Plasma osmolality estimated from plasma concentrations of Na+, Cl-, K+, glucose, and urea was compared with measured osmolality in preweaned Holstein calves. When calves (n = 5) were fed only milk replacer after fasting for 24 h, measured osmolality fluctuated almost in parallel with estimated osmolality during the 8-h period after feeding, although estimated values were about 90% of measured values. When calves (n = 5) were fed only calf starter after fasting for more than 16 h, measured osmolality did not parallel the estimated osmolality during the 8-h period after feeding. We describe the development of a new transformation system, using multiple auxotrophic marker genes, for the fission yeast Schizosaccharomyces pombe. We developed three new auxotrophic marker genes (arg12(+), tyr1(+) and ade7(+)) and generated a new host strain, YF043, by Cre-loxP-mediated gene disruption. YF043 possessed six mutated biosynthetic genes (leu1-32, ura4-M190T, arg12::loxP, tyr1::loxP, ade7::loxP and his2::loxP). The combination of this host strain and the new selectable markers can be used for gene disruption using the same preexisting transformation systems. In addition, Sz. pombe vectors were constructed, containing selectable marker genes that complement the auxotrophies of YF043. These new vectors are available for gene disruption and heterologous protein expression in strain YF043. The new Sz. pombe host strain will be a useful tool for molecular genetic studies of Sz. pombe where multiple recombinant modifications or multiple mutations are needed. 相似文献

Abstract: The effect of sensory and extrinsic attributes on consumer intentions to purchase the Japanese traditional fermented soybean product natto was evaluated using conjoint analysis. Six attributes with 2 levels each were chosen and manipulated: price (high compared with low), the country of origin of the soybeans (domestic compared with imported), stickiness (strong compared with moderate), smell (rich compared with moderate), attached seasonings (attached compared with no attached seasonings), and the environmental friendliness of the packaging (high compared with low). A fractional factorial design was applied and 8 hypothetical product labels were produced. A sample of 479 Japanese housewives ranked these product labels based on their purchase intentions. Overall purchase intention was affected by country of origin, attached seasonings, and price; those attributes accounted for 81.0%, while the sensory attributes of the product accounted for 19.0% of purchase intents. In order to estimate market segments for the natto products based on consumer preference, a cluster analysis was performed. It identified 4 segments of consumers: 1 oriented to attached seasonings, another conscious of the price, and the other 2 oriented to origins. The behavioral and demographic characteristics of the respondents had a limited influence on segment membership. Practical Application: This research was conducted to understand how consumers valuate various sensory and nonsensory product attributes based on their assessment of the overall product in the case of Japanese fermented soy product (natto). We developed a simple method for quantification of the peroxide value (PV) in colored lipids on the basis of the reaction between triphenylphosphine (TPP) and oxidized oil to afford triphenylphosphineoxide (TPPO). Diphenylphosphineoxide (DPPO) was employed as internal standard. The formed TPPO was analyzed by high-pressure liquid chromatography–UV spectroscopy with absorption at 260 nm. The conditions that gave the highest correlative calibration curve between the peak area on the chromatogram and peroxide value were identified: the optimum TPP–oxidized oil mix ratio, reaction temperature, and reaction time were found to be 2:1, 40 °C, and 30 min, respectively. Linear calibration curves, passing through the origin, were obtained for PV versus TPPO and TPPO versus DPPO. The quantification limit for this method was 2.01 pmol hydroperoxyl group, which corresponds to a PV value of 0.2 meq/kg in a 10-mg oil sample. This method was used to measure the PV in colored fats and oils or lipids extracted from dark meat and processed food containing a coloring agent. Though the official method could not measure the PVs in the colored lipids, the method proposed here, which uses an inexpensive chemical reagent and machine, could. Using the pH-jump (neutral to above 11) for Tyr-OH ionization as a probe, two glucoamylases from Rhizopus niveus and Rhizopus delemar were found to be caused the change in conformation, which is in a kinetically single step. The pH titration at pHs below 12 was carried out on a multidimensional correlation spectrophotometer. Correlations among the spectrophotometric properties: fluorescence, absorption and circular dichroism are almost identical for the two enzymes. Pyrolysis is a promising technique for the recovery of useful gas, tar, and solid products from biomass waste. However, the low tar yields obtained from lignocellulosic biomass are a significant drawback. To enhance tar yields, sugarcane bagasse, which is the most abundant agricultural waste in Fiji, was pretreated at ambient temperature and atmospheric pressure using various sulfuric acid (H2SO4) concentrations. Here, the ether bonds of cellulose, hemicellulose, and lignin were partially hydrolyzed. The pretreated samples were then pyrolyzed at 500 °C, and it was confirmed that H2SO4-pretreatment disrupted the bagasse cell structure, with the thermogravimetry and differential thermogravimetry results confirming that decomposition occurred at lower temperatures after pretreatment. In addition, tar yields were significantly enhanced from 5.6 wt% to 13.4 wt% for the untreated and 3 M H2SO4-pretreated samples respectively. The main components detected in this tar product were levoglucosan, andcellulose-and hemicellulose-derived products, whose proportions were increased following pretreatment. Thus, our work demonstrates that dilute acid pretreatment enhances tar production from sugarcane bagasse due to the production of shorter chain components via the partial hydrolysis of ether bonds. 相似文献

