Unlike the general sliding vector design, the poleassignment method is applied to the overall system, not to a reduced system in sliding mode. High density lipoprotein (HDL) is throught to play a significant role in the process of reverse cholesterol transport. It
has become clear that the apolipoprotein (apo) composition of HDL is important in determining the metabolic fate of this particle.
The major proteins of human HDL are apoAI and APOAII; the latter protein is a disulfide-linked dimer in humans and higher
primates but monomeric in the other species. The consequences of the apo Cys6-Cys6 disulfide bridge in apoAII for human HDL
structure and function are not known. To address this issue, the influence of the Cys6-Cys6 disulfide bridge on the interaction
of human apoAII with palmitoyl-oleoyl phosphatidylcholine has been studied. The size and valence of a series of homogeneous
discoidal complexes containing either monomeric (reduced and carboxymethylated) or dimeric apoAII have been determined, and
their ability to remove cholesterol from rat Fu5AH hepatoma cells grown in culture has been compared. The apoAII dimer and
monomer form discoidal complexes of similar size, with twice as many of the latter molecule required per disc. Removal of
the disulfide bond influences the stability of the helical segments around the edge of the disc as seen by a decrease in α-helix
content of the monomeric protein. The discoidal particles containing the monomeric form of apoAII are somewhat more effective
than particles containing either dimeric apoAII or apoAI in removing cellular cholesterol. Overall, reduction of the disulfide
bridge of apoAII probably does not have a major effect in the determination of HDL particle sizein vivo. It follows that the evolution of the Cys6-Cys6 disulfide bond in higher primates probably has not had a major effect on

Molecular cloning of secretogranin II (SgII) in phylogenetically distant species has recently revealed the existence of a highly conserved 66-amino acid peptide flanked by preserved pairs of basic residues. This observation suggested that this peptide, named EM66, which had not been described to date, could be an important processing product of SgII. The aim of the present study was to investigate the possible occurrence of EM66 in the human adrenal gland. The EM66 peptide was generated in Escherichia coli, which was programmed to make a fusion protein containing the human EM66 sequence. The affinity-purified fusion protein was used to raise polyclonal antibodies in rabbits. The free EM66 peptide was obtained by cleavage of the fusion protein followed by high performance liquid chromatography purification. Immunohistochemical analysis using the EM66 antibodies revealed intense labeling of adrenochromaffin cells in the adult adrenal medulla and the fetal adrenal gland. A sensitive and specific RIA was developed and applied to the detection of EM66-like immunoreactivity in extracts of adult adrenal medulla and whole fetal adrenal gland after high performance liquid chromatographic analysis. A major immunoreactive species exhibiting the same retention time as recombinant EM66 was detected in both adult and fetal adrenal extracts. Taken together, these data demonstrate that posttranslational processing of SgII actually generates EM66 in the adrenal gland. The strong conservation of the amino acid sequence of EM66 in the vertebrate phylum and the occurrence of the mature peptide in both fetal and adult chromaffin cells suggest that EM66 could play an important physiological role in the human adrenal gland. 相似文献

Inviscid melt-spun calcia-alumina fibre-reinforced aluminium-silicon alloy (4032) composites were produced using a melt-infiltration technique. Scanning electron microscopy, energy dispersive X-ray spectroscopy (EDS), and X-ray diffraction (XRD) were used to investigate interfacial wetting and interphase formation, and identify the crystalline phase of the interphase of these composites. The composites processed at 700°C showed a good interfacial wetting and silicon accumulation at the interface. The composites processed at 927°C showed formation of an interphase region of about 10–20 m thick, as well as excellent interfacial wetting. EDS analysis gave averaged compositions of this interphase region at 74 wt % Al and 26 wt % Si, which corresponds to the composition of mullite (3Al2O3 · 2SiO2). The formation of mullite at the interface was confirmed by XRD analysis. 相似文献

The retinal pigment epithelium (RPE), situated upon Bruch’s membrane, plays multiple roles in the ocular system by interacting with photoreceptors and. Therefore, dysfunction of the RPE causes diseases related to vision loss, such as age-related macular degeneration (AMD). Despite AMD being a global cause of blindness, the pathogenesis remains unclear. Understanding the pathogenesis of AMD is the first step for its prevention and treatment.
