The article presents a new idea of the application of polymer track membranes (PTM) for immobilization of a reagent in fiber optic chemical sensors. PTM was made of a poly(ethylene terephthalate) foil (10 μm in thickness, pores of 0.2 μm in diameter). The usefulness of membranes additionally covered by poly(vinyl chloride) was tested in a fiber optic redox titrator. The titrator utilized N,N′-diphenylbenzidine as a reagent which changes its absorbance in dependence on the redox potential. The measuring system is based on a lightemitting diode and a silicon photodiode connected to a bifurcated fiber optic bundle. Dynamic self-diffraction processes have been observed and analysed for methylene blue-sensitised water-swollen gelatine (MBSG). A degenerate two-wave mixing experiment performed on thick samples of MBSG with 10 mW, 632.8 nm wavelength He Ne laser light allowed for the formation of phase- and amplitude-transient holographic gratings. Single-beam propagation characteristics through MBSG were measured with a Mach-Zehnder interferometer. The observed energy transfer between the writing beams in the two-beam coupling experiment and the considerable temporal oscillations of diffraction efficiency are explained by simple formulae. Two processes are considered: the time-dependent phase shift between the writing beams and the transverse self-phase modulation effects (self-focusing and interference ring formation).  相似文献

Pectobacterium parmentieri is a pectinolytic plant pathogenic bacterium causing high economic losses of cultivated plants. The highly devastating potential of this phytopathogen results from the efficient production of plant cell wall-degrading enzymes, i.e., pectinases, cellulases and proteases, in addition to the impact of accessory virulence factors such as motility, siderophores, biofilm and lipopolysaccharide (LPS). LPS belongs to pathogen-associated molecular patterns (PAMPs) and plays an important role in plant colonization and interaction with the defense systems of the host. Therefore, we decided to investigate the heterogeneity of O-polysaccharides (OPS) of LPS of different strains of P. parmentieri, in search of an association between the selected genomic and phenotypic features of the strains that share an identical structure of the OPS molecule. In the current study, OPS were isolated from the LPS of two P. parmentieri strains obtained either in Finland in the 1980s (SCC3193) or in Poland in 2013 (IFB5432). The purified polysaccharides were analyzed by utilizing 1D and 2D NMR spectroscopy (1H, DQF-COSY, TOCSY, ROESY, HSQC, HSQC-TOCSY and HMBC) in addition to chemical methods. Sugar and methylation analyses of native polysaccharides, absolute configuration assignment of constituent monosaccharides and NMR spectroscopy data revealed that these two P. parmentieri strains isolated in different countries possess the same structure of OPS with a very rare residue of 5,7-diamino-3,5,7,9-tetradeoxy-l-glycero-l-manno-non-2-ulosonic acid (pseudaminic acid) substituted in the position C-8: 3)-β-d-Galf-(13)-α-d-Galp-(18)-β-Pse4Ac5Ac7Ac-(26)-α-d-Glcp-(16)-β-d-Glcp-(1. The previous study indicated that three other P. parmentieri strains, namely IFB5427, IFB5408 and IFB5443, exhibit a different OPS molecule than SCC3193 and IFB5432. The conducted biodiversity-oriented assays revealed that the P. parmentieri IFB5427 and IFB5408 strains possessing the same OPS structure yielded the highest genome-wide similarity, according to average nucleotide identity analyses, in addition to the greatest ability to macerate chicory tissue among the studied P. parmentieri strains. The current research demonstrated a novel OPS structure, characteristic of at least two P. parmentieri strains (SCC3193 and IFB5432), and discussed the observed heterogenicity in the OPS of P. parmentieri in a broad genomic and phenotype-related context.  相似文献

