium initially present in the wastewater solutions is recovered in the form of ettringite.  相似文献

A model was developed and tested to investigate the effects of various parameters on the processing costs of anaerobic digestion. The model was based on empirical data collected from full-scale plants which varied in size from 35,000 to 900,000 population equivalents. (Equivalent to loading rates of 0.5 to 2.7 kg VS/m3/d.) In spite of different operating regimes and conditions of the plants analysed, various relationships were discovered between: sludge quality and volatile solids destruction; sludge quality and biogas yield; and, effluent volatile solids and dewatering. Where correlations existed, these were used to make predictions on operating costs of the digestion and dewatering plant based on sensitivity analysis. The results of the model were used to make recommendations on how to optimise the operation of an anaerobic digestion plant.  相似文献

Most researchers agree that preserving, creating, and restoring urban wildlife habitats has to be an interdisciplinary undertaking if it is to succeed. Ecologists and wildlife biologists must be key participants. The culturally modified context of most urban habitats suggests that landscape architects also have a primary role to play.Open space in the urban/suburban environment is a scarce and valuable resource. Private and public sector pressure to convert these spaces to commercial or intensive recreational uses are immediate and intense. Presenting arguments to preserve or restore urban open space solely as habitat for wildlife is seldom successful. Decision makers are more likely to support urban wildlife habitat programs if other uses are also accommodated.There is a growing body of research data that suggests that wildlife are being adversely impacted by multiple uses of urban habitats. The challenge for planners and designers is to minimize adverse impacts and capitalize on those attributes of other uses that enhance habitat value. Planning for the temporal as well as spatial dimensions of site uses is required if the needs of wildlife are to be met.The paper presents three general areas in which landscape architects have traditionally contributed to an interdisciplinary approach to urban wildlife habitat planning: contextual perspective, problem solving process, and design/technical execution.Reflecting back on his past 10 years of experience as a landscape architect planning urban wildlife habitats, the author presents several case study projects in which: (A) accommodation of multiple use objectives, including wildlife habitat, was required; (B) interdisciplinary teams participated in the planning and design process; (C) key principles from the fields of landscape ecology, conservation biology and wildlife biology were applied to facility and activity location, habitat configuration, and management.Plans of completed projects are used to illustrate how habitat values were preserved or enhanced and multiple uses accommodated in public open spaces.
