Two extraction techniques, dynamic headspace extraction (DHE) and solid-phase microextraction (SPME), were compared to assess the effect of high-pressure treatment (400 MPa, 10 min, 12 °C) on the volatile compounds of low-acid fermented sausage “espetec” and sliced cooked pork shoulder stored at 4 °C. DHE was more efficient at extracting low-boiling compounds such as ethanal, 2,3-butanedione and alcohols, while SPME extracted more efficiently a higher number of chemical families, especially fatty acids. The effect of pressurisation on the volatile fraction of “espetec” was better categorized by DHE, whereas SPME was more appropriate for cooked pork shoulder. The volatile fraction of “espetec” changed slightly after pressurisation, mainly showing a decrease in the levels of lipid-derived compounds, like linear alkanes, aldehydes, or 1-alcohols in pressurised samples. The volatile profile of cooked pork shoulder underwent substantial changes during refrigerated storage, mainly due to microbial metabolism, most of these changes being limited by HPP.  相似文献

The bactericidal activity of lactoferrin (LF), amidated lactoferrin (AMILF), pepsin digested lactoferrin (PDLF), and its activated (ALF) commercial form, against six strains of three gram-positive bacterial species was investigated. Listeria monocytogenes was most sensitive in vitro, Staphylococcus aureus showed a moderate resistance, and Enterococus faecalis was highly resistant to antimicrobials. When chicken breast fillets were inoculated with L. monocytogenes CECT5725 and treated with antimicrobials, reductions were below 0.5 log CFU/ml in all cases. In combination with high pressure (HHP) treatment at 400 MPa for 10 min, antimicrobials showed a slight additional bactericidal effect, always below 1 log CFU/g. Incorporation of antimicrobials 18 h before or 1 h after HHP treatment generally yielded better results than incorporation 1 h before HHP treatment, although reductions remained below 1.5 log CFU/g in all cases. LF and its derivatives showed a limited potential for pathogen control in meat.  相似文献

The United States Environmental Protection Agency has identified quantification of trichloroethylene (TCE), an industrial solvent, in breast milk as a high priority need for risk assessment. Water and milk samples were collected from 20 households by a lactation consultant in Nogales, Arizona. Separate water samples (including tap, bottled, and vending machine) were collected for all household uses: drinking, bathing, cooking, and laundry. A risk factor questionnaire was administered. Liquid-liquid extraction with diethyl ether was followed by GC-MS for TCE quantification in water. Breast milk underwent homogenization, lipid hydrolysis, and centrifugation prior to extraction. The limit of detection was 1.5 ng/mL. TCE was detected in 7 of 20 mothers' breast milk samples. The maximum concentration was 6 ng/mL. TCE concentration in breast milk was significantly correlated with the concentration in water used for bathing (ρ = 0.59, p = 0.008). Detection of TCE in breast milk was more likely if the infant had a body mass index <14 (RR = 5.2, p = 0.02). Based on average breast milk consumption, TCE intake for 5% of the infants may exceed the proposed U.S. EPA Reference Dose. Results of this exploratory study warrant more in depth studies to understand risk of TCE exposures from breast milk intake.  相似文献

Pediocin PA-1 is an antimicrobial peptide produced by lactic acid bacteria (LAB) that has been sufficiently well characterised to be used in food industry as a biopreservative. Sulphur dioxide is the traditional antimicrobial agent used during the winemaking process to control bacterial growth and wine spoilage. In this study, we describe the effect of pediocin PA-1 alone and in combination with sulphur dioxide and ethanol on the growth of a collection of 53 oenological LAB, 18 acetic acid bacteria and 16 yeast strains; in addition, production of pediocin PA-1 by Pediococcus acidilactici J347-29 in presence of ethanol and grape must is also reported. Inhibitory concentrations (IC) and minimal bactericide concentrations of pediocin PA-1 were determined against LAB, and revealed a bacteriostatic effect. Oenococcus oeni resulted more sensitive to pediocin PA-1 (IC50 = 19 ng/ml) than the other LAB species (IC50 = 312 ng/ml). Cooperative inhibitory effects of pediocin PA-1 and either sulphur dioxide or ethanol were observed on LAB growth. Moreover, the pediocin PA-1 producing P. acidilactici strain J347-29 was able to grow and produce the bacteriocin in presence of ethanol (up to 4% ethanol in the fermentation broth) and grape must (up to 80%), which indicated that pediocin PA-1 can be considered as a potential biopreservative in winemaking.  相似文献

