Migration from can-coatings into retorted canned food simulants (canned oil and water, 121 degrees C, 30 min) was investigated through HPLC with a fluorescence detector and evaporative light scattering detector, and by measurements of residue on evaporation and consumption of potassium permanganate. HPLC analysis revealed that migration into the canned oil was hundreds of times more than that into n-heptane (25 degrees C, 60 min, the official test conditions according to the Japanese Food Sanitation Law), whereas it was similar to the migration into isooctane-butyl acetate mixtures (60 degrees C, 60 min), and that migration into the canned water was several times more than that into water (95 degrees C, 30 min, the official test conditions). Residue on evaporation for the n-heptane extract was several-fold lower than 30 ppm (the official limit), whereas that for the isooctane-butyl acetate mixtures exceeded 30 ppm. Consumption of potassium permanganate for the canned water was 30 times higher than that for the water extract (95 degrees C, 30 min). The official test conditions for can-coatings, in particular the use of n-heptane as an oil simulant, were suggested to lead to substantial underestimation of migration into canned food. 相似文献

Increased public awareness of health benefits of green tea is generally based on the high polyphenol content of tea leaves and the resulting beverage. A number of factors, such as species, season, agronomic condition and age of the leaves, are known to affect the composition of commercial teas. In the present study the effects of factors associated with domestic preparation and analytical methods, such as brewing time, concentration, solvent and type of tea product, on levels of catechins and caffeine, antioxidant activity and estimated daily intakes were investigated. There were large variations in the levels of total catechins: 43 and 117 mg g?1 dry matter (DM) (brewed for 30 s and 5 min respectively); 72 and 161 mg g?1 DM (extracted in boiling water and 50% acetonitrile respectively); 72 and 117 mg g?1 DM (a tea bag and tea leaves respectively). The effects on caffeine content were comparatively smaller. These variations consequently led to considerable variations in estimated daily intakes based on three cups (600 ml), ranging between 538 and 2014 mg g?1 DM of total catechins and between 103 and 466 mg g?1 DM of caffeine. The antioxidant activity was highest (26 680 µmol g?1 DM) for tea leaves brewed for 5 min and lowest (10 110 µmol g?1 DM) for a tea bag product brewed for 1 min. Phosphatidylinositol (PI) 3-kinase (encoded by the pik3(+) gene) in Schizosaccharomyces pombe has been identified as a homologue of VPS34p, a protein required for proper vesicular protein sorting. The clone defective in this protein carries enlarged vacuoles and exhibits sensitivity to high temperature or high ion concentration. The effect of disruption of pik3(+) on sporulation of Sz. pombe was examined. The diploid cells underwent G(1) arrest and meiosis. However, the spores formed by the deltapik3 cells were not viable. Electron-microscopic analysis revealed that the growth of the forespore membrane of deltapik3 cells was not correctly orientated, failing to engulf the nucleus or forming extremely small spores, as was confirmed by the use of Spo3p-GFP and GFP-Psy1p, which are markers of the forespore membrane. The coating materials found along the forespore membrane of the wild-type were greatly reduced in these cells. PI 3-P, the product of Pik3p, was detected on the forespore membrane, suggesting that PI 3-P-dependent vesicle transport may take place in formation of the forespore membrane. Misshaped forespore membrane, accumulation of vesicles, formation of small non-viable spores, and suppression by over expression of Psy1p were the phenotypes commonly seen in deltapik3 and deltaspo3 cells, suggesting a relationship between the functions of Pik3p and Spo3p in formation of the forespore membrane in Sz. pombe. 相似文献