Simple SummaryA new sources of plant growth-promoting (PGP) bacteria are needed to increase the agricultural crops without increasing the usage of chemicals. The aim of this study was to test the hypothesis that insect-gut bacteria promote tomato plant growth. The insect gut bacteria were screened for the presence of PGP traits. The plants treated with bacterial consortium showed a significant increase in fruit yield, in both number of fruits (+41%) and weight of fruits (+44%). Our results showed that: (i) D. virgifera gut’s bacteria significantly promote the growth of tomato plants, and (ii) bacteria other than plant-related can be considered as PGP. It must be underlined that even though the insect gut bacteria were proven to stimulate the plant’s growth, their practical usage must be preceded by an examination of their influence on ecological and biological safety.AbstractWe investigated gut bacteria from three insect species for the presence of plant growth properties (PGP). Out of 146 bacterial strains obtained from 20 adult specimens of Scolytidae sp., 50 specimens of Oulema melanopus, and 150 specimens of Diabrotica virgifera, we selected 11 strains displaying the following: PGP, phosphate solubility, production of cellulase, siderophore, lipase, protease, and hydrogen cyanide. The strains were tested for growth promotion ability on tomato (Lycopersicon esculentum) plants. Each strain was tested individually, and all strains were tested together as a bacterial consortium. Tomato fruit yield was compared with the negative control. The plants treated with bacterial consortium showed a significant increase in fruit yield, in both number of fruits (+41%) and weight of fruits (+44%). The second highest yield was obtained for treatment with Serratia liquefaciens Dv032 strain, where the number and weight of yielded fruits increased by 35% and 30%, respectively. All selected 11 strains were obtained from Western Corn Rootworm (WCR), Diabrotica virgifera. The consortium comprised: Ewingella americana, Lactococcus garvieae, L. lactis, Pseudomonas putida, Serratia liquefaciens, and S. plymuthica. To our knowledge, this is the first successful application of D. virgifera gut bacteria for tomato plant growth stimulation that has been described.  相似文献

4-(5-methyl-1,3,4-thiadiazole-2-yl) benzene-1,3-diol (C1) and 4-[5-(naphthalen-1-ylmethyl)-1,3,4-thiadiazol-2-yl] benzene1,3-diol (NTBD) are representative derivatives of the thiadiazole group, with a high antimycotic potential and minimal toxicity against normal human fibroblast cells. The present study has proved its ability to synergize with the antifungal activity of AmB. The aim of this work was to evaluate the cytotoxic effects of C1 or NTBD, alone or in combination with AmB, on human renal proximal tubule epithelial cells (RPTECs) in vitro. Cell viability was assessed with the MTT assay. Flow cytometry and spectrofluorimetric techniques were used to assess the type of cell death and production of reactive oxygen species (ROS), respectively. The ELISA assay was performed to measure the caspase-2, -3, and -9 activity. ATR-FTIR spectroscopy was used to evaluate biomolecular changes in RPTECs induced by the tested formulas. The combinations of C1/NTBD and AmB did not exert a strong inhibitory effect on the viability/growth of kidney cells, as evidenced by the negligible changes in the apoptotic/necrotic rate and caspase activity, compared to the control cells. Both NTBD and C1 displayed stronger anti-oxidant activity when combined with AmB. The relatively low nephrotoxicity of the thiadiazole derivative combinations and the protective activity against AmB-induced oxidative stress may indicate their potential use in the therapy of fungal infections.  相似文献

Two types of polymeric sols, resorcinol‐formaldehyde (RF) and resorcinol‐furfural (RFur), were mixed in a water‐containing medium with aqueous solutions of inorganic salts: NH4ClO4, Mg(ClO4)2, and NH4NO3. After gelation and an ambient pressure drying, hybrid nanocomposites with properties of energetic materials were obtained. It was stated that salt concentration and addition of a second solvent (e.g., methanol or N,N‐dimethylformamide) to the mixture of reagents have crucial meaning for gel formation. In the case when only water was used, the mixture of organic sol/inorganic salt did not transfer from sol to gel, and precipitates were formed. Conventional drying of wet gelled composites leads to rigid material called xerogels. The RF xerogels are red and RFur xerogels are black. Typically, xerogels are transparent at low salt concentration (below 30%). The microstructure, morphology, and some other properties of chosen composites were studied by means of HR SEM, AFM, XRD, DTA/TG, and N2 adsorption isotherm techniques. SEM observation revealed that sizes of the oxidizer particles vary from less than 100 nm to ca. 1000 nm. XRD analyses also confirmed the presence of nanometer‐sized crystals of oxidizers in some formulations. The specific surface area of polymeric matrix/oxidizer composites was found to be in the range from 0.002 to 0.3 m2 g−1. After removing the salt from the composites (by extraction with boiling water), the specific surface area grows even up to 210 m2 g−1. TG/DTA analyses showed that the tested composites decompose as typical energetic materials. If pre‐heated and exposed to flame, some of them (especially RF/Mg(ClO4)2 composites) undergo violent deflagration with loud sound and flash effect.  相似文献

