Cultivated Agaricus bisporus possess the ability to bioaccumulate seven heavy metals (Pb, Cd, Hg, Fe, Cu, Mn, and Zn). The cultivated champignon mushroom A. bisporus was grown in soil composts of 16 different compositions. An edible mushroom, A. bisporus was also cultivated on humic compost artificially fortified with the seven heavy metals at five different concentrations (0.1 mg/kg, 0.5 mg/kg, 1.0 mg/kg, 3.0 mg/kg, and 10.0 mg/kg) and on control substrate (0 mg/kg) on a dry weight basis. The method of measuring bioaccumulation was flame atomic absorption spectrophotometry.  相似文献

In this study, activated carbons were prepared from sugar beet bagasse, which is side product and waste in sugar plants, by chemical activation with ZnCl2. Influence of activation temperature was investigated on to pore structure. ZnCl2/sugar beet bagasse ratio (impregnation ratio) was selected as 1:1. The impregnated sample was raised to the activation temperature under N(2) (100ml/min) atmosphere with 10 degrees C/min heating rate and hold at this temperature for 1h. The activation temperature was varied over the temperature range of 400-900 degrees C. BET surface area values were determined in the range of 832-1697 m(2)/g. Under the experimental conditions, 500 degrees C was found to be the optimal condition for producing high surface area carbons with ZnCl2 activation. Sugar beet bagasse was suitable for preparation of activated carbon with essentially microporous structure. Activated carbon ash content was found in the range of 1.2-2.7 (%w/w d.b.). Activated carbon samples and raw material were characterized by XRD, FT-IR, DTA and TGA.  相似文献

In this research, changes in volatile compounds of olive oil were investigated in terms of cultivar, harvest year, and geographic regions (Mediterranean, Aegean, Southeastern Anatolia, Marmara, and Black Sea) were investigated. Volatile compounds of olive oils were extracted by using solid-phase microextraction (SPME) method and were identified with gas chromatography–mass spectrometry. A total of 59 volatile compounds from seven different chemical groups including aldehydes, ketones, hydrocarbons, alcohols, esters, terpenes, and furans were identified. Aldehydes were detected as the major group in all geographic regions. Among aldehydes, trans-2-hexenal was found to be relatively higher. Alcohols and esters were determined as other important compounds. While many volatile compounds were affected by olive cultivar factor, this factor had no significant effect on esters. Principal component analysis showed that volatile compounds could play an important role in the separation of regions.  相似文献

High doses of morphine produce a state of behavioural inactivity and muscular rigidity. This type of 'catalepsy' is clearly different from the state which is produced by the administration of neuroleptics, e.g. haloperidol. While haloperidol-induced catalepsy can easily be antagonised by NMDA receptor antagonists, there has been a report that the non-competitive NMDA receptor antagonist (+)-5-methyl-10,11-dihydro-5H-dibenzo[a,d] cyclohepten-5,10-imine (MK-801) potentiates morphine-induced catalepsy. The aim of this study was to further examine the role of glutamate receptors in the mediation of morphine-induced catalepsy. To this end we coadministered morphine (20, 40, 60 mg/kg i.p.) with MK-801 (0.1 and 0.3 mg/kg i.p.), the competitive NMDA receptor antagonist DL-(E)-2-amino-4-methyl-5-phosphono-3-pentoic acid (CGP 37849) (2 and 6 mg/kg i.p.), or 1-(4-aminophenyl)-4-methyl-7,8-methylen-dioxy-5H-2,3- benzodiazepine (GYKI 52466) (2 and 4 mg/kg), an antagonist of the AMPA type of glutamate receptors, respectively. The degree of catalepsy was assessed using two different methods, the 'bar/podium/grid' test which is commonly used to measure neuroleptic-induced catalepsy, and a test for the presence or absence of righting reflexes after turning the animals into a supine position. It was found that in the 'bar/podium/grid' test coadministration of both NMDA receptor antagonists significantly and dose-dependently augmented morphine-induced catalepsy. The results using the AMPA receptor antagonist were less clear since the lower dose of GYKI 52466 tended to attenuate the morphine effect whereas the higher dose augmented morphine-induced catalepsy in some cases. While placing the animals on the bar and on the podium produced essentially the same results, the grid was found to be inapplicable for the measurement of morphine-induced catalepsy since the animals did not cling to the grid and fell off almost immediately after being released from the experimenter's hand. With respect to the righting reflexes it was found that the number of animals not showing these responses increased when MK-801 or CGP 37849 was coadministered with morphine. In contrast, most of the animals treated with GYKI 52466 and morphine displayed intact righting reflexes. It is concluded that glutamatergic transmission plays an important role in the mediation of morphine-induced catalepsy, though different to that of haloperidol-induced catalepsy, and that NMDA and AMPA receptors are differentially involved in different aspects of the associated behavioural state.  相似文献

Using the two-hybrid method, we isolated a Saccharomyces cerevisiae cDNA encoding a protein homologous to Schizosaccharomyces pombe protein Dis3sp, using as bait, human GTPase Ran. The DIS3 gene is essential for viability and complements S.pombe mutant dis3-54 which is defective in mitosis. Although Dis3sc has no homology to RanBP1, it bound directly to Ran and the S.cerevisiae Ran homologue Cnr1, but not to the S.cerevisiae RCC1 homologue Srm1. Upon binding to Ran with a 1:1 molar ratio, Dis3sc enhanced a nucleotide-releasing activity of RCC1 on Ran. In the presence of Dis3sc, the K(m) of RCC1 on Ran decreased by half, while the kcat was unchanged. In vivo, Dis3sp was present as oligomers of M(r) 670-200 kDa as previously reported, and the 200 kDa oligomer of Dis3sp was found to include Spi1 and Pim1, the S.pombe homologues of Ran and RCC1, respectively. Although the biological function of the heterotrimeric oligomer consisting of Dis3, Spi1 and Pim1 is unknown, our results indicate that Dis3 is a component of the RCC1-Ran pathway.  相似文献

