The objective of the study was to determine the effect of active dry Saccharomyces cerevisiae (ADSC) supplementation on dry matter intake, milk yield, milk components, ruminal pH, and microbial community during a dietary regimen that leads to subacute ruminal acidosis (SARA). Sixteen multiparous, rumen-cannulated lactating Holstein cows were randomly assigned to 1 of 2 dietary treatments that included ADSC (Biomate; AB Vista, Marlborough, UK; 8 × 1010 cfu/head per day) or control. During wk 1 to 6, all cows received a high-forage (HF) diet (77:23, forage:concentrate). Cows were then abruptly switched during wk 7 to a high-grain (HG) diet (49:51, forage:concentrate) and remained on the HG until the end of wk 10. Feed intake and milk yields were recorded daily. Ruminal pH was recorded continuously using an indwelling system for 1 to 2 d per week during the pre-experimental phase, and wk 6, 7, and 10. Ruminal digesta samples were collected at the end of the experiment and analyzed for relative change in microbial communities using real-time quantitative PCR. Cows were considered to have SARA if the duration below pH 5.6 was ≥300 min/d. Ruminal pH during wk 6 (HF plateau) was not different across treatments (15 ± 46 min/d at pH <5.6). The dietary regimen successfully induced SARA during wk 7 (transition from HF to HG diet), and ruminal pH (551 ± 46 min/d at pH <5.6) was not different across treatments. However, cows receiving ADSC had an improved ruminal pH (122 ± 57 vs. 321 ± 53 min/d at pH <5.6) during wk 10 (HG plateau) compared with control. Additionally, cows receiving ADSC had a better dry matter intake (23.3 ± 0.66 vs. 21.6 ± 0.61 kg/d) and 4% fat-corrected milk yield (29.6 ± 1.2 vs. 26.5 ± 1.2 kg/d) than control cows during the HG phase (wk 8 to 10). During HG feeding, cows receiving ADSC had greater total volatile fatty acid and propionate concentrations (175 ± 7.5 vs. 154 ± 7.5 and 117 ± 6.1 vs. 94 ± 5.7 mM for ADSC and control, respectively) and lower acetate:propionate ratio (0.26 ± 0.5 vs. 0.36 ± 0.05 for ADSC and control, respectively). Microbial analyses conducted on samples collected during wk 10 showed that cows supplemented with S. cerevisiae had a 9-fold, 2-fold, 6-fold, 1.3-fold, and 8-fold increase in S. cerevisiae, Fibrobacter succinogenes, Anaerovibrio lipolytica, Ruminococcus albus, and anaerobic fungi, respectively, which suggested an increase in cellulolytic microbes within the rumen. Cows supplemented with ADSC had 2.2-fold reduction in Prevotella albensis, which is a gram-negative bacterium predominant during SARA. Prevotella spp. are suggested to be an important source of lipopolysaccharide responsible for inflammation within the rumen. Cows supplemented with ADSC had a 2.3-fold increase in Streptococcus bovis and a 12-fold reduction in Megasphaera elsdenii. The reduction in M. elsdenii may reflect lower concentration of lactic acid within the rumen for ADSC cows. In conclusion, ADSC supplementation to dairy cows was demonstrated to alleviate the condition of SARA caused by abrupt dietary changes from HF to HG, and can potentially improve rumen function, as indicated by greater numbers of cellulolytic microorganisms within the rumen.  相似文献

State estimation procedures using the extended Kalman filter, particle filter, and least squares are investigated for a transient heat transfer problem in which a high heat flux concentrated source is applied on one side of a thin plate and ultrasonic pulse time of flight is measured between spatially separated transducers on the opposite side of the plate. This work is an integral part of an effort to develop a system capable of locating the boundary layer transition region on a hypersonic vehicle aeroshell. Results from thermal conduction experiments involving one-way ultrasonic pulse time of flight measurements are presented. Comparisons of heating source localization measurement models are conducted where ultrasonic pulse time of flight readings provide the measurement update to the extended Kalman filter, particle filter, and least squares. Two different measurement models are compared: (1) directly using the one-way ultrasonic pulse time of flight as the measurement vector and (2) indirectly obtaining distance from the one-way ultrasonic pulse time of flight and then using these obtained distances as the measurement vector. For the direct model, the Jacobian required by the extended Kalman filter and least squares is obtained numerically using finite differences and a finite element forward conduction solution. For the indirect model, the derivatives with respect to the state variables are obtained in closed form. Heating source localization results and convergence behavior are compared for the three inverse methods and the two measurement models. The extended Kalman filter, least squares, and particle filter methods using the one-way ultrasonic pulse time of flight measurement model (direct model) produced similar results when considering accuracy of converged solution, ability to converge to the correct solution, and smoothness of convergence behavior. The results provide quantified justification for moving forward with development of an extended Kalman filter-based localization solution.  相似文献