Formation of ferrihydrite and associated iron corrosion products in permeable reactive barriers of zero-valent iron 总被引:1,自引:0,他引:1
Ferrihydrite, which is known to form in the presence of oxygen and to be stabilized by the adsorption of Si, PO4 and SO4, is ubiquitous in the fine-grained fractions of permeable reactive barrier (PRB) samples from the U.S. Coast Guard Support Center (Elizabeth City, NC) and the Denver Federal Center (Lakewood, CO) studied by high-resolution transmission electron microscopy and selected area electron diffraction. The concurrent energy-dispersive X-ray data indicate a strong association between ferrihydrite and metals such as Si, Ca, and Cr. Magnetite, green rust 1, aragonite, calcite, mackinawite, greigite and lepidocrocite were also present, indicative of a geochemical environment that is temporally and spatially heterogeneous. Whereas magnetite, which is known to form due to anaerobic Fe0 corrosion, passivates the Fe0 surface, ferrihydrite precipitation occurs away from the immediate Fe0 surface, forming small (<0.1 microm) discrete clusters. Consequently, Fe0-PRBs may remain effective for a longer period of time in slightly oxidized groundwater systems where ferrihydrite formation occurs compared to oxygen-depleted systems where magnetite passivation occurs. The ubiquitous presence of ferrihydrite suggests that the use of Fe0-PRBs may be extended to applications that require contaminant adsorption rather than, or in addition to, redox-promoted contaminant degradation. 相似文献

A rapid and simple method has been developed for the determination of 2-alkylcyclobutanone, 2-dodecylcyclobutanone (DCB), and 2-tetradecylcyclobutanone (TCB) in irradiated meat and processed foods. The procedure consists of extraction with n-hexane, following defatting and cleanup with a silica gel mini-column before gas chromatograph–mass spectrometry analysis. The method was evaluated using samples of beef, pork, Parmesan cheese, fried chicken, hamburger, gyoza (Chinese dumplings), and gyudon (boiled beef and onion seasoned with soy sauce and sugar). The recoveries of spiked DCB were 67–88 %, and those of TCB were 70–86 %. Furthermore, the method could detect DCB and TCB from samples irradiated at 1.0 and 2.6 kGy at levels dependent on dose; DCB and TCB were not detected in any nonirradiated samples. The method did not require special equipment, such as Soxhlet extraction, accelerated solvent extraction, or supercritical fluid extraction, for sample preparation. Brazil is the sixth largest producer of cocoa beans in the world, after Côte d’Ivoire, Ghana, Indonesia, Nigeria and Cameroon. The southern region of Bahia stands out as the country’s largest producer, accounting for approximately 60% of production. Due to damage caused by infestation of the cocoa crop with the fungus Moniliophthora perniciosa, which causes ‘witch’s broom disease’, research in cocoa beans has led to the cloning of species that are resistant to the disease; however, there is little information about the development of other fungal genera in these clones, such as Aspergillus, which do not represent a phytopathogenicity problem but can grow during the pre-processing of cocoa beans and produce mycotoxins. Thus, the aim of this work was to determine the presence of aflatoxin (AF) and ochratoxin A (OTA) in cocoa clones developed in Brazil. Aflatoxin and ochratoxin A contamination were determined in 130 samples from 13 cocoa clones grown in the south of Bahia by ultra-performance liquid chromatography with a fluorescence detector. The method was evaluated for limit of detection (LOD) (0.05–0.90 μg kg?1), limit of quantification (0.10–2.50 μg kg?1) and recovery (RSD) (89.40–95.80%) for AFB1, AFB2, AFG1, AFG2 and OTA. Aflatoxin contamination was detected in 38% of the samples in the range of ?1, with AFB1 in 25% of the total samples, whereas ochratoxin A was positive in 18% of the samples in the range of ?1. 相似文献

Hybrid Langmuir and Langmuir–Blodgett (LB) films consisting of amphiphilic cyclodextrins (CDs) and hydrophobic azobenzene were fabricated. Due to the larger cross-sectional area of the CD core compared with the sum of the cross-sectional areas of the attached alkyl chains, the films had free space in the hydrophobic regions. Azo(p-Hexyl) having hydrophobic terminal chains was accommodated in the free space, whereas the usage of an azobenzene derivative bearing hydrophilic groups led to the phase separation. In the present paper. a numerical model for MAG (metal active gas) arc welding of thin plate has been developed. In MAG arc welding, the electrode wire is melted and supplied into the molten pool intermittently. Accordingly, it is assumed on the modeling that the thermal energy enters the base-plates through two following mechanisms, i.e., direct heating from arc plasma and "indirect" heating from the deposited metal. In the second part of the paper, MAG arc welding process is numerically-analyzed by using the model. and the calculated weld bead dimension and surface profile have been compared with the experimental MAG welds on steel plate.
