In ruminants, IFN-tau (IFNT) is a pregnancy recognition signal secreted by the embryonic trophectoderm before implantation, and it induces the expression of IFN-stimulated genes (ISG) in the uterine endometrium and blood leukocytes. The expression of ISG in blood leukocytes could indicate the presence of a viable conceptus before return of the next estrus; however, expression levels have high variation for confirming pregnancy. We hypothesized that the secreted IFNT in the uterus would affect ISG expression in cervical and vaginal tissues because they are directly adjacent to the uterus. To prove the hypothesis, we investigated the expression of 3 ISG (ISG15, MX1, and MX2) in cervical and vaginal mucosal membranes collected from pregnant (n = 12) and nonpregnant (n = 11) lactating Holstein cows at 17 to 18 d after artificial insemination. Mucosal membrane samples of the cervical canal near the external os (cervix) and deep vaginal wall surrounding the external os (vagina) were collected separately by simply scraping with a curette on d 17 or 18 of pregnancy (d 1 = ovulation), at which time IFNT secretion into the maternal uterus is maximal. After pregnancy diagnosis on d 30 and 60, separately collected samples confirmed as pregnant and nonpregnant were used for evaluation of the expression of IFN-stimulated protein 15 kDa (ISG15) and myxovirus-resistance protein 1 and 2 (MX1, MX2) with quantitative real-time PCR. The collected mucosal membrane samples from cervix contained mostly cell clots showing membrane structure and a low content of blood cells. The expression levels of all 3 genes were significantly increased in pregnant cows compared with nonpregnant cows in both cervical and vaginal samples. These results suggest that increased expression of ISG in the cervix and vagina is a pregnancy-associated phenomenon and is highly affected by IFNT secreted from the conceptus through the uterus.  相似文献

Carbon fibers are widely used as a reinforcement in composite materials because of their high specific strength and modulus. Current trends toward the development of carbon fibers have been driven in two directions; ultrahigh tensile strength fiber with a fairly high strain to failure (~2%), and ultrahigh modulus fiber with high thermal conductivity. Today, a number of ultrahigh strength polyacrylonitrile (PAN)-based (more than 6 GPa), and ultrahigh modulus pitch-based (more than 900 GPa) carbon fibers have been commercially available. In this study, the tensile strengths of PAN- and pitch-based carbon fibers have been investigated using a single filament tensile test at various gauge lengths ranging from 1 to 250 mm. Carbon fibers used in this study were ultrahigh strength PAN-based (T1000GB, IM600), a high strength PAN-based (T300), a high modulus PAN-based (M60JB), an ultrahigh modulus pitch-based (K13D), and a high ductility pitch-based (XN-05) carbon fibers. The statistical distributions of the tensile strength were characterized. It was found that the Weibull modulus and the average tensile strength increased with decreasing gauge length, a linear relation between the Weibull modulus, the average tensile strength and the gauge length was established on log–log scale. The results also clearly show that for PAN- and pitch-based carbon fibers, there is a linear relation between the Weibull modulus and the average tensile strength on log–log scale.  相似文献

In Japan, a large number of old small earth dams are in critical need of repair due to leakage and poor earthquake resistance. In addition to cohesive soils, geosynthetic clay liners (GCLs) are used as impervious materials to repair such dams. This paper discusses the seismic performance of small earth dams, with reservoirs on their upstream side, repaired with a sloping core zone and a GCL on the basis of the results of full-scale shaking table tests performed at the E-Defense facility. The main focus is on the differences in mechanical behavior between the upstream and downstream sides of the dam. The results elucidate that the effective stress of the upstream embankment materials increased because of the undrained shear behavior of the compacted soils, although the deformation on the upstream side was larger than that on the downstream side. A large phase difference in the measured accelerations between the upstream slope and the downstream slope was also observed. Therefore, it is concluded that significant differences occurred in the dynamic behavior of the upstream side and the downstream side.  相似文献

A multidimensional computational fluid dynamics (CFD) simulation of a constructed syngas chemical kinetic mechanism was performed to evaluate the combustion of syngas in a supercharged dual-fuel engine for various syngas initial compositions under lean conditions. The modelled results were validated by comparing predictions against corresponding experimental data for a supercharged dual-fuel engine. The predicted and measured in-cylinder pressure, temperature, and rate of heat release (ROHR) data were in good agreement. The effect of the hydrogen peroxide chain-propagation reaction on the progress of combustion under supercharged conditions was examined for different types of syngas using various initial H2 concentrations. The effect of the main syngas kinetic mechanism reactions on the combustion progress was analysed in terms of their contribution to the total heat of the reaction. The best results compared with experimental data were obtained in the range of equivalence ratios below about 0.8 for all types of syngas considered in this paper. As the equivalence ratio increased above 0.8, the results deviated from the experiment data. The spatial distribution of the in-cylinder temperature and OH within this equivalence-ratio range showed the completeness of the combustion. The present CFD model captured the overall combustion process well and could be further developed into a useful tool for syngas-engine combustion simulations.  相似文献

To maintain the efficiency of proton exchange membrane fuel cells (PEFCs) without flooding, it is necessary to control the liquid water transport in the gas diffusion layer (GDL). This experimental study investigates the effects of the GDL fiber direction on the cell performance using an anisotropic GDL. The results of the experiments show that the efficiency of the cell is better when the fiber direction is perpendicular to the channel direction, and that the cells with perpendicular fibers are more tolerant to flooding than cells with fibers parallel to the channel direction. To determine the mechanism of the fiber direction effects, the liquid water behavior in the channels was observed through a glass window on the cathode side. The observations substantiate that the liquid water produced under the ribs is removed more smoothly with the perpendicular fiber direction. Additionally, the water inside the GDL was frozen to observe its distribution using a specially made cell broken into two pieces. The photographic results show that the amount of water under the ribs is larger than that under the channels using the parallel fiber direction GDL while the water distributions in these two places are almost equal level with the perpendicular fiber direction GDL. This freezing method confirmed the better liquid water removal ability and better reactant gas transportation in the GDL with the fiber direction perpendicular to the channel direction.  相似文献

The selective oxidation of specific components in alumina-forming alloy such as CoNiCrAlY under precisely regulated oxygen partial pressures (PO2) can be used to control polymorphism in Al2O3 scale formed on the alloy. Dense, smooth α-Al2O3 scale was formed rapidly by treatment at 1323 K under a thermodynamically determined PO2, where both aluminum and chromium in the alloy were oxidized and elements such as cobalt and nickel were not oxidized. By contrast, under a higher PO2 all the components in the alloy were oxidized, the transformation was obviously retarded, and (Co,Ni)(Al,Cr)2O4 was produced.  相似文献