Morpholine is a precursor of carcinogenic nitrosamines and although the possibility of their formation in the human stomach after ingestion of morpholine-treated apples is reported as highly unlikely morpholine has not been authorised as a food additive in the EU. Methods for its detection are required since it is permitted in other jurisdictions and may be present on food through direct treatment of fruit with waxes containing the compound, through steam treatment during processing or from packaging. Methods using derivatising agents with the inclusion of UV chromophores such as dansyl chloride yield good separation and high sensitivity but with mass spectrometric fragment ions predominantly originated from the derivatising group rather than the morpholine moiety. An amine acetylation derivatisation method is proposed from which fragment ions originating from the morpholine group are detected using widely available GC–MS. With full validation, a forensically robust confirmation of the presence of morpholine via its N-acetyl derivative is possible in support of regulatory analysis.  相似文献

The paradoxical behaviour of a new command and control concept called Network Enabled Capability (NEC) provides the motivation for this paper. In it, a traditional hierarchical command and control organisation was pitted against a network centric alternative on a common task, played thirty times, by two teams. Multiple regression was used to undertake a simple form of time series analysis. It revealed that whilst the NEC condition ended up being slightly slower than its hierarchical counterpart, it was able to balance and optimise all three of the performance variables measured (task time, enemies neutralised and attrition). From this it is argued that a useful conceptual response is not to consider NEC as an end product comprised of networked computers and standard operating procedures, nor to regard the human system interaction as inherently stable, but rather to view it as a set of initial conditions from which the most adaptable component of all can be harnessed: the human.  相似文献

The oxyacetylene torch facility is used to measure the ablation rates of graphite and the surface temperatures of different aerospace materials. The free‐stream flame environment is characterized as a function of flame chemistry for heat flux, pO2, and flow velocity. Measured ablation rates for graphite increase as a function of increasing heat flux and pO2, which are validated by applying an oxygen diffusion based model. The model uses experimentally measured values for temperature, pO2, and gas velocity in order to confirm torch testing results are reliable and reproducible. Surface temperatures of ultra‐high temperature ceramic composites are measured as a function of increasing heat flux and show an enthalpic cooling effect on the flame during oxidation testing.  相似文献

This review evaluates the role of α-adrenoceptor antagonists as a potential treatment of prostate cancer (PCa). Cochrane, Google Scholar and Pubmed were accessed to retrieve sixty-two articles for analysis. In vitro studies demonstrate that doxazosin, prazosin and terazosin (quinazoline α-antagonists) induce apoptosis, decrease cell growth, and proliferation in PC-3, LNCaP and DU-145 cell lines. Similarly, the piperazine based naftopidil induced cell cycle arrest and death in LNCaP-E9 cell lines. In contrast, sulphonamide based tamsulosin did not exhibit these effects. In vivo data was consistent with in vitro findings as the quinazoline based α-antagonists prevented angiogenesis and decreased tumour mass in mice models of PCa. Mechanistically the cytotoxic and antitumor effects of the α-antagonists appear largely independent of α 1-blockade. The proposed targets include: VEGF, EGFR, HER2/Neu, caspase 8/3, topoisomerase 1 and other mitochondrial apoptotic inducing factors. These cytotoxic effects could not be evaluated in human studies as prospective trial data is lacking. However, retrospective studies show a decreased incidence of PCa in males exposed to α-antagonists. As human data evaluating the use of α-antagonists as treatments are lacking; well designed, prospective clinical trials are needed to conclusively demonstrate the anticancer properties of quinazoline based α-antagonists in PCa and other cancers.  相似文献