This study aimed to formulate antibacterial dental adhesives. Phosphate-substituted methacrylate adhesives were modified with 0–20 wt % copper-doped glass microparticles. Two shapes of microparticles were used: regular shaped (microspheres) and irregular shaped (microparticles). The morphology/composition, roughness, monomer conversion (DC%), thermogravimetric analysis, and antibacterial action against S. mutans and P. aeruginosa and ion release were investigated. The results showed that microspheres produced adhesives with a relatively smoother surface than microparticles did. The DC% of adhesives increased with increasing glass filler content. Filled adhesives showed polymer decomposition at ~315 °C and glass melting at 600–1000 °C. The weight loss percent of adhesives decreased with increasing weight percent of fillers. Glass microparticles at 0–20 wt % significantly increased the antibacterial action of adhesives against both bacteria. Glass microspheres at 0–5 wt % significantly increased the antibacterial action of adhesives against both bacteria. Only 20 wt % microparticle-filled adhesive showed an inhibition zone similar to tobramycin (positive control). Microparticle-filled adhesives (with >5 wt % filler) significantly reduced S. mutans more than their microsphere counterparts. Microsphere-filled adhesives (with ≤5 wt % filler) significantly reduced P. aeruginosa more than their microparticle counterparts. Microsphere-filled adhesives showed higher Cu release than their microparticle counterparts. Antimicrobial resistance has become a serious concern for the treatment of urinary tract infections. In this context, an anti-adhesive approach targeting FimH, a bacterial lectin enabling the attachment of E. coli to host cells, has attracted considerable interest. FimH can adopt a low/medium-affinity state in the absence and a high-affinity state in the presence of shear forces. Until recently, mostly the high-affinity state has been investigated, despite the fact that a therapeutic antagonist should bind predominantly to the low-affinity state. In this communication, we demonstrate that fluorination of biphenyl α-d -mannosides leads to compounds with perfect π–π stacking interactions with the tyrosine gate of FimH, yielding low nanomolar to sub-nanomolar KD values for the low- and high-affinity states, respectively. The face-to-face alignment of the perfluorinated biphenyl group of FimH ligands and Tyr48 was confirmed by crystal structures as well as 1H,15N-HSQC NMR analysis. Finally, fluorination improves pharmacokinetic parameters predictive for oral availability.  相似文献

Soft liner materials in oral cavity environments are easily colonized both by fungi and dental plaque. These factors are the cause of mucosal infections. The microorganism that most frequently colonizes soft liner materials is Candida albicans. Colonization occurs on the surface of materials and within materials. A solution to this problem might involve modification of soft liner materials with silver nanoparticles (AgNPs). In this article, we present results showing the antifungal efficacy of silicone soft lining materials modified with AgNPs. The modification process was conducted by dissolving both material components (base and catalyst) in a colloidal solution of AgNPs and evaporating the solvent. Composites with various AgNP concentrations (10, 20, 40, 80, 120 and 200 ppm) were examined. The in vitro antifungal efficacy (AFE) of composite samples was 16.3% to 52.5%.  相似文献

Voltamperometric studies on the indirect electrochemical α-methoxylation of Boc-Pro-Gly-OMe and Boc-Val-Gly-OMe in MeOH in the presence of NaCl or NaBr as the mediator suggested that the first reaction step was a direct N-halogenation of the dipeptide by active chlorine or bromine adsorbed on the electrode surface. The kind of mediator (NaCl or NaBr), its concentration, the current density, and the applied electric charge had a significant influence on the reaction course. In the case of Boc-Pro-Gly-OMe, the use of sodium bromide was necessary to obtain a relatively high ratio of α-monomethoxylation to α,α-dimethoxylation. For Boc-Val-Gly-OMe, the selectivity for α-monomethoxylation was close to 100%, independently of the mediator. Optimisation of the selected electrolysis parameters allowed us to significantly improve the yield and selectivity of the α-methoxylation of Boc-Pro-Gly-OMe (Kardassis et al. Tetrahedron 54:3471, 1998) and to obtain good results in the α-methoxylation of Boc-Val-Gly-OMe.  相似文献