The effect of nonproportional strain path on fatigue/creep-fatigue properties was investigated with 304 stainless steel at 550°C under strain controlled biaxial conditions. The fatigue/creep-fatigue life reduction due to nonproportional strain path occurred even at the lowest strain range investigated, that is, 0.2% for fatigue loading and 0.3% for creep-fatigue loading. The Mises-type path-dependent equivalent strain range was employed in order to evaluate the fatigue/creep-fatigue strength under nonproportional loading conditions. Stress relaxation behavior under nonproportional loading was examined. It was shown that stress relaxes proportionally toward the origin of stress plane even under nonproportional loading. Fatigue damage and creep damage were calculated based on the linear damage summation rule. Life prediction was shown to be possible within an accuracy of a factor of about 2 for nonproportional loading along with other waveforms including pure axial loading, pure torsional loading and combined proportional loading. 相似文献

The influence of the turbulence of reactant on the radiation-induced polymerization of ethylene in 40 mole-% Freon-114 (C2Cl2F4) was studied using a tubular reactor at 400 kg/cm2 and 25°C with a dose rate of 1.3 × 105 rad/hr. At constant linear velocity and tube diameter, the polymer concentration was shown to increase linearly with the reactor tube length. This indicates that the polymerization is in a stationary state. By changing the linear velocity from 3.5 to 42.7 cm/sec and the tube diameter from 5 to 14 mm, the space time yield and the molecular weight of polymer were found to vary between 0.21 and 0.46 mole ethylene/1.-hr and from 5.0×103 to 10.5×103, respectively. The space time yield and molecular weight decreased sharply to about one half those in the static polymerization with increasing fluid turbulence and then slowly increased in the highly turbulent state. To make a Mn2+-doped red glass phosphor that can be excited with ultraviolet (UV) light of light-emitting diodes (LEDs), 60P2O5-35ZnO-5Al2O3-8MnO-xCu2O glasses (x = 0-1.00) were prepared by a melt-quenching method at 1200-1400°C for 30-180 minutes in atmospheric air, and the redox of Mn and Cu as well as fluorescence properties were investigated. The Mn2+ ion was not reduced and oxidized in the melting, quenching, and annealing processes. The valence of Cu in the glasses changed in the order of 0, 1+, and 2+ with the increase in the amount of Cu2O and in the melting temperature and time. In this study, a 60P2O5-35ZnO-5Al2O3-8MnO-0.10Cu2O glass melted at 1250°C for 90 minutes, having the highest Cu+ concentration, showed the strongest Mn2+ red fluorescence under the UV light at 275 nm. This strong Mn2+ red fluorescence has been caused by the energy transfer from excited Cu+ ions to Mn2+ ions. 相似文献

Synthesis of the γ-Ga2O3-Al2O3 solid solutions by spray pyrolysis was examined. Spherical particles were obtained using an aqueous solution of Al(NO3)3 and Ga(NO3)3 with HNO3. For Ga-rich composition, γ-phase solid solutions were directly crystallized by the spray pyrolysis. For Al-rich composition, spray pyrolysis gave amorphous products unless a sufficient thermal energy was supplied during the spray pyrolysis. Subsequent calcination of the amorphous products gave γ-Ga2O3-Al2O3 solid solutions. However, physical properties of the solid solutions were affected by the spray pyrolysis conditions. 相似文献
