Anchietia salutaris tea is traditionally used in Brazil to treat allergies, suggesting it contains compounds with antagonistic activity on the allergic mediators. We have evaluated extracts and semi-purified fractions of Anchietia salutaris as a source of compounds having this type of antagonism on the contraction induced in guinea-pig lung parenchymal strips and on platelet aggregation and shape change. After 10 min pre-incubation dichloromethane extracts containing 30 or 100 microg mL(-1) inhibited the contraction induced by prostaglandin D2 (PGD2) in guinea-pig lung parenchymal strips with dose ratios (DR) of 0.76+/-0.14 and 0.93+/-0.19, respectively; the amount of inhibition depended both on the concentration and on the time of pre-incubation (DR after 30 min pre-incubation was 1.21+/-0.51). The dichloromethane extract and its semi-purified fractions also inhibited the contractions induced by U46619, a more potent, stable, synthetic agonist of thromboxane A2 (TxA2) prostanoid (TP) receptors, the receptors acted upon by PGD2 to produce lung contractions. The dichloromethane extract did not inhibit the lung parenchymal contractions induced by histamine, leukotriene D4 (LTD4) or platelet-activating factor (PAF). Platelet aggregation induced by U46619, adenosine 5'-diphosphate (ADP) or PAF was not inhibited by the dichloromethane extract. Indeed, the extract potentiated platelet aggregation induced by low concentrations of these agonists and also potentiated the shape change induced by U46619. These results imply that the dichloromethane extract of Anchietia salutaris and its semi-purified fractions contain an active principle that competitively inhibits TxA2 TP receptors, the stimulation of which causes lung parenchymal contraction. The inhibition seems to be selective for this receptor subtype, because the extract fails to inhibit platelet aggregation or shape change. This provides additional support of earlier reports suggesting the occurrence of TP receptor subtypes.  相似文献

Tumor necrosis factor-alpha (TNF-alpha) levels were measured in the serum (sTNF-alpha) or bone marrow (BM) biopsies of 43 patients with myelodysplastic syndromes (MDS) who subsequently received therapy with a combination of pentoxifylline and ciprofloxacin (PC) with or without dexamethasone (PCD). All 43 patients received only PC therapy for 12 weeks, after which 18 of 36 nonresponders received PCD. A total of 18 of 43 patients showed a hematologic or cytogenetic response or both. BM TNF-alpha levels were semiquantitatively assessed using immunohistochemistry on a scale of 0-8+ and in the serum using enzyme linked immunoassay. The median TNF-alpha for the entire group was 3.0 in BM and 6.9 pg/ml in the serum, and 14 patients had no detectable levels. Responders had higher BM levels (median 3.5 vs. 2.0) than nonresponders, although this was not statistically significant. During PC therapy, a decline in BM TNF-alpha level was seen in the entire group, which was significant at 2 weeks (p = 0.02), 8 weeks (p = 0.001), and 12 weeks (p = 0.0001). Both responders (p = 0.01) and nonresponders (p = 0.03) had a decline at 8 weeks, but at 12 weeks, only the responders continued to show a significant decline (p = 0.03). We conclude that MDS patients with high BM TNF-alpa levels have a better chance of responding to PCD therapy and that the therapy is quite successful in reducing the TNF-alpha levels in a sustained fashion. Future studies need to be directed at identifying agents that would be more potent suppressors of the proapoptotic cytokines in these patients.  相似文献

The photosynthetic and anatomical responses of bryophytes to changes in gravity will provide crucial information for estimating how these plant traits evolved to adapt to changes in gravity in land plant history. We performed long-term hypergravity experiments at 10g for 4 and 8 weeks using the moss Physcomitrella patens with two centrifuges equipped with lighting systems that enable long-term plant growth under hypergravity with irradiance. The aims of this study are (1) to quantify changes in the anatomy and morphology of P. patens, and (2) to analyze the post-effects of hypergravity on photosynthesis by P. patens in relation to these changes. We measured photosynthesis by P. patens for a population of gametophores (e.g., canopy) in Petri dishes and plant culture boxes. Gametophore numbers increased by 9% for a canopy of P. patens, with 24–27% increases in chloroplast sizes (diameter and thickness) in leaf cells. In a canopy of P. patens, the area-based photosynthesis rate (A canopy) was increased by 57% at 10g. The increase observed in A canopy was associated with greater plant numbers and chloroplast sizes, both of which involved enhanced CO2 diffusion from the atmosphere to chloroplasts in the canopies of P. patens. These results suggest that changes in gravity are important environmental stimuli to induce changes in plant growth and photosynthesis by P. patens, in which an alteration in chloroplast size is one of the key traits. We are now planning an ISS experiment to investigate the responses of P. patens to microgravity.  相似文献

