Niemann–Pick disease type C (NPC) is a recessive hereditary disease caused by mutation of the NPC1 or NPC2 gene. It is characterized by abnormality of cellular cholesterol trafficking with severe neuronal and hepatic injury. In this study, we investigated the potential of glycoprotein nonmetastatic melanoma protein B (GPNMB) to act as a biomarker reflecting the therapeutic effect of 2-hydroxypropyl-β-cyclodextrin (HP-β-CD) in an NPC mouse model. We measured serum, brain, and liver expression levels of GPNMB, and evaluated their therapeutic effects on NPC manifestations in the brain and liver after the intracerebroventricular administration of HP-β-CD in Npc1 gene-deficient (Npc1−/−) mice. Intracerebroventricular HP-β-CD inhibited cerebellar Purkinje cell damage in Npc1−/− mice and significantly reduced serum and cerebellar GPNMB levels. Interestingly, we also observed that the intracerebral administration significantly reduced hepatic GPNMB expression and elevated serum ALT in Npc1−/− mice. Repeated doses of intracerebroventricular HP-β-CD (30 mg/kg, started at 4 weeks of age and repeated every 2 weeks) drastically extended the lifespan of Npc1−/− mice compared with saline treatment. In summary, our results suggest that GPNMB level in serum is a potential biomarker for evaluating the attenuation of NPC pathophysiology by intracerebroventricular HP-β-CD treatment.  相似文献

Immunoglobulin A (IgA)-albumin complexes may be associated with pathophysiology of multiple myeloma, although the etiology is not clear. Detailed structural analyses of these protein–protein complexes may contribute to our understanding of the pathophysiology of this disease. We analyzed the structure of the IgA-albumin complex using various electrophoresis, mass spectrometry, and in silico techniques. The data based on the electrophoresis and mass spectrometry showed that IgA in the sera of patients was dimeric, linked via the J chain. Only dimeric IgA can bind to albumin molecules leading to IgA-albumin complexes, although both monomeric and dimeric forms of IgA were present in the sera. Molecular interaction analyses in silico implied that dimeric IgA and albumin interacted not only via disulfide bond formation, but also via noncovalent bonds. Disulfide bonds were predicted between Cys34 of albumin and Cys311 of IgA, resulting in an oxidized form of albumin. Furthermore, complex formation prolongs the half-life of IgA molecules in the IgA-albumin complex, leading to excessive glycation of IgA molecules and affects the accumulation of IgA in serum. These findings may demonstrate why complications such as hyperviscosity syndrome occur more often in patients with IgA dimer producing multiple myeloma.  相似文献

Receptor-interacting protein kinase 1 (RIPK1) is a key component of the tumor necrosis factor (TNF) receptor signaling complex that regulates both pro- and anti-apoptotic signaling. The reciprocal functions of RIPK1 in TNF signaling are determined by the state of the posttranslational modifications (PTMs) of RIPK1. However, the underlying mechanisms associated with the PTMs of RIPK1 are unclear. In this study, we found that RING finger protein 4 (RNF4), a RING finger E3 ubiquitin ligase, is required for the RIPK1 autophosphorylation and subsequent cell death. It has been reported that RNF4 negatively regulates TNF-α-induced activation of the nuclear factor-κB (NF-κB) through downregulation of transforming growth factor β-activated kinase 1 (TAK1) activity, indicating the possibility that RNF4-mediated TAK1 suppression results in enhanced sensitivity to cell death. However, interestingly, RNF4 was needed to induce RIPK1-mediated cell death even in the absence of TAK1, suggesting that RNF4 can promote RIPK1-mediated cell death without suppressing the TAK1 activity. Thus, these observations reveal the existence of a novel mechanism whereby RNF4 promotes the autophosphorylation of RIPK1, which provides a novel insight into the molecular basis for the PTMs of RIPK1.  相似文献

The main function of skeletal muscles is to generate force. The force developed by myofiber contraction is transmitted to the tendon. There are two pathways of force transmission from myofibers to tendons: longitudinal transmission that depends on tension elicited via the myotendinous junction and lateral transmission that depends on shear elicited via the interface between the myofiber surface and surrounding connective tissue. Experiments using animal muscle and mathematical models indicated that lateral transmission is the dominant pathway in muscle force transmission. Studies using rat muscle showed that the efficiency of lateral force transmission declines with age. Here, the lateral transmission of force was measured using the extensor digitorum longus muscle from young and old mice. Dependence on longitudinal transmission increased in the old muscle, and there was a trend for lower efficiency of lateral force transmission in the old muscle compared to the young muscle. There was a noticeable increase in the connective tissue volume in the old muscle; however, there was no significant change in the expression of dystrophin, a critical molecule for the link between the myofiber cytoskeleton and extracellular matrix. This study demonstrates the measurement of lateral force transmission in mouse muscles and that alteration in force transmission property may underlie age-related muscle weakness.  相似文献

