In this study, the relationship between stunning techniques and protein oxidation which are accepted as the main cause of food spoilage was investigated. For this purpose, the antioxidant status, in vivo myofibrillary protein (MP) oxidation and sensitivity, and postmortem oxidation (inducted with hydroxyl radical system) of rainbow trout (Oncorhynchus mykiss) fillets killed by hitting to head (T1), neck crushing (T2), and convulsion (T3) methods, were investigated. Statistically significant differences (p < .05) were found among all parameters examined and it was observed that the most stressing technique was the convulsion method. It was determined that in protein profiles myosin were influenced too much from stunning technique and in actin observed oxidation-induced reductions. Reductions in S-S and S-H were also found to be increased in carbonyl concentrations, but the most effective values in both processes were determined by the convulsive technique. Our results show that short-term stunning techniques (hitting to head, neck crushing) give better meat quality results in terms of O. mykiss welfare and low MP oxidation rates. In general, we can say that T3 group fillets are more sensitive to oxidative damage, while T1 and T2 groups give better results in maintaining meat quality with low MP oxidation rates.  相似文献

This study aims to determine the effects of chitosan obtained from Metapenaeus stebbingi shells on the shelf life of refrigerated gilthead sea bream. It was determined that 1% chitosan‐coated samples had the lowest thiobarbituric acid (TBA) (3.05 mg malondialdehyde (MDA) kg?1) and free fatty acids (FFA) value (2.79% oleic acid), while the control group had the highest TBA (5.08 mg MDA kg?1) and FFA value (6.13% oleic acid) on the 27th day of storage. In the last day of storage, TVB‐N was found higher in control group (25.62 mg 100 g?1) than chitosan‐coated samples (14.57 mg 100 g?1). Total viable count value of the control group exceeded maximum permissible limit on the 27th day of storage. However, it was lower than 7.0 log CFU/g in chitosan‐coated samples during the refrigerated storage. As a result of this study, it was determined that shelf life of refrigerated gilthead sea bream can be increased up to 27 days with chitosan.  相似文献

Waste or by-products contains a substantial amount of nutrients. Fish silage is an alternative way for assessment of fish discards and waste generated in the fish processing industries. The aim of the study was to evaluate the potential use of discard species (Equulites klunzingeri, marine fish and Carassius gibelio, freshwater fish) for fish silage and chemical quality of fish oil extracted from fish silages prepared with acid and fermented methods. Lipid quality of fish oils extracted from fish silages with LAB strains was evaluated in terms of thiobarbituric acid (TBA), peroxide value (PV), p-Anisidine (AV), totox values and fatty acid composition. The results showed that PV, TBA, AV and totox values of all extracted fish oils were within the acceptable limits for edible oils. Recovered fish oils from silage contained high level of polyunsaturated fatty acids (PUFAs), especially eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), serving as a source of good quality fish oil for human consumption.  相似文献

Possible adulteration of canned products containing spirit vinegar pickle by adding synthetic acetic acid is a significant problem of the food industry. Isotope analyses, which determine botanical origin of acetic acid and also can detect synthetic acid, were applied to detect undeclared addition of synthetic acetic acid to canned products. The aim of the study was to improve the extraction technique for the SNIF-NMR (2H/1H; site-specific natural isotopic fractionation-nuclear magnetic resonance) and IRMS (13C/12C; isotope ratio mass spectrometry) isotope methods and for an atypical matrix and to determine isotope ratios in canned vegetables pickle to prove their adulteration or authenticity. The following set of canned products was analysed: pickled cucumbers (n = 16) and one vinegar pickle purchased in the Czech market and six model (cucumber) pickles. The determined ratios of 2H/1H and 13C/12C for the pickled cucumbers proved to be authentic ranged from 89.4 to 107.0 ppm and from ?28.7 to ?15.6 ‰, respectively; for the synthetic acetic acids diluted with water they ranged from 114.2 to 129.0 ppm and from ?44.9 to ?33.4 ‰, respectively. Isotope analyses were confirmed as a reliable tool for assessing authenticity of canned products. The method enables detection of synthetic acetic acid addition into vinegars and canned vegetables containing vinegar pickle up from 20 % (of total acidity).  相似文献