A method is described that allows simultaneous measurement of two spectrally distinguishable green fluorescent protein (GFP) mutants with a confocal microscope. In contrast to previously described methods, neither UV excitation nor repetition of scans is required. Therefore the method is well-suited to the long-time observation of living cells in three-dimensional microscopy and time series recording, as demonstrated with GFP-expressing Dictyostelium discoideum cells.  相似文献

Traumatic events frequently produce false fear memories. We investigated the effect of hypothalamic corticotropin-releasing factor (CRF) knockdown (Hy-Crf-KD) or overexpression (Hy-CRF-OE) on contextual fear memory, as fear stress-released CRF and hypothalamic–pituitary–adrenal axis activation affects the memory system. Mice were placed in a chamber with an electric footshock as a conditioning stimulus (CS) in Context A, then exposed to a novel chamber without CS, as Context B, at 3 h (B-3h) or 24 h (B-24h). The freezing response in B-3h was intensified in the experimental mice, compared to control mice not exposed to CS, indicating that a false fear memory was formed at 3 h. The within-group freezing level at B-24h was higher than that at B-3h, indicating that false context fear memory was enhanced at B-24h. The difference in freezing levels between B-3h and B-24h in Hy-Crf-KD mice was larger than that of controls. In Hy-CRF-OE mice, the freezing level at B-3h was higher than that of control and Hy-Crf-KD mice, while the freezing level in B-24h was similar to that in B-3h. Locomotor activity before CS and freezing level during CS were similar among the groups. Therefore, we hypothesized that Hy-Crf-KD potentiates the induction of false context fear memory, while Hy-CRF-OE enhances the onset of false fear memory formation.  相似文献

Blended elemental W–xTiC (x = 0, 5, 10, 15 wt%) powders were mechanically alloyed (MA’d) for 30 h in a SPEX Mixer/Mill at room temperature. About 1 wt% Ni was added to each MA’d batch as sintering aid which were further milled for 1 h. MA’d powders were sintered at 1400 °C for 2 h under Ar, H2 gas flowing conditions and annealed at 1600 °C for 6 h under Ar atmosphere. Microstructural characterizations of as-sintered and annealed samples were conducted using XRD and SEM. XRD patterns of the as-sintered and annealed samples revealed the presence of the matrix W and Ni phases, whereas (Tix,W1−x) solid solution phase came into existence after annealing. In addition to XRD patterns, hot combustion and infrared detection measurements revealed the decarburization of TiC. Relative density values varied between 85.2% and 96.4% after sintering. The density values of sintered samples decreased with increasing TiC content. After annealing, a maximum relative density value of 99.8% was achieved. Vickers microhardness values varied between 5.11 GPa and 10.79 GPa for as-sintered samples and a maximum microhardness value of 8.1 GPa was measured after annealing. Wear resistance of the as-sintered samples increased with increasing TiC content.  相似文献

Abstract: Effect of radio frequency (RF) postdrying of partially baked cookies on acrylamide content, texture, and color of the final product was investigated in this study. Control cookies were prepared by baking in a conventional oven at 205 °C for 11 min. Cookies partially baked for 8 and 9 min were postdried in a 27.12 MHz RF tunnel oven until attainment of the moisture content of control cookies. Internal temperature of cookies was monitored during the experiments to better explain the results. Cookies were analyzed for acrylamide content using a liquid chromatography‐mass spectrometry (LC‐MS) method. Texture measurements were performed using a Texture Analyzer, while digital image analysis was used for color measurement. The results showed that RF postdrying of partially baked cookies resulted in lower acrylamide levels (107.3 ng/g for control cookies, 74.6 ng/g upon RF postdrying of cookies partially baked for 9 min, 51.1 ng/g upon RF postdrying of cookies partially baked for 8 min). Instrumental texture analysis showed no significant difference among the texture of cookies, whereas RF postdried samples had a lower degree of browning. According to sensory evaluation results, control had a more crumbly texture, and RF postdried sample that was conventionally baked for 8 min had a slightly uncooked flavor. Practical Application: Combining radio frequency and conventional baking may provide cookie manufacturers with ability to make cookies with lower levels of acrylamide.  相似文献

Unhydrogenated and hydrogenated sunflowerseed oils were exposed to the autoxidation process by sunlight under atmospheric conditions. Experiments were carried out in equal-sized glass, PET (polyethylene terephthalate) polymer, and metal (covered by tin) containers. The reaction time was 30 d, and the reaction course was observed by determining weight changes and peroxide values (PV) of the oil samples at the same time within 2-d intervals. The logarithm of the PV was plotted against time, and straight lines were obtained from the 4th or 6th d. The autoxidation reaction constants were obtained for each oil in each container. When comparing the reaction constants, the unhydrogenated oils autoxidize easily, and the autoxidation reaction occurs faster in sunlight in glass than in the PET polymer container and much faster than in the darkness of the metal container.  相似文献