This study describes a basic theory for reconstructing pure Raman signals of materials composing a multilayer sample from Raman spectra obtained using two types of miniaturized Raman probes. An illustrative example is demonstrated using a multilayer system of samples composed of the transparent plastics polymethylmethacrylate (PMMA) and polyethylene (PE) as a model of thin-layered biomedical tissues. When the same region of an object is measured using Raman probes with different focal properties, the Raman spectra provide different depth profile information depending on the level of light penetration. Thus, a detailed comparison of the spectra can provide an interesting opportunity to probe the differences between the layers. A simple analytic form is presented for reconstructing the pure Raman spectra of the embedded layer. The method applies an understanding of the Raman sampling volume in layered transparent materials to the interpretation of Raman spectra experimentally measured by multiple probes. The basic theory described here is necessary for the expansion of the technique to turbid media, such as biological samples, where light-scattering effects must be considered. The potential applications of the proposed method include material and catalyst subsurface probing through different embedded materials, such as assessment of silicon wafers, effective noninvasive screening for catalyst synthesis, and biomedical tissue research.  相似文献

A series of 2H- and 13C-labeled glutamates were used as substrates for coenzyme B12-dependent glutamate mutase, which equilibrates (S)-glutamate with (2S,3S)-3-methylaspartate. These compounds contained the isotopes at C-2, C-3, or C-4 of the carbon chain: [2-2H], [3,3-2H2], [4,4-2H2], [2,3,3,4,4-2H5], [2-13C], [3-13C], and [4-13C]glutamate. Each reaction was monitored by electron paramagnetic resonance (EPR) spectroscopy and revealed a similar signal characterized by g'xy = 2.1, g'z = 1.985, and A' = 5.0 mT. The interpretation of the spectral data was aided by simulations which gave close agreement with experiment. This approach underpinned the idea of the formation of a radical pair, consisting of cob(II)alamin interacting with an organic radical at a distance of 6.6 +/- 0.9 A. Comparison of the hyperfine couplings observed with unlabeled glutamate with those from the labeled glutamates enabled a principal contributor to the radical pair to be identified as the 4-glutamyl radical. These findings support the currently accepted mechanism for the glutamate mutase reaction, i.e., the process is initiated through hydrogen atom abstraction from C-4 of glutamate by the 5'-deoxyadenosyl radical, which is derived by homolysis of the Co-C sigma-bond of coenzyme B12.  相似文献

The relationships between morphology and proton conduction for Nafion membranes and hydrocarbon-type proton exchange membranes, namely, sulfonated poly(arylene ether ether ketone) (S-PEEK) and sulfonated poly(arylene ether sulfone) (S-PES), were investigated by small-angle X-ray scattering (SAXS) and atomic force microscopy (AFM). The direct simultaneous observation of surface morphology and active regions of proton conduction on membranes by combined high-resolution AFM phase imaging and an electrochemical technique at controlled humidity provided significant morphological information, particularly for the hydrocarbon-type membranes that exhibit few or no features on SAXS profiles. For the Nafion membranes, the active proton paths became denser and congregated with each other at over 60% RH, resulting in the formation of well-connected networks. For the hydrocarbon-type membranes, however, only the relatively small and dispersed proton paths were observed, which showed no significant change even as water content increased. We have demonstrated that the differences in microscopic morphology between the Nafion and hydrocarbon-type membranes are associated with the differences between their macroscopic proton conductivities.  相似文献

