Efficient lipase production by two-step fed-batch culture of an organic solvent-tolerant bacterium, Pseudomonas aeruginosa LST-03, was investigated. When FB synthetic medium was used in flask culture, no lipase activity was detected, whereas lipase was produced at 2.3 I.U./ml in C2 complex medium. However, lipase production was induced in FB medium when a fatty acid was added to the culture broth in the stationary phase. Among fatty acids tested, long chain saturated fatty acids, such as C18 (stearic acid) and C20 (arachidic acid), were found to function as effective inducers for the production of lipase, giving an activity level almost the same as that obtained in C2 medium in flask culture. Two-step lipase production, comprised of a growth phase in fed-batch mode and a production phase in which lipase was induced by the addition of 5% (v/v) stearic acid, was carried out in a jar-fermentor. In the growth phase, the maximum cell concentration at 16 h was only 20 in terms of the optical density at 660 nm (OD660), and a low level of lipase production (8 I.U./ml) was obtained after 167 h. This was considered to be due to the exhaustion of several medium components brought about by the use of an unsuitable medium or feeding solution. After analyzing the contents of the compounds in the culture broth by inductively coupled plasma spectrometry for metal ions and HPLC for anions, a modified FB medium was designed. When this modified FB medium was used in two-step fed-batch culture, the maximum cell concentration reached an OD660 of 55 (30.2 g-dry cells/l) at 16.5 h, and lipase was produced at 96 I.U./ml after 35 h, which is approximately 40 times higher than the production level obtained in flask culture using C2 medium.  相似文献

Two-dimensional anti-ferromagnetic solid 3He on graphite, so called 4/7 phase, is a highly frustrated magnetic system and its ground state is considered to be a gapless spin liquid. Then how is the behavior in high magnetic fields? We tried to observe the magnetization curve at around 1 mK in high magnetic fields up to 10 T, using a Faraday-type magnetometer. To eliminate a large background signal from graphite as a substrate and copper as a thermal link, a double gradient coil system is employed to produce opposite field gradients in two regions equidistant from the field center. Preliminary results of the 4/7 phase on graphite show no saturated magnetization even at 1 mK and 10 T, suggesting the existence of magnetization plateau.  相似文献

This paper presents a low-power low-voltage 10-bit 100-MSample/s pipeline analog-to-digital converter (ADC) using capacitance coupling techniques. A capacitance coupling sample-and-hold stage achieves high SFDR with 1.0-V supply voltage at a high sampling rate. A capacitance coupling folded-cascode amplifier effectively saves the power consumption of the gain stages of the ADC in a 90-nm digital CMOS technology. The SNDR and the SFDR are 55.3 dB and 71.5 dB, respectively, and the power consumption is 33 mW  相似文献

Abstract— A new design for an FED envelope, composed of box‐shaped front glass, sheet‐like rear glass, and metal members, has been devised. This design structure is effective in reducing tensile stress induced by vacuum at the sealing points. Also, a new glass composition, a new physical tempering method, and a lead‐free hermetic sealing material have been developed. As a result, a novel lightweight spacer‐free panel structure for FEDs has been developed by integrating these new technologies. It will assist in maximizing the essential advantages of FED such as high image quality, high reliability, and low cost.  相似文献

The increase in electric power consumption in industry has led to the development of modern distribution systems such as control centers having high kVA ratings and high prospective fault current. In such a situation, the improvement of the system usefulness such as reliability and cost effectiveness is the essential requirement. An index is presented to evaluate the system usefulness using the cost effectiveness for fault protection in a system of given bank kVA. The index is estimated on four different system structures of control centers, including one which utilizes Permanent Power Fuses (PPF). It is presented that the PPF combined with molded case circuit breaker in main circuit effectively improves the cost effectiveness in control centers with high fault current. Operating principles of the PPF and the control center utilizing the PPF are briefly outlined.  相似文献

