Continuous manufacturing and closed-loop quality control are emerging technologies that are pivotal for next-generation pharmaceutical modernization. We develop a process control framework for a continuous carousel for integrated filtration-drying of crystallization slurries. The proposed control system includes model-based monitoring and control routines, such as state estimation and real-time optimization, implemented in a hierarchical, three-layer quality-by-control (QbC) framework. We implement the control system in ContCarSim, a publicly available carousel simulator. We benchmark the proposed control system against simpler methods, comprising a reduced subset of the elements of the overall control system, and against open-loop operation (the current standard in pharmaceutical manufacturing). The proposed control system demonstrates superior performance in terms of higher consistency in product quality and increased productivity, proving the benefits of closed-loop control and of model-based techniques in pharmaceutical manufacturing. This study represents a step forward toward end-to-end continuous pharmaceutical processing, and in the evolution of quality-by-design toward quality-by-control. 相似文献

A photoacoustic spectroscopic (PAS) and a direct optical absorption spectroscopic (OAS) gas sensor, both using continuous-wave room-temperature diode lasers operating at 1531.8 nm, were compared on the basis of ammonia detection. Excellent linear correlation between the detector signals of the two systems was found. Although the physical properties and the mode of operation of both sensors were significantly different, their performances were found to be remarkably similar, with a sub-ppm level minimum detectable concentration of ammonia and a fast response time in the range of a few minutes. 相似文献

Thermal evolution of amorphous calcium phosphate (ACP) powder from a fast nitrate synthesis with a Ca/P ratio of 1:1 were studied in the range of 20–980?°C. The powder consisted of amorphous dicalcium phosphate anhydrate (CaHPO4) after heating to 200?°C. CaHPO4 gradually condensed to amorphous calcium pyrophosphate Ca2P2O7 (CPP) between 200 to 620?°C. Amorphous CPP crystallized at 620–740?°C to a metastable polymorph α′-CPP of the high-temperature phase α-CPP and β-CPP. The α′-CPP/ β-CPP phase ratio reached a maximum at 800?°C (60?wt% α′-CPP/40?wt% β-CPP), and α′-CPP gradually transformed to β-CPP at a higher temperature. Some β-TCP occurred at 900?°C, so that a three-phasic mixture was obtained in the powder heated to 980?°C. The occurrence of metastable α′-CPP is attributed to Ostwald’s step rule, and a mechanism for β-TCP formation is proposed. The advantages of prospective biomaterials from these powders are discussed. 相似文献

A new mixer/modulator for millimetre-wave applications is presented. The circuit is implemented with a combination of finlines, coplanar lines and microstrip lines. By applying the described mixer/modulator to millimetre-wave systems a twofold use of active elements such as the Gunn element and Schottky diodes is possible. Cerium oxide (CeO2), nanoparticles were prepared using chitosan as template, cerium nitrate as a starting material and sodium hydroxide as a precipitating agent. The resultant ceria–chitosan spheres were calcined at 350 °C. The synthesized powders were characterized by, XRD, HRTEM, UV–vis, FTIR, and TG-DTA. The average size of the nanoparticles obtained was ∼4 nm and BET specific surface area ∼105 m2 g−1. Blueshifts in the ultraviolet absorption spectra have been observed in cerium oxide nanocrystallites. The band-gap was found to be 4.5 eV. The blueshifts are well explained for diameters down to less than a few nanometers by the change in the electronic band structure. 相似文献

The expression of the membrane ABCB1 transporter in neoplastic cells is one of the most common causes of reduced sensitivity to chemotherapy. In our previous study, we investigated the effect of a single culture of ABCB1-negative (S) and ABCB1-positive variants of L1210 cells (R and T) in the presence of sulforaphane (SFN). We demonstrated that SFN induces the onset of autophagy more markedly in S cells than in R or T cells. In the current study, we focused on the effect of the repeated culture of S, R and T cells in SFN-containing media. The repeated cultures increased the onset of autophagy compared to the simple culture, mainly in S cells and to a lesser extent in R and T cells, as indicated by changes in the cellular content of 16 and 18 kDa fragments of LC3B protein or changes in the specific staining of cells with monodansylcadaverine. We conclude that SFN affects ABCB1-negative S cells more than ABCB1-positive R and T cells during repeated culturing. Changes in cell sensitivity to SFN appear to be related to the expression of genes for cell-cycle checkpoints, such as cyclins and cyclin-dependent kinases. 相似文献