Due to the increasing interest on laterally diffused MOS (LDMOS) transistors as a part of power electronics in the high energy physics (HEP) experiments, the effect of total ionising dose (TID) on their electrical performances has been experimentally measured. The analysis of the experimental results requires the aid of physics-based simulations to study the impact of TID effects on the LDMOS drift oxide layer. In this work, a simulation methodology is developed in order to analyse the changes in the electric field distribution as a consequence of the TID induced trapped charge, and its relationship with the technological parameters and the bias conditions. Emerging evidence has suggested that dysbiosis of the gut microbiota may influence the drug efficacy of colorectal cancer (CRC) patients during cancer treatment by modulating drug metabolism and the host immune response. Moreover, gut microbiota can produce metabolites that may influence tumor proliferation and therapy responsiveness. In this study we have investigated the potential contribution of the gut microbiota and microbial-derived metabolites such as short chain fatty acids and polyamines to neoadjuvant radiochemotherapy (RCT) outcome in CRC patients. First, we established a profile for healthy gut microbiota by comparing the microbial diversity and composition between CRC patients and healthy controls. Second, our metagenomic analysis revealed that the gut microbiota composition of CRC patients was relatively stable over treatment time with neoadjuvant RCT. Nevertheless, treated patients who achieved clinical benefits from RTC (responders, R) had significantly higher microbial diversity and richness compared to non-responder patients (NR). Importantly, the fecal microbiota of the R was enriched in butyrate-producing bacteria and had significantly higher levels of acetic, butyric, isobutyric, and hexanoic acids than NR. In addition, NR patients exhibited higher serum levels of spermine and acetyl polyamines (oncometabolites related to CRC) as well as zonulin (gut permeability marker), and their gut microbiota was abundant in pro-inflammatory species. Finally, we identified a baseline consortium of five bacterial species that could potentially predict CRC treatment outcome. Overall, our results suggest that the gut microbiota may have an important role in the response to cancer therapies in CRC patients.  相似文献

The first step of this work is to study the susceptibility of a radiofrequency oscillator to deterministic disturbance sources. A Colpitts oscillator, working around a 4 GHz frequency, contains a heterojunction bipolar transistor with a silicon–germanium base as an active device. A mixed‐mode analysis is involved, applying a microscopic drift diffusion model to the device, whereas the rest of the circuit used is governed by Kirchhoff's laws. We assume that this tool is very relevant to grasp the influence of intrinsic or extrinsic noisy sources of the oscillator. Our first simulation raw results motivate us to discuss, and perhaps extend, via some analytical models, the so‐called impulse sensitivity function model. Control of meat shelf-life includes the time that it remains in the exhibitor of sale (such as the supermarket) until its rejection for the consumer, or withdrawal due to expiry date. Near infrared spectroscopy (NIRS) is one of the most promising techniques for large-scale meat quality control. This study investigated the potential of on-site NIRS portable instrumentation-based models to predict three microbiological parameters to establish if pork meat is acceptable or not for consumption (aerobic Mesophilous microorganisms, Enterobacteriaceae, and lactic acid bacteria) and pH to quality control food preservation and shelf-life extension on intact slices of pork meat packaged under two different modified atmospheres. NIR calibrations were developed by using an on-site Phazir instrument (Polychromix, Wilmington, MA, USA) in the range 1,600–2,400 nm. A total of 252 samples of pork meat slices were directly scanned twice in reflectance mode on trays, once before and another one after removing the film cover at 1, 3, 5, 7, 9, 12, and 15 days of storage. Results showed that spectra of meat acceptable or not for consumption have marked differences around 1,660 nm. NIRS quantitative prediction models showed r 2 values between 0.19 and 0.65 for the microbiological parameters assayed. The developed NIRS methodology makes possible on-site prediction of microbiological status of pork meat with a standard error of cross-validation around 1 log cfu/g. Results have shown that modified atmosphere packaging has no influence on calibration statistics.  相似文献

The effect of high-pressure processing (pressure levels of 400, 500 and 600 MPa, and exposure times of 5 and 10 min) on the volatile profile of vacuum-packaged sliced cooked pork shoulder held for 28 days at 4 °C was assessed. The volatile fraction of pressurized samples scarcely changed immediately after treatment and remained stable for 14 days, regardless the pressure and time of exposure. After 21 days of storage, significant differences were observed in the profile of volatile compounds in pressurized samples as compared with control samples, these differences being treatment dependent. At the end of the storage period, control and 400 MPa samples showed higher levels of acetic and fatty acids, ethanol and ethyl esters, whereas 500 and 600 MPa samples contained higher levels of ethanal, branched-chain aldehydes, diacetyl, acetoin, and 2,3-butanediol among other compounds. These results suggest that the high-pressure treatment had a discriminant effect on the microbiota of cooked pork shoulder, which led to the accumulation of different volatile compounds during the refrigerated storage of control and pressurized samples.  相似文献
