Oxysterols play a key role in many (patho)physiological processes and they are potential biomarkers for oxidative stress in several diseases. Here we developed a rapid gas chromatographic-mass spectrometry-based method for the separation and quantification of 11 biologically relevant oxysterols bearing hydroxy, epoxy, and dihydroxy groups. Efficient chromatographic separation (resolution ≥ 1.9) was achieved using a medium polarity 35%-diphenyl/65%-dimethyl polysiloxane stationary phase material (30 m × 0.25 mm inner diameter and 0.25 μm film thickness). Based on thorough analysis of the fragmentation during electron ionization we developed a strategy to deduce structural information of the oxysterols. Optimized sample preparation includes (i) extraction with a mixture of n-hexane/iso-propanol, (ii) removal of cholesterol by solid phase extraction with unmodified silica, and (iii) trimethylsilylation. The method was successfully applied on the analysis of brain samples, showing consistent results with previous studies and a good intra- and interday precision of ≤20%. Finally, we used the method for the investigation of oxysterol formation during oxidative stress in HepG2 cells. Incubation with tert-butyl hydroperoxide led to a massive increase in free radical formed oxysterols (7-keto-chol > 7β-OH-chol >> 7α-OH-chol), while 24 h incubation with the glutathione peroxidase 4 inhibitor RSL3 showed no increase in oxidative stress based on the oxysterol pattern. Overall, the new method described here enables the robust analysis of a biologically meaningful pattern of oxysterols with high sensitivity and precision allowing us to gain new insights in the biological formation and role of oxysterols.  相似文献

Within the framework of an interdisciplinary research project “investigation of forest ecosystems in the ore-mountains as basis for an ecologically oriented silviculture” supported by the Federal-ministry for education, science, research and technology (BMBF), Bonn, a study of the wood quality of heavily SO2-polluted and wind-exposed spruce trees from high altitude stands was conducted. For this study wood from 6 standing, wind-exposed and 6 storm-tossed trees with brash fractures was considered. By means of light microscopy (polarization, phase contrast) and electron microscopy the formation, distribution and extension of slip planes within the cell walls were examined and their significance for fracture initialization quantified. Wood discolouring and wood destroying fungi were isolated from the fracture zones and taxonomically determined. The extent of wood decay was characterized on a cellular and subcellular scale by means of fluorescence microscopy and UV spectroscopy. In the heavily SO2-polluted and thinned-out spruce stands at the high altitudes of the ore-mountains extreme wind stresses initiate in some tress slip planes which accumulate in the vicinity of rays and branch tracers. The slip plane zones are subsequently infected by wood degrading fungi, in particular by Stereum sanguinolentum (Alb. et Schw.: Fr.) Fr. resulting in a simultaneous decay of carbohydrates and lignin in the cell walls. The combined effect of wind stress and cell-wall degradation apparently induces brash, staircase-like fractures in some stems, which limitates the utilization of the timber. Eventually reduced SO2 immissions and an improved stand structure will help avert this kind of damage and spruce grown at this site will again satisfy standard wood quality requirements.  相似文献

Atom chips are a promising candidate for a scalable architecture for quantum information processing provided a universal set of gates can be implemented with high fidelity. The difficult part in achieving universality is the entangling two-qubit gate. We consider a Rydberg phase gate for two atoms trapped on a chip and employ optimal control theory to find the shortest gate that still yields a reasonable gate error. Our parameters correspond to a situation where the Rydberg blockade regime is not yet reached. We discuss the role of spontaneous emission and the effect of noise from the chip surface on the atoms in the Rydberg state.  相似文献

Open porous mechanically stable ceramic foams are developed by a simple direct foaming process. The new processing route is based on the transition of a surfactant stabilized highly concentrated alkane phase homogeneously distributed in a stabilized aqueous ceramic powder suspension into high performance ceramic foams with porosities up to 90% and cell sizes ranging from 3 to 200 μm. The droplet size distribution of the high alkane phase emulsified suspensions (HAPES) is efficiently controlled by the stirring velocity during emulsification experimentally investigated for varying powder particle contents. Stable foams with tailored structural features can be prepared by adjusting the rheological characteristics of HAPES being dependent on the system and process parameters. The influence of the emulsification stirring velocity on the resulting HAPES droplet size is analysed on the basis of the Taylor model of mechanical shearing describing the stresses responsible for the fragmentation of the droplets.  相似文献

Two paintings on metal support have been imaged by terahertz time-domain imaging (THz-TDI) in a reflection setup and the X-ray radiographs were also recorded. The study was performed for testing the terahertz radiation (THz) as an imaging method alternative to X-ray radiography, which suffers several limitations in imaging paint layers on metal support. While the information regarding the paint layers of the paintings was almost lost in the records provided by the X-ray radiography, THz-TDI demonstrates the ability to provide important information about them, despite the presence of the underlying metal.  相似文献

We present an interesting processing route for obtaining alumina/mullite‐based ceramics with controlled porosity and airflow resistance leading to promising microstructures for application as sound absorbers. The use of ceramic materials aims for potential applications where high temperatures or corrosive atmospheres are predominant, e.g., in combustion chambers of gas turbines. For the production of the porous ceramics we combined freeze gelation and sacrificial templating processes to produce near‐net‐shaped parts with low shrinkage (<3%) based on environmental‐friendly and low cost conditions. The obtained microstructure presents a bimodal pore size distribution, with small pores derived from the freeze gelation process (~30 μm) connecting large pores (2–5 mm diameter) originated from the expanded polystyrene template particles. These connections, called “windows” in this study, show a significant impact on the sound absorption properties, allowing the pressure diffusion effect to take place, resulting in a significant improvement of the sound absorption coefficient. By varying the template particle content and the slurry solid content, it is possible to control the sound absorption behavior at different frequencies of the open‐celled ceramics. These ceramics feature a high open porosity, from 77% to 82%, combined with sufficient compressive strength ranging from 0.27 to 0.68 MPa and sound absorption coefficients of 0.30–0.99, representing a highly promising combination of properties for noise control and reduction at corrosive environments and high temperatures.  相似文献