Beside ethanol, carbohydrates are the main source of total energy in beer. While analyses of fermentable carbohydrates are important from the technological point of view, the total content of carbohydrates is relevant in terms of nutrition. A high-performance liquid chromatography (HPLC) method with refractometric (RI) detection was developed for determination of total carbohydrate content in beer. Using enzymatic reaction with amyloglucosidase, the carbohydrates were cleaved to yield glucose and short glucose oligomers of less than 10 units, and separated on HPLC ion exchanger Rezex RSO-Oligosaccharide column in Ag+ mode. Optimum parameters were established for the enzymatic sample treatment and for the HPLC separation of reaction products. Calibration curves of glucose, fructose, maltose and simultaneously analyzed glycerol ranged from 0.001 to 0.5 g/100 ml, correlation coefficients of all calibration curves were 0.9999. The instrumental limits of quantification were 0.001 g/100 ml and they were verified using repetitive injections, with coefficient of variation (CV) less than 10 % in five replicates. The method limit of quantification was 0.01 g/100 ml since it was necessary to dilute the beer samples before chromatographic analysis. Recovery of the method in non-alcoholic and alcoholic beer was 98.5 %, and 92.3 %, respectively. Finally, ten non-alcoholic and 15 alcoholic beers from Czech market were analyzed using the method, the average content of total carbohydrates in non-alcoholic and alcoholic beers being 4.21 and 3.70 g/100 ml, respectively. These results are in a good correlation with the real extract of beer, which is on average 4.58 and 4.27 g/100 ml.  相似文献

Common starter cultures used in fermented mutton sausages were substituted by probiotic strains of Lactobacillus acidophilus CCDM 476 and Bifidobacterium animalis 241a. Technological properties of the traditional and the probiotic sausages were compared. The potential probiotic effect was evaluated by enumeration of bifidobacteria and lactobacilli in stool samples of 15 volunteers before and after a 14-day consumption period. The numbers of lactobacilli (107 cfu/g) and bifidobacteria (103 cfu/g) in the final product did not affect the technological properties. The use of L. acidophilus as a starter culture was found more beneficial than the use of B. animalis. Even after 60 days of storage, high counts of L. acidophilus (106 cfu/g) were detected; on the other hand, the counts of B. animalis were under the detection limit. Regarding sensory properties, the probiotic products showed better texture, and, curiously, a reduction of the typical smell of mutton. The numbers of lactobacilli in stool samples increased significantly after the consumption of the probiotic sausages.  相似文献

The aim of the present study was to analyze sugar levels (namely maltose, maltotriose, glucose and fructose) and alcohols (ethanol and glycerol) during the fermentation process in wort samples by amperometric enzymatic biosensors developed by our research group for industrial application, HPLC and spectrophotometry, and to compare the suitability of the presented methods for determination of individual analytes. We can conclude that for the specific monitoring of maltose or maltotriose only the HPLC method was suitable. On the other hand, biosensors and spectrophotometry reflected a decrease in total sugar concentration better and were able to detect both glucose and fructose in the later stages of fermentation, while HPLC was not. This can be attributed to the low detection limits and good sensitivity of the proposed methods. For the ethanol and glycerol analysis all methods proved to be suitable. However, concerning the cost expenses and time analysis, biosensors represented the best option.  相似文献

BACKGROUND: This study attempts to compare two possibilities of enhancing the colour of egg yolk. One of them is based on the ecological rearing of laying hens on natural green grass whereas the other uses a feeding dose supplemented with natural pigments in laying hens reared in individual cages. Is it possible to distinguish these two technologies using yolk colour determination in the CIELAB system? RESULTS: Yolk colour parameters such as L*, a*, and b* in the group of grazed hens are significantly different (α = 0.001) from those observed in hens reared in cages. The yolk colour shows a darker, redder and more yellow colour. The greatest difference was seen in the red colour parameter, a*, that increased more than twice. Visually, this means a shift towards a more orange colour. Compared to grazing in the meadow (ΔE* = 13.257), the addition of artificial pigments in the feed resulted in a more significant increase in the parameter ΔE* (CIE total colour difference), with the greatest value of ΔE* being observed with the use of both pigments (ΔE* = 24.265). CONCLUSION: Grazing increases the parameter a* whereas the values of the parameter C*ab remain relatively low. The parameter ΔE* is significantly lower in the case of grazing as compared to the supplementation of the feed with pigments. However, colourity parameters cannot be used as a specific standard to identify a particular grazing technology as their values vary during the laying period. Hops contain large amounts of secondary metabolites, many of which have notable bioactive and sensory characteristics. Many of these properties are affected by the processing of raw hops into products. We studied the influence of high‐pressure processing (HPP) on the content and composition of secondary metabolites in hop homogenates prepared from fresh green cones of several Czech hop varieties. Homogenates contained more hop oils (27% on average) compared to dried hops. The composition of essential oils in homogenates after HPP showed a decrease in fatty acid methyl and thioesters fractions (80 and 100% respectively). Conversely, the number of other bioactive compounds from the group of resins and prenylflavonoids that remained in HPP homogenates was retained to a greater extent than in the dried hops.