Part I briefly described environmental and societal issues that are mandating the incorporation of weight-adding technologies while requiring overall vehicular weight reduction for purposes of improving fuel economy. Part II describes the status of weight-reduction technologies that are currently available to car makers and projects how these technologies may evolve in the coming years. Homogeneous-eutectic microstructure of Y3Al5O12–Al2O3 system without coarse primary crystals was formed at an off-eutectic composition. This method utilizes a low migration rate in an amorphous phase. A mixture of Y2O3 and Al2O3 having the off-eutectic composition was melted and quenched rapidly to form an amorphous phase. A heat-treatment of the amorphous phase at 1000 °C and 1300 °C for 30 min formed Y3Al5O12 and Al2O3 phases. SEM observation of this material, which was formed from the amorphous phase at 1300 °C for 30 min, showed homogeneous eutectic-like microstructure. The formation of the primary crystals (coarse Al2O3), which are always observed in the off-eutectic compositions by ordinary method, was completely suppressed.  相似文献

Fibrates have been reported to elevate the hepatic proportion of oleic acid (18:1n‐9) through inducing stearoyl‐CoA desaturase (SCD). Despite abundant studies on the regulation of SCD in the liver, little is known about this issue in the small intestine. The present study aimed to investigate the effect of clofibric acid on the fatty acid profile, particularly monounsaturated fatty acids (MUFA), and the SCD expression in intestinal mucosa. Treatment of rats with a diet containing 0.5 % (w/w) clofibric acid for 7 days changed the MUFA profile of total lipids in intestinal mucosa; the proportion of 18:1n‐9 was significantly increased, whereas those of palmitoleic (16:1n‐7) and cis‐vaccenic (18:1n‐7) acids were not changed. Upon the treatment with clofibric acid, SCD was induced and the gene expression of SCD1, SCD2, and fatty acid elongase (Elovl) 6 was up‐regulated, but that of Elovl5 was unaffected. Fat‐free diet feeding for 28 days increased the proportions of 16:1n‐7 and 18:1n‐7, but did not effectively change that of 18:1n‐9, in intestinal mucosa. Fat‐free diet feeding up‐regulated the gene expression of SCD1, but not that of SCD2, Elovl6, or Elovl5. These results indicate that intestinal mucosa significantly changes its MUFA profile in response to challenges by clofibric acid and a fat‐free diet and suggest that up‐regulation of the gene expression of SCD along with Elovl6 is indispensable to elevate the proportion of 18:1n‐9 in intestinal mucosa.  相似文献

Eukaryotic microalgae have been known for high competency in the accumulation of triacylglycerol (TAG), a representative class of storage lipid. The snow algal species, Chlamydomonas nivalis, is a unique green eukaryotic microalga that can grow and survive in a wide range of temperatures. Although a few metabolomic studies of C. nivalis were conducted, no study has reported on TAG accumulation in C. nivalis. Herein, the present work aimed to investigate TAG production in C. nivalis under nutrient-starved conditions at 22 °C. Compared to phosphorus starvation, C. nivalis under nitrogen starvation showed a less severe growth defect, greater capacity for TAG production, and simple acyl composition in TAG enriched with 18:1. These features suggest that C. nivalis may be a significant model species to investigate glycerolipid metabolism for basic and applied research.  相似文献

The influence of the mechanically applied load on the piezoelectric properties and domain switching characteristics has been investigated experimentally using a soft lead zirconate titanate (PZT) piezoceramic, which de-poled to make randomly orientated domain structures. Mechanical loads were applied to rectangular PZT ceramics under compressive and tensile stresses. With the applied stress, 90° domain switching occurred in many locations in the PZT ceramics, where the higher stress caused more severe domain switching. The domain switching occurred with different directions, depending on the stress direction. For instance, the (2 0 0) peak increased and the (0 0 2) peak decreased when more severe compressive stress was applied. The opposite trend was detected as more severe tensile stress was applied; the (2 0 0) peak decreased and the (0 0 2) peak increased. With the applied loading process, the domains (long tetragonal c-axis) were orientated regularly in the PZT ceramics. Furthermore, like the electrically poled PZT ceramics, the domain orientations were more aligned when the sample surfaces were constrained during the loading process.  相似文献

Because plastics contribute to healthy and sound everyday life, global usage volume of plastic is expected to increase. However, the chemical industry is facing challenges in plastics from the viewpoints of resource depletion and environmental burden. These trends have led to discussions on how plastics should move forward in a sustainable society and a circular economy considering resource conversion, efficient after‐use utilization, and environmental protection. Bioplastics, both bio‐based and biodegradable, have reemerged as potential solutions. To clarify the role of bio‐based, biodegradable, and fossil‐based plastics, it is meaningful to assess lessons learned from experiences in the 1990s and 2000s. Although industries have been delivering solutions through the provision of materials, a coordinated and innovative approach throughout the value chain is necessary to achieve an integrated business model that incorporates efficient resource utilization, applications, and after‐use utilization, including chemical recycling, mechanical recycling, and energy/thermal recovery.  相似文献
