Angiostatin, a proteolytic fragment of plasminogen, inhibits the growth of primary and metastatic tumors by suppressing angiogenesis. When used in combination with ionizing radiation (IR), angiostatin demonstrates potent antitumor synergism, largely caused by inhibition of the tumor microvasculature. We report here the temporal interaction of angiostatin and IR in Lewis lung carcinoma (LLC) tumors growing in the hind limbs of syngeneic mice. Tumors with an initial mean volume of 510 +/- 151 mm3 were treated with IR alone (20 Gy x 2 doses on days 0 and 1), angiostatin alone (25 mg/kg/day divided twice daily) on days 0 through 13, or a combination of the two as follows: (a) IR plus angiostatin (days 0 through 13); (b) IR plus angiostatin (days 0 and 1); and (c) IR followed by angiostatin beginning on the day after IR completion and given daily thereafter (days 2 through 13). By day 14, tumors in untreated control mice had grown to 6110 +/- 582 mm3, whereas in mice treated with: (a) IR alone, tumors had grown to 2854 +/- 338 mm3 (P < 0.05 compared with untreated controls); and (b) angiostatin alone, tumors had grown to 3666 +/- 453 mm3 (P < 0.05 compared with untreated controls). In combined-treatment groups, in mice treated with: (a) IR plus longer-course angiostatin, tumors reached 2022 +/- 282 mm3 (P = 0.036 compared with IR alone); (b) IR followed by angiostatin, tumors reached 2677 +/- 469 mm3 (P > 0.05 compared with IR alone); and (c) IR plus short-course angiostatin, tumors reached 1032 +/- 78 mm3 (P < 0.001 compared with IR alone). These findings demonstrate that the efficacy of experimental radiation therapy is potentiated by brief concomitant exposure of the tumor vasculature to angiostatin.  相似文献

We report a very efficient semianalytical approach for the performance evaluation of differential detection schemes for GMSK signals of the DECT standard. Precisely, for a given channel, the performance is determined by means of an analytical procedure which includes the saddlepoint approximation. We consider both static channels (with impulse response generated by the simulation program SIRCIM) and two-ray Rayleigh and log-normal fading channels. As a departure from previous works, our receiver includes an all-digital part after the analog differential detection scheme. The digital part includes: (1) a block for the estimation of both the optimum sampling phase and the nonlinear channel coefficients (by making use of the DECT training sequence), (2) a one-tap decision feedback (DF) equalizer, and (3) a block for the evaluation of the approximate optimum bias level (γ e) in the threshold detector. Both the DF equalizer coefficient and γe are based on the nonlinear channel coefficients estimate. For channels with a normalized delay spread up to 0.2, the use of the optimum threshold together with the DF equalizer permits a gain of about 2 dB at BER=10-6 with respect to a receiver without equalization and a zero-level decision threshold. In addition, we discover that, in indoor environments, the 2-bit GMSK detector performs roughly the same as the 1-bit detector. The threshold optimization is also effective in the presence of channels affected by fading. To support this statement, we report the performance of the 1-bit differential detection scheme combined with antenna selection diversity in the presence of a two-ray log-normal and Rayleigh fading channel  相似文献

During the molding of industrial parts using injection molding, the molten polymer flow through converging and diverging sections as well as in areas presenting thickness and flow direction changes. A good understanding of the flow behavior and thermal history is important in order to optimize the part design and molding conditions. This is particularly true in the case of automotive and electronic applications where the coupled phenomena of fluid flow and heat transfer determine to a large extent the final properties of the part. This paper presents a 3D finite element model capable of predicting the velocity, pressure, and temperature fields, as well as the position of the flow fronts. The velocity and pressure fields are governed by the generalized Stokes equations. The fluid behavior is predicted through the Carreau Law and Arrhenius constitutive models. These equations are solved using a Galerkin formulation. A mixed formulation is used to satisfy the continuity equation. The tracking of the flow front is modeled by using a pseudo-concentration method and the model equations are solved using a Petrov-Galerkin formulation. The validity of the method has been tested through the analysis of the flow in simple geometries. Its practical relevance has been proven through the analysis of an industrial part.  相似文献

