We cloned and characterized a novel gene (abfA) encoding alpha-L-arabinofuranosidase (alpha-L-AFase) from Aspergillus oryzae. One clone homologous to the alpha-L-AFase gene of Thermotoga maritima was found in an expressed sequence tag (EST) library of A. oryzae and a corresponding gene was isolated. Molecular analysis showed that the abfA gene carried six exons interrupted by five introns and had an open reading frame encoding 481 amino acid residues. The amino acid sequence similarity at active sites to the alpha-L-AFases from other organisms indicated that the alpha-L-AFase encoded by abfA was classified as a family 51 glycoside hydrolase. When the abfA was overexpressed in the homologous hyperexpression system of A. oryzae, a large amount of alpha-L-AFase was produced as intracellular protein. The apparent molecular mass of the purified enzyme was estimated to be 228,000 by gel filtration and that of its subunit as 55,000 by SDS-PAGE, suggesting that the enzyme is a tetramer. The enzyme hydrolyzed p-nitrophenyl-alpha-L-arabinofuranoside but not other p-nitrophenyl glycosides. These results demonstrated that the abfA gene encodes a functional alpha-L-AFase.  相似文献

Wavelength-tunable ultra-flat optical frequency comb generation is demonstrated using only a conventional Mach-Zehnder modulator that is asymmetrically dual-driven by large-amplitude sinusoidal signals with different amplitudes. A 10 GHz-spaced frequency comb with a 10 dB bandwidth of 230 GHz was experimentally generated. In addition, 50 nm widely wavelength tunable operation was achieved.  相似文献

Patients with muscle weakness such as muscular dystrophy usually need someone’s assistance in their daily activities. In order to reduce the caregiver burden and to improve quality of life (QOL) of the patients, various robotic technologies have been developed. This paper presents an exoskeletal assistance system EMAS II for the patients, which assists the upper extremity for the purpose of daily activities such as eating, writing, or other desk works. The EMAS II assists four DOF; shoulder flexion-extension, shoulder abduction-adduction, shoulder medial-lateral rotation, and elbow flexion-extension. The EMAS II has three kinds of user interfaces which are operated by residual functions of the patients, because it is important for patients’ health and initiative to use the residual functions. In order to control the four DOFs exoskeleton system using the interfaces with less DOF, the EMAS II simulates upper limb motion patterns of healthy people. The patterns are modeled by extracting correlations between the height of the wrist joint and that of the elbow joint. Therefore, users have only to control the position of their wrist joint to do tasks at a table. Through an experiment with a healthy subject, the feasibility of meal assistance by the EMAS II was confirmed. Furthermore, the system was applied to a spinal muscular atrophy patient in a clinical trial to check the usability. The experimental results indicated that the EMAS II could support the patient’s upper extremity to do tasks at a table.  相似文献

The preliminary results of mode measurement in the ITER relevant 40 m long transmission test line composed from 63.5 mm diameter corrugated waveguides and miter bends are presented. The field patterns were measured by taking temperature profiles on a paper screen placed in front of the waveguide end using an infrared imaging camera. The complex electric field at the waveguide end was retrieved from the measured temperature profiles. As a result, the transmission power includes 87% of HE11 mode and 6% of LP11 odd (HE21+TE01) mode and small ratios of other modes. The mode content had small dependence on length of the transmission line. This result indicated that the higher order mode content generated at the input of the transmission line is conserved and propagated through the transmission line. This suggests that the initial RF coupling to the waveguide is critical since it affects the launcher efficiency.  相似文献

We have developed a novel microchip equipped with a microchannel and Pt microelectrode array for electrochemically controlling valences of actinide (An) species. The square wave voltammograms of the redox reaction of potassium hexacyanoferrate(II) in the microchannel were measured. We found that the fabricated Pt microelectrode array has superior performances for the detection of the electrochemically active species in the microchannel. Therefore, the potentiostatic electrolysis experiments of uranium ions were carried out in the microchannel, and the concentration changes of uranium ions accompanied by the potentiostatic electrolysis were examined using thermal lens microscope. The results showed that the redox reactions between U(VI) and U(IV) can be performed completely in a microchannel in a few minutes, that is, the microscale reaction is accelerated by a factor of more than 10 compared with the bulk solution reactions taking hours mostly. The developed microchip was found to have enough performances for realizing rapid and highly efficient redox reactions for An species, which are impossible in the bulk reactions.  相似文献

Investigations were conducted on the VFT (Very Fast Transient) surges that propagate into a 500‐kV transformer. The disconnector restriking surge and ground fault surged were discussed. It was regarded that a large part of the surge voltage was applied just at the entrance of the transformer winding for the VFT surges. Thus, an equivalent circuit that models the windings was used for the analysis. The overvoltages that appear between the first winding sections at the entrance of the transformer were computed. The following was established. (1) The overvoltage between the first winding sections becomes greater when the magnitude of the voltage change at the transformer terminal is larger. The overvoltage between the first winding sections is not affected by the magnitude of the transformer terminal voltage. (2) For the disconnectors that are not connected directly to the transformer, the voltage change at the transformer terminal is not so large. (3) In the case of a ground fault at the GIS near the transformer, the voltage change at the transformer terminal is the same as that for a disconnector directly connected to the transformer. (4) In actual GIS, the disconnector that is connected directly to the transformer is not usually used. In this situation, the overvoltages that threaten the transformer insulation will not be generated by the restriking of the disconnectors. © 2003 Wiley Periodicals, Inc. Pulmonary arterial impedance is an important and interesting characteristic that can be used to evaluate the physiological properties of the pulmonary vessel. However, power spectrum analysis of the pulmonary artery pressure and flow pattern have suggested that peak power in the relatively high frequency range (> 10 Hz) is significantly low; thus, we cannot analyze the vessel properties in the high frequency range. In this study, we used the newly developed vibrating flow pump (VFP), which can generate oscillated blood flow with a relatively high frequency (10-50 Hz) for right heart bypass, to evaluate the pulmonary arterial impedance pattern in the high frequency range. Acute animal experiments of the right heart bypass from the right atrium to the pulmonary artery using 6 healthy adult goats were performed. The flow pattern and pressure of the pulmonary artery, electrocardiograms (ECGs), and arterial and right atrial pressures were continuously monitored during the experiments. Spectral analysis of the hemodynamic parameters using the fast Fourier transform (FFT) method was performed to evaluate the spectral properties. The coherence function, transfer function, and phase patterns were calculated to analyze the impedance pattern in the relatively high frequency area. Previously, various investigators had tried to analyze the impedance patterns of the pulmonary artery; however, they could not analyze the impedance patterns over 10 Hz because the spectral patterns of the pulmonary flow do not have high power at high frequencies. These physiological analyses may be useful in designing the optimal pulmonary circulation.  相似文献

We demonstrated a synchronous control technique for external optical modulation in a format of continuous-phase frequency-shift keying (CPFSK) at 10 Gb/s. In this method, the FSK signal in the upper- or lower-sideband state synchronously shifts to the other state at the timing when their phases are the same. We investigated the accuracy of the timing control required for the synchronous control. Experimental results show that the allowable timing misalignment to keep power penalty of the receiver sensitivity less than 1 dB was more than 25 ps, 25% of each bit period.  相似文献