Registration of 3D data is a key problem in many applications in computer vision, computer graphics and robotics. This paper provides a family of minimal solutions for the 3D-to-3D registration problem in which the 3D data are represented as points and planes. Such scenarios occur frequently when a 3D sensor provides 3D points and our goal is to register them to a 3D object represented by a set of planes. In order to compute the 6 degrees-of-freedom transformation between the sensor and the object, we need at least six points on three or more planes. We systematically investigate and develop pose estimation algorithms for several configurations, including all minimal configurations, that arise from the distribution of points on planes. We also identify the degenerate configurations in such registrations. The underlying algebraic equations used in many registration problems are the same and we show that many 2D-to-3D and 3D-to-3D pose estimation/registration algorithms involving points, lines, and planes can be mapped to the proposed framework. We validate our theory in simulations as well as in three real-world applications: registration of a robotic arm with an object using a contact sensor, registration of planar city models with 3D point clouds obtained using multi-view reconstruction, and registration between depth maps generated by a Kinect sensor.  相似文献

Land cover is classified over East Asia using 250‐m Moderate Resolution Imaging Spectroradiometer (MODIS) land surface reflectance, MODIS snow cover and Operational Linescan System (OLS) human settlement data. The classification method includes a decision tree classification scheme that considers 11 kinds of land surface features derived from the OLS product and the time series of two MODIS products in 2000. The decision tree was defined manually based on the experiment because of insufficient training data, ease of tuning by visual interpretation, and extensibility to further research. The resulting classification is compared to three kinds of reference data, i.e. MODIS land cover product, Chinese digital land cover map, and Chinese census. The land cover classification can be input into a hydrological model applied to the Yellow River in China.  相似文献

Satellite‐based monitoring is an indispensable tool to guide soil‐specific crop management. However, it has attained little success in the estimation of soil nutrients due to the limitations incurred from inherent spectral characteristics. In this study, spectral band cloning (SBC) is developed and proposed to augment the soil nutrient predictive capabilities of broadband satellite data. Fine‐spectral channels of spectrometers were synchronized with coarse resolution of IRS satellite data to generate nutrient‐sensitive cloned IRS bands. Soil samples, collected at the time of satellite image acquisition in Lop Buri, Thailand, were analyzed both spectrally and chemically, viz., soil organic matter (OM), phosphorus, potassium and iron. The resulting SBC‐based models showed acceptable correlations, which otherwise were unattainable from raw IRS bands through prevailing models. Accuracy and validation measures showed good agreements between the measured and estimated nutrient surfaces. It is concluded that the SBC is a promising method of quantitative soil nutrient mapping, and could further be used for identification and mapping of other indiscernible biophysical parameters.  相似文献

Leaf area index (LAI) and actual evapotranspiration (ETa) from satellite observations were used to estimate simultaneously the soil hydraulic parameters of four soil layers down to 60 cm depth using the combined soil water atmosphere plant and genetic algorithm (SWAP–GA) model. This inverse model assimilates the remotely sensed LAI and/or ETa by searching for the most appropriate sets of soil hydraulic parameters that could minimize the difference between the observed and simulated LAI (LAIsim) or simulated ETa (ETasim). The simulated soil moisture estimates derived from soil hydraulic parameters were validated using values obtained from soil moisture sensors installed in the field. Results showed that the soil hydraulic parameters derived from LAI alone yielded good estimations of soil moisture at 3 cm depth; LAI and ETa in combination at 12 cm depth, and ETa alone at 28 cm depth. There appeared to be no match with measurement at 60 cm depth. Additional information would therefore be needed to better estimate soil hydraulic parameters at greater depths. Despite this inability of satellite data alone to provide reliable estimates of soil moisture at the lowest depth, derivation of soil hydraulic parameters using remote sensing methods remains a promising area for research with significant application potential. This is especially the case in areas of water management for agriculture and in forecasting of floods or drought on the regional scale.  相似文献
