(1) Background: The size and surface charge are the most significant parameters of nanocarriers that determine their efficiency and potential application. The poor cell uptake of encapsulated drugs is the main limitation in anticancer treatment. The well-defined properties of nanocarriers will enable to target specific tissue and deliver an active cargo. (2) Methods: In the current study, poly(D,L -lactide) (PLA) nanocarriers loaded with curcumin (CUR) and differing surface charge were evaluated for transport efficacy in combination with electroporation (EP) in dependence on the type of cells. The obtained CUR-loaded nanoparticles with diameters ranging from 195 to 334 nm (derived from dynamic light scattering (DLS)) were characterized by atomic force microscopy (AFM) (morphology and shape) and Doppler electrophoresis (ζ-potential) as well as UV-vis spectroscopy (CUR encapsulation efficiency (about 90%) and photobleaching rate). The drug delivery properties of the obtained PLA nanocarriers enhanced by electroporation were assessed in human colon cancer cells (LoVo), excitable normal rat muscle cells (L6), and free of voltage-gated ion channels cells (CHO-K1). CLSM studies, viability, and ROS release were performed to determine the biological effects of nanocarriers. (3) Results: The highest photodynamic activity indicated anionic nanocarriers (1a) stabilized by C12(COONa)2 surfactant. Nanocarriers were cytotoxic for LoVo cells and less cytotoxic for normal cells. ROS release increased in cancer cells with the increasing electric field intensity, irradiation, and time after EP. Muscle L6 cells were less sensitive to electric pulses. (4) Conclusions: EP stimulation for CUR-PLA nanocarriers transport was considered to improve the regulated and more effective delivery of nanosystems differing in surface charge. 相似文献

Tellurite glasses with the composition of xTm_2O_3-(6-x)Y_2O_3-3Na_2O-25ZnO-66TeO_2(where 0≤x≤6)were obtained by the melt-quenching technique.Absorption(300 K),excitation(300 K) and fluorescence spectra(300 K) as well as fluorescence decay curves of Tm~(3+)-doped title glasses are presented and discussed in details.The Judd-Ofelt analysis based on the room temperature absorption spectrum was applied for determination of fundamental fluorescence properties such as radiative transition probabilities(A_T),branching ratios(β_R),radiative lifetimes(τ_R) of the emitting levels of the Tm~(3+) ion and stimulated emission cross-sections(σ_(em)).Fluorescence spectra were recorded and analysed in the visible and near-infrared spectral range.The emission and effective cross-section were calculated for the ~3F_4→~3H_6 transition,showing that the investigated glasses are promising laser host materials,operating at 1.8 μm.The observed concentration quenching and non-exponential decay curves from the ~1 G_4 and ~3H_4 states indicate nonradiative energy transfer between Tm~(3+) ions.The analysis of non-exponential fluorescence decay curves from the ~1 G_4 and ~3H_4 levels was carried out in framework of the InokutiHirayama and Yokota-Tanimoto models and energy transfer microparameters were determined.The self-quenching model was proposed for describing relaxation of the first excited state of the Tm~(3+) ion. 相似文献

The structure of cell wall of yeasts (genus Saccharomyces) is one of the factors that determine their health-promoting properties connected to the presence of β-glucans and mannoprotein. The aim of the study was to determine the influence of glycerol as a carbon source on structural polymers of cell wall (β-glucan and mannoprotein) of probiotic yeasts Saccharomyces cerevisiae var. boulardii and brewer’s yeasts S. cerevisiae R9. Significant increase of the percentage of polysaccharide content in the cell wall dry weight of S. cerevisiae R9 brewer’s yeasts was noted (in the range of 10–20 %) after cultivation in medium containing glycerol at a concentration of 2–5 % and pH 4.0. The highest content of carbohydrates in probiotic yeasts’ cell wall (58 %) was observed after cultivation in medium containing 3 % of glycerol and pH 5.0. The cell wall of probiotic yeasts was characterized by higher content of mannoprotein comparing with cell wall preparation of brewer’s yeasts S. cerevisiae R9 composed mainly of β-glucans. After cultivation in mediums with 2 and 3 % of glycerol, the cell of brewer’s yeasts contained the highest amount of β(1,3/1,6)-glucan in dry weight of the cell wall (about 36 %). Glycerol at a concentration of 3 and 5 % also intensified mannoprotein biosynthesis in cell wall of S. cerevisiae R9, approximating their content to those noted in the cells of probiotic yeasts (about 29 % (w/w) of dry weight of the cell wall) after cultivation in a medium of pH 5.0 containing 3 % of glycerol. 相似文献