This study was performed to contribute to the analysis of alpha-lactalbumin "molten globule" state by using spectral and proteolysis techniques. Samples of holo and apo alpha-lactalbumin in the presence of different concentrations of ethanol were analyzed. Results of fluorescence spectroscopy of both forms showed that as ethanol concentration increased, the tryptophanyl residues became more accessible to the solvent. Near circular dichroism spectra of holo alpha-lactalbumin indicated that its tertiary structure was maintained in 20% ethanol whereas it was altered in 30 and 40% ethanol. For apo alpha-lactalbumin, spectra were similar in all samples studied. Holo alpha-lactalbumin was resistant to trypsinolysis in 0% ethanol, whereas it was easily hydrolyzed in 20 and 30% ethanol. In the case of the apo form and in the absence of ethanol, 70% of the protein was degraded after 1 h. However, in the presence of 20 and 30% ethanol, the overall reaction rate was lowered. Peptides obtained after tryptic hydrolysis were identified by reversed-phase high-performance liquid chromatography coupled to mass spectrometry. Differences in population of produced peptides indicate the changes of folding intermediates present in the studied alpha-lactalbumin solutions. This study demonstrated that proteolytic enzymes are suitable tools to determine protein structure complementing physico-chemical studies.  相似文献

A 28-year-old man with Wiskott-Aldrich syndrome presented with ulcerative-proliferative lesions on his face from which herpes simplex type 1 (HSV-1) was isolated. He was initially treated with 10 mg/kg of acyclovir (Zovirax) intravenously every 8 h, but his skin lesions worsened. Clinical resistance to acyclovir was suspected, and therapy with this drug was intensified. The dosage of acyclovir was increased to 45 mg/kg, administered by continuous infusion, and the lesions subsequently resolved. The strain of HSV recovered from the patient showed acyclovir-resistance in vitro, using the colorimetric method with neutral red. Herpes simplex virus resistance to acyclovir is rare. It is more common in immunocompromised patients if subtherapeutic doses are administered in the treatment of chronic persistent forms of infection. Whenever clinical resistance to acyclovir is suspected, the dosage should be increased to 2 mg/kg per h administered via an infusion pump. If no improvement is observed in the patient's condition with this regimen, a phosphorylated medication whose mechanism of action is not dependent on viral thymidine kinase, such as foscarnet (phosphonoformic acid), should be substituted.  相似文献

The synthesis, processing, and characterization of new crosslinked functional polymer thin films derived from fumaryl chloride and maleic anhydride is presented. Experimental data demonstrated that this is a versatile, convenient, and cost‐effective method of fabricating ultrastructure crosslinked and functional polymer thin films for potential nonlinear optical (NLO) or other applications where molecular orientation is required. The unsaturated and processable polyester thin films are capable of crosslinking in air to form a hardened lattice under a variety of conditions, including both thermal and photoinitiated crosslinking. The thermal stability of the second harmonic (SHG) signal for a crosslinked NLO thin film was stable at temperatures up to 150°C, which is in contrast to uncrosslinked polymers whose SHG signals typically decreased over 50% below 100°C. Because of the lack of NH/OH groups and their vibrational overtones in the polymer, these crosslinked polyester systems have a great potential for low optical loss applications at 1550 nm communication wavelength. © 2004 Wiley Periodicals, Inc. A counter flow membraneless microfluidic fuel cell is presented, where a non-reacting electrolyte separates the reacting streams. In this fuel cell design, vanadium reactants flow through porous carbon electrocatalysts. A sulfuric acid stream is introduced in the gap between the electrodes and diverts the reactants to opposite and independent outlets. This fuel cell differs from other membraneless designs in its ability to maintain a constant separation between the reactants without diffusive mixing.  相似文献

In light of their environmental and economic interests, food byproducts have been increasingly exploited and valorized for their richness in dietary fibers and antioxidants. Phenolic compounds are antioxidant bioactive molecules highly present in grape byproducts. Herein, the accelerated solvent extraction (ASE) of phenolic compounds from wet and dried grape pomace, at 45 °C, was conducted and the highest phenolic compounds yield (PCY) for wet (16.2 g GAE/100 g DM) and dry (7.28 g GAE/100 g DM) grape pomace extracts were obtained with 70% ethanol/water solvent at 140 °C. The PCY obtained from wet pomace was up to two times better compared to the dry byproduct and up to 15 times better compared to the same food matrices treated with conventional methods. With regard to Resveratrol, the corresponding dry pomace extract had a better free radical scavenging activity (49.12%) than the wet extract (39.8%). The drying pretreatment process seems to ameliorate the antiradical activity, especially when the extraction by ASE is performed at temperatures above 100 °C. HPLC-DAD analysis showed that the diversity of the flavonoid and the non-flavonoid compounds found in the extracts was seriously affected by the extraction temperature and the pretreatment of the raw material. This diversity seems to play a key role in the scavenging activity demonstrated by the extracts. Our results emphasize on ASE usage as a promising method for the preparation of highly concentrated and bioactive phenolic extracts that could be used in several industrial applications.  相似文献