Climatic change is recognized as an important factor capable of influencing the structural properties of aquatic ecosystems. Lake ecosystems are particularly sensitive to climate change. Several long time-series studies have shown close coupling between climate, lake thermal properties and individual organism physiology, population abundance, community structure, and food-web structure. Understanding the complex interplay between climate, hydrological variability, and ecosystem structure and functioning is essential to inform water resources risk assessment and fisheries management. The purpose of this paper is to present the current understanding of climate-induced changes on lake ecosystem phenology. We first review the ability of climate to modulate the interactions among lake hydrodynamics, chemical factors, and food-web structure in several north temperate deep lakes (e.g., Lake Washington, Lake Tahoe, Lake Constance, Lake Geneva, Lake Baikal, and Lake Zurich). Our aim is to assess long-term trends in the physical (e.g., temperature, timing of stratification, and duration of ice cover), chemical (e.g., nutrient concentrations), and biological (e.g., timing of the spring bloom, phytoplankton composition, and zooplankton abundance) characteristics of the lakes and to examine the signature of local weather conditions (e.g., air temperature and rainfall) and large-scale climatic variability (e.g., ENSO and PDO) on the lake physics, chemistry and biology. We also conducted modeling experiments to quantify the relative effect of climate change and nutrient loading on lake phenology. These modeling experiments focused on the relative changes to the major causal associations underlying plankton dynamics during the spring bloom and the summer stratified period. To further understand the importance of climate change on lakes, we propose two complementary directions of future research. First, additional research is needed to elucidate the wide array of in-lake processes that are likely to be affected by the climate change. Second, it is essential to examine the heterogeneity in responses among different water bodies. The rationale of this approach and its significance for dealing with the uncertainty that the climate signals cascade through lake ecosystems and shape abiotic variability and/or biotic responses have been recently advocated by several other synthesis papers.  相似文献

ABSTRACT

Since a particular trivalent lanthanide (Ln(III)) often plays a unique role in different industrial applications, the development of a selective separation method to get only the requisite Ln(III) would be most convenient. Synergistic extraction with β-diketone (HA) and Lewis base (B), which has been extensively used for the separation of lanthanides, relies on two processes: the extraction with HA as LnA3 into the organic phase and the complexation of LnA3 with B as LnA3B in the organic phase. The variation in the pattern of the extraction constants of LnA3 across the Ln series correlates with the number of CF3- group in the β-diketones, and with the stability constants of LnA3B of different Lewis bases. It is seen that the constants with bipyridyl increase with increasing atomic number, while those with POPO (methylenediphosphonate) and MePhPA (methylphenylphenanthroline carboxamide) decrease with increasing Ln atomic number. Using pivaloyltrifluoroacetone as the β-diketone and MePhPA, the best extractability appeared at Sm(III) among the series. As a conclusion, to get only a required Ln(III), the combination of β-diketone and Lewis base is very important. In addition, the separation of Lns(III) from other metals and the use of new extractants developed recently and reported in literature were referred to find out more effective extractants and selective separation methods.  相似文献

Manganese dioxide electrodes were prepared by a thermal decomposition of manganese nitrate solution on a titanium or a platinum substrate, and their anodic characteristics were investigated mainly in 1N H2SO4 and 1N KOH. The platinum-supported manganese dioxide electrode shows the good anodic characteristics with a relatively low overvoltage for oxygen evolution while the use of the titanium-supported one as an anode needs further modification to reduce its high resistivity resulting from the thick film of titanium dioxide in spite of good adhesion of the oxide film with the titanium substrate.The primary water or hydroxide ion discharge step is rate-controlling in the anodic evolution of oxygen on the platinum-supported manganese dioxide electrode in both acidic and alkaline solutions. The oxygen overvoltage is raised and the mechanical strength of the catalytic oxide on the platinum substrate is weakened by the anodic polarization in acidic solution. These phenomena are explicable on the basis of an increase in the oxygen content in the oxide. In conclusion, the presented results suggest that the manganese dioxide film is a practical usable material for the anodic evolution of oxygen as well as chlorine, especially in alkaline solutions.  相似文献
