Matrix metalloproteinases are thought to be key players in the remodelling activity of cells associated with both physiological and pathological processes. They share a relatively conserved structure with a number of identifiable modules linked to their specific functions. The structure of the individual domains of a number of matrix metalloproteinases have now been elucidated. Here we review these data in the light of complementary studies on the behaviour of these enzymes in biological systems. In particular we focus on the C-terminal hemopexin-like domain which has intriguingly specific roles in individual matrix metalloproteinases.  相似文献

From the analysis of the temperature dependence of the EPR line width, an estimate of the anisotropic parts of the dipolar and quadrupolar hyperfine couplings of lanthanum was derived. BACKGROUND: Alveolar macrophages from patients with sarcoidosis were analyzed for their ability to secrete tumor necrosis factor-alpha (TNF-alpha), interleukin-1-beta (IL-1-beta), and interleukin-6 (IL-6). RESULTS: Constitutive release of all three monokines in these patients was concomitantly increased in the active state of disease in comparison with inactive sarcoidosis or healthy control subjects. Alveolar macrophages from patients with inactive sarcoidosis compared with cells from healthy subjects showed increased spontaneous secretion of TNF-alpha and IL-6 only, whereas the constitutive release of IL-1-beta was similar as in healthy volunteers. In vitro stimulation of alveolar macrophages from healthy control subjects with lipopolysaccharide or pokeweed mitogen led to a time- and dose-dependent enhanced secretion of TNF-alpha, IL-1-beta, and IL-6. In a similar manner, with corresponding cells from patients with sarcoidosis the secretion of all three cytokines could be further increased by stimulation with lipopolysaccharide or pokeweed mitogen. CONCLUSIONS: The data presented indicate that an increased release of TNF-alpha, IL-1-beta, and IL-6 correlates to disease activity and may play a critical part in the pathogenesis of sarcoidosis.  相似文献

The rate of nosocomial respiratory syncytial virus (RSV) infection was measured in a large pediatric hospital using an incidence density method. The at-risk days for nosocomial RSV were summed during a defined winter period in which there were 54 admissions with community-acquired RSV infection giving a rate of 2.9 cases per 1,000 at-risk days (95% confidence interval, 0.3-5.4 per 1,000).  相似文献

The direct synthesis of hydrogen peroxide from H2 and O2 using a range of supported Au–Pd alloy catalysts is compared for different supports using conditions previously identified as being optimal for hydrogen peroxide synthesis, i.e. low temperature (2 °C) using a water–methanol solvent mixture and short reaction time. Five supports are compared and contrasted, namely Al2O3, -Fe2O3, TiO2, SiO2 and carbon. For all catalysts the addition of Pd to the Au only catalyst increases the rate of hydrogen peroxide synthesis as well as the concentration of hydrogen peroxide formed. Of the materials evaluated, the carbon-supported Au–Pd alloy catalysts give the highest reactivity. The results show that the support can have an important influence on the synthesis of hydrogen peroxide from the direct reaction. The effect of the methanol–water solvent is studied in detail for the 2.5 wt% Au–2.5 wt% Pd/TiO2 catalyst and the ratio of methanol to water is found to have a major effect on the rate of hydrogen peroxide synthesis. The optimum mixture for this solvent system is 80 vol.% methanol with 20 vol.% water. However, the use of water alone is still effective albeit at a decreased rate. The effect of catalyst mass was therefore also investigated for the water and water–methanol solvents and the observed effect on the hydrogen peroxide productivity using water as a solvent is not considered to be due to mass transfer limitations. These results are of importance with respect to the industrial application of these Au–Pd catalysts.  相似文献

E. J. Wisniewski and G. L. Murphy (see record 2004-22496-015) suggested that the apparent effects of relation frequency in C. L. Gagné and E. J. Shoben's (1997; see record 1997-02349-004) conceptual combination experiments could be explained by differences between the familiarity and plausibility of their stimuli (noun-noun phrases). However, C. L. Gagné and T. L. Spalding (see record 2006-20573-017) argued that Wisniewski and Murphy's measures of plausibility and frequency are both sensitive to relation frequency. They also suggested that the stimuli were mostly novel, such that differences in familiarity could not explain Gagné and Shoben's findings. The current authors focus on the theoretical rationale for the plausibility and familiarity variables, arguing that the original interpretation of their findings is correct. The ingestion of I-131 by pregnant women can have consequences for the developing foetus, in particular brain function. As the foetal thyroid accumulates iodine from the twelfth week of gestation onwards, the determination of foetal brain dose resulting from such I-131 accumulation is essential. Normal dosimetric methods fail to treat the case of foetus. Using an approximation method based on the MIRD approach, a foetal dose estimation scheme is developed to allow the determination of foetal brain dose from foetal thyroid irradiation. Dose values are obtained for the foetus based on the maternal intake of I-131. It was found that the choice of biokinetic model for the mother/foetus has a large impact on the determined dose estimate.  相似文献

When the phenotype of neurons in pre- and paravertebral sympathetic ganglia are compared, there are marked differences in NGF dependence, neuropeptide content, connectivity and electrophysiological properties. The trophic interactions that induce these differences are currently poorly understood. One explanation is that prevertebral neurons receive a second neurotrophic signal, other than NGF, from their target of innervation. If this is the case, neurons in the prevertebral ganglia should express another neurotrophin receptor, in addition to the NGF receptor (trkA). To test this prediction, the level of expression of three neurotrophin receptors, trkA, trkB and trkC, were examined in one paravertebral sympathetic ganglia, the SCG, and two prevertebral ganglia, the celiac and superior mesenteric ganglia. It was found that mRNA encoding the full-length form of the trkB receptor was barely expressed in the SCG. Significantly higher levels of full-length trkB mRNA expression were found in the prevertebral ganglia. Ligands of the trkB receptor may, therefore, contribute to the differentiation and/or survival of some prevertebral sympathetic neurons.  相似文献