Campylobacter-associated gastroenteritis remains an important cause of morbidity worldwide, and some evidence suggests that poultry is an important source of this foodborne infection in humans. This study was conducted to analyze the prevalence and genetic background of resistance of 149 Campylobacter jejuni and 54 Campylobacter coli strains isolated from broiler chicken carcasses and from stool samples of infected children in Poland from 2003 through 2005. Nearly all isolates were susceptible to macrolides and aminoglycosides. The highest resistance in both human and chicken strains was observed for ciprofloxacin (more than 40%), followed by ampicillin (13 to 21%), and tetracycline (8 to 29%). Resistance to ampicillin and tetracycline rose significantly between 2003 and 2005. Slight differences in resistance between human and chicken isolates indicate that although chicken meat is not the only source of Campylobacter infection in our population, it can be involved in the transmission of drug-resistant Campylobacter strains to humans. 相似文献

The paper presents the analysis of the profile composition of fatty acids in the molecules of phosphatidylcholine and phosphatidylethanolamine, by using hydrophilic interaction liquid chromatography and gas chromatography coupled with mass spectrometry. The profiles of 15 phosphatidylcholine and 8 phosphatidylethanolamine species were analyzed with a newly developed hydrophilic interaction liquid chromatography (HILIC)–electrospray ionization (ESI)–tandem mass spectroscopy (MS/MS) method, by using a new stationary bonded phase. The application of the new method in control and experimental groups of egg yolk revealed significant differences in the composition of phospholipid species containing mainly polyunsaturated fatty acids. Additionally, using GC-MS, the profile of fatty acids in four groups with different dietary supplementation of hens was analyzed and 20 fatty acids in egg yolks were determined. Monounsaturated fatty acids were found in higher amounts than saturated fatty acids and polyunsaturated fatty acids in egg yolks. Oleic acid (18:1) was the major monounsaturated fatty acid in egg yolk while palmitic acid (16:0) was the major saturated fatty acid. Linoleic acid (18:2), arachidonic acid (20:4), and docosahexaenoic acid (22:6) reached the highest levels among the polyunsaturated fatty acids. 相似文献

Dihydroxyacetone (1,3-dihydroxy-2-propanone, DHA) is applied in the food and cosmetic industries as well as in pharmacy and medicine. It is produced as a result of incomplete oxidation of glycerol by acetic acid bacteria Gluconobacter oxydans. This reaction is catalyzed by PQQ-dependent membrane-bound glycerol dehydrogenase. The research developed a method of obtaining DHA by oxidation of a 3?% aqueous solution of glycerol (pH 7.5) at a temperature of 23?°C, with the only reaction biocatalyst being an immobilized cell preparation obtained from G. oxydans cells. After 5?days of the process, DHA concentration in the solution accounted for 27.2?g/L and the reaction efficiency for 94?%. After 4?days of the reaction run in culture media with pH 5.0, at a temperature of 28?°C, free or immobilized cells of G. oxydans produced on average 25?g of DHA/L at the reaction efficiency of 87?%. 相似文献

In 2007 and 2008, a monitoring study was carried out in Poland to examine the occurrence of thermotolerant Campylobacter spp. in raw and cooked chicken products available on the retail market. A total of 912 samples were tested: 443 samples of raw chicken meat, 146 samples of giblets, and 323 ready-to-eat poultry products (150 samples of spit-roasted chicken, 56 samples of smoked chicken, and 117 samples of paté and cold meats). A high level of contamination of raw chicken meat (51.7% of samples) and chicken giblets (47.3% of samples) was detected. The study focused on the comparison of carcass and fillet yields, fillet color, the content of basic components, and the fatty acid composition of tissue from cultivated and wild pikeperch. There was no variation in yield between carcasses and fillets. The brightness and redness of the color of the fillet surfaces of both types of fish were comparable, although the yellowness, chromaticity and shade of color differed. The tissue of cultivated fish had three times more fat (2.87%) than did that of wild fish; the difference was compensated for by water content. The protein and mineral component contents did not vary significantly. The total levels of saturated and monounsaturated acids in the tissue lipids in 1 g of fat varied. No differences were observed between the total amount of polyunsaturated fatty acids (PUFA), PUFAn-3, PUFAn-6, or in the n-3/n-6 proportion. One consequence of the higher fat content in cultivated pikeperch tissue was that the contents of eicosapentaenoic (EPA; 20:5n-3) and docosahexaenoic (DHA; 22:6n-3) acids per 100 g of fillet were significantly higher than in the tissue of wild pikeperch. 相似文献
