We cloned and characterized a novel gene (abfA) encoding alpha-L-arabinofuranosidase (alpha-L-AFase) from Aspergillus oryzae. One clone homologous to the alpha-L-AFase gene of Thermotoga maritima was found in an expressed sequence tag (EST) library of A. oryzae and a corresponding gene was isolated. Molecular analysis showed that the abfA gene carried six exons interrupted by five introns and had an open reading frame encoding 481 amino acid residues. The amino acid sequence similarity at active sites to the alpha-L-AFases from other organisms indicated that the alpha-L-AFase encoded by abfA was classified as a family 51 glycoside hydrolase. When the abfA was overexpressed in the homologous hyperexpression system of A. oryzae, a large amount of alpha-L-AFase was produced as intracellular protein. The apparent molecular mass of the purified enzyme was estimated to be 228,000 by gel filtration and that of its subunit as 55,000 by SDS-PAGE, suggesting that the enzyme is a tetramer. The enzyme hydrolyzed p-nitrophenyl-alpha-L-arabinofuranoside but not other p-nitrophenyl glycosides. These results demonstrated that the abfA gene encodes a functional alpha-L-AFase.  相似文献

Remineralization effects of xylitol on demineralized enamel   总被引:3,自引:0,他引:3  
We morphologically determined the effects of xylitol on the remineralization of artificially demineralized enamel. The samples were demineralized and then immersed in a remineralizing solution with or without 20% xylitol at 37 degrees C for 2 weeks. Samples were observed using contact microradiography, a multipurpose image processor (MIP) and a high-resolution electron microscope (HRTEM). Contact microradiography indicated that remineralization occurred in the surface and deep layers of samples immersed in a non-xylitol solution. Samples immersed in a xylitol solution demonstrated less mineralization in the outer 10 microm of the outermost surface layers, but more mineralization in the middle and deep layers, than was observed in the non-xylitol samples. The MIP evaluation indicated that remineralization was more prominent in layers at depths of 50-60 microm in the xylitol samples than in the non-xylitol samples. Observation of the xylitol samples by HRTEM revealed crystals of various sizes and irregular shapes with unclear crystal angles in the outermost surface layers. In the middle layers, they had thickened and angles of crystals are clear. These results indicate that xylitol can induce remineralization of deeper layers of demineralized enamel by facilitating Ca2+ movement and accessibility.  相似文献

When localized transverse loading is applied to a sandwich structure, the facesheet locally deflects and the core crushes. A residual dent induced by the core crushing significantly degrades the mechanical properties of the sandwich structure. In a previous paper, the authors established a “segment-wise model” for theoretical simulation of barely visible indentation damage in honeycomb sandwich beams with composite facesheets. Honeycomb sandwich beam was divided into many segments based on the periodic shape of the honeycomb and complicated through-thickness characteristics of the core were integrated into each segment. In this paper, the new model is validated by experiments using specimens with different types of honeycomb cores. In addition, the damage growth mechanism under indentation load was clarified from the viewpoint of the reaction force from the core to the facesheet. The applicability of the model to other types of core materials is also discussed.  相似文献

Go beyond the level that can be achieved, using only conventional natural language processing and vast repositories of sample patterns. Physical therapists are trained in manual examination techniques to test the impaired motor functions of patients. In this study, we have introduced a wearable robotic dummy joint to simulate disordered joint resistances or behaviors to support physical therapists in learning such techniques. We developed a discontinuous joint friction model based on a stick-slip phenomenon to simulate knee joint resistances caused by crepitus, a typical symptom accompanied by osteoarthritis. Practicing therapists participated in a reality-based evaluation test and specified acceptable parameter sets to adjust the simulated crepitus for the exoskeletal patient robot. The simulated crepitus and wearable dummy joint are expected to support the training of physical therapists.  相似文献

This paper proposes new sensorless vector control methods for permanent‐magnet synchronous motors (PMSMs), which are based on a new minimum‐order back‐EMF state observer taking acceleration into account, and examines potentiality of the improved back‐EMF observer through experiments. Conventional sensorless vector control methods for PMSMs using an estimate of back‐EMF are established under the assumption of constant speed or zero acceleration, and consequently cannot operate properly at modes requiring rapid speed change especially in low speed region. On the other hand, the proposed back‐EMF observer has the following features: (1) it is a new back‐EMF state observer taking acceleration into account and requiring no additional approximation to motor mathematical model; (2) it is a minimum‐order state observer; (3) it utilizes motor parameters in the simplest manner; (4) it can be applied to both salient‐pole and non‐salient‐pole PMSMs; (5) it can be realized in both rotor and stator reference frames. Detailed designs and analyses for the improved state observer and the sensorless vector control systems in both rotor and stator reference frames are given. In relation to the sensorless vector control systems, this paper newly proposes a double‐integral‐type PLL method and an integral‐feedback‐type acceleration/speed estimation method. The water wettability and non-wettability onto various kinds of polymers, which have so far been macroscopically described by the water contact angle, were investigated using the molecular orbital theory. An atomistic model consisting of an H2O molecule and a selected polymer crystal surface was analyzed in the isolated and the physisorbed states. The degree of interaction between the water molecule and the polymer surface was evaluated by considering the change of Mulliken charge at the oxygen site in the water molecule, and also the work of adhesion during the physisorption process. Such energetic quantities for several polymers were compared with the macroscopic water wettability/non-wettability. We found fairly good agreement between these approaches for the highly non-wettability polymers.  相似文献

This study was conducted to elucidate the effects of annealing and hydrogenation on the tensile properties of an Fe–0.01 mass% C alloy processed by high-pressure torsion (HPT). By HPT processing, the tensile strength was increased to ∼1500 MPa through grain refinement. Low-temperature annealing further strengthened the HPT-processed specimen because of a simultaneous effect of carbide precipitation and grain refinement. Reduction in the dislocation density and the fraction of low-angle grain boundaries through warm-temperature annealing led to a decrease in hydrogen uptake when the specimens were exposed to high-pressure gaseous hydrogen, and they became less sensitive to hydrogen embrittlement (HE).  相似文献
