A decision support system for water quality issues in the Manzanares River (Madrid, Spain)

The Manzanares River, located in Madrid (Spain), is the main water supplier of a highly populated region, and it also receives wastewater from the same area. The effluents of eight Waste Water Treatment Plants (WWTPs) downstream of the river, which represent 90% of the flow in the middle and lower parts of the river, are the primary sources of water pollution. Although the situation has improved slightly in the last two years, the water in the river is highly polluted, making it uninhabitable for aquatic life. Water quality modelling is typically used to assess the effect of treatment improvements in water bodies. In this work, the GESCAL module of the Aquatool Decision Support System Shell was used to simulate water quality in the Manzanares River. GESCAL is appropriate for modelling in an integrated way water quality for whole water resources systems, including reservoirs and rivers. A model was built that simulates conductivity, phosphorous, carbonaceous organic matter, dissolved oxygen, organic nitrogen, ammonia, and nitrates. The period from October 2006 to September 2008 was selected for calibration due to the many treatment modifications that occurred during this time. An earlier and longer period, from October 2000 to September 2006, was used for validation. In addition, a daily model was used to analyse the robustness of the GESCAL model. Once the GESCAL model was validated, different scenarios were considered and simulated. First, different combinations of nutrient elimination among the different WWTPs were simulated, leading to the conclusion that investments have to focus on three of the proposed WWTPs. Moreover, these treatments will not be sufficient to maintain fish habitat conditions at all times. Additional measures, such as the increment of the flow in the river or oxygen injection, were simulated. Incrementing the flow of the Manzanares River has been shown to be an efficient means of increasing water quality, but this implies an increment in the risk of water scarcity situations in the Madrid water supply system.     

Antimicrobial peptides and their superior fluorinated analogues: structure-activity relationships as revealed by NMR spectroscopy and MD calculations

The conformations of two synthetic pentapeptides with antimicrobial activity and their 4-fluorophenylalanine (Pff)-containing analogues (ArXArXAr-NH(2); Ar=Phe, Pff; X=Lys, Arg) have been studied. NMR experiments carried out both in aqueous fluoroalcohol solutions and SDS micelles permitted their interactions with membrane-like environments to be explored. WaterLOGSY experiments and Mn(2+)-based paramagnetic probes were also applied to assess their orientations with respect to the SDS micelles. In addition, pulse-field gradient (PFG) diffusion NMR spectroscopy studies were conducted, under different experimental conditions (i.e., concentration, temperature) to characterize the possible changes in the peptides' aggregation states as a putative critical factor for their antimicrobial activity. Finally, molecular dynamics simulations on a variety of conformations showed the intrinsic flexibility of these peptides in both aqueous solutions and membrane-mimetic systems.     

Detection of anti-Histoplasma capsulatum antibodies by the ELISA technique. Preliminary study

An indirect micro-ELISA system is presented for diagnosing histoplasmosis. The diagnostic criteria are defined by using sera from 12 patients who are histoplasmosis carriers. For this group, the optical density values were superior to 1,000; use was made of 43 sera from blood bank donors and 9 sera from children without a history of exposure. The optical density values in these cases were inferior to 0,200. The significant difference found led to the diagnostic criterion for confirming 3 histoplasmosis carriers who showed clinical manifestations but had been negative to double immunodiffusion. Thus, the usefulness of the proposed micro-ELISA system for early diagnosis was proved.     

Effectiveness of nitrogen incorporation to enhance the photoelectrochemical activity of nanostructured TiO2:NH3 versus H2-N2 annealing

Highly ordered TiO(2) nanohole layers were synthesized by anodic oxidation of titanium foils using ethylene glycol and ammonium fluoride as the electrolyte. The effectiveness of different methods, namely annealing at 500?°C in NH(3) and in H(2) diluted in N(2), to incorporate nitrogen into TiO(2) and thus extend its photoelectrochemical (PEC) activity to the visible range was studied. The intra-gap levels introduced by both processes were identified by means of XPS and PL measurements. Water splitting experiments demonstrated that annealing in H(2) improved the photocatalytic activity of pure TiO(2), while annealing in ammonia led to a decrease in the PEC performance.     

Pathogenicity of Vibrio alginolyticus for cultured gilt-head sea bream (Sparus aurata L.)

The in vivo and in vitro pathogenic activities of whole cells and extracellular products of Vibrio alginolyticus for cultured gilt-head sea bream were evaluated. The 50% lethal doses ranged from 5.4 x 10(4) to 1.0 x 10(6) CFU/g of body weight. The strains examined had the ability to adhere to skin, gill, and intestinal mucus of sea bream and to cultured cells of a chinook salmon embryo cell line. In addition, the in vitro ability of V. alginolyticus to adhere to mucus and skin cells of sea bream was demonstrated by scanning electron microscopy. The biological activities of extracellular products of V. alginolyticus were hydrolytic activities; the products were able to degrade sea bream mucus. V. alginolyticus was cytotoxic for fish cell lines and lethal for sea bream. Moreover, the extracellular products could degrade sea bream tissues. However, experiments performed with the bath immersion inoculation technique demonstrated that V. alginolyticus should be considered a pathogen for sea bream only when the mucus layer is removed and the skin is damaged.     

Urinary infection and pregnancy: a public health problem?

A clinical case of carditis associated with Mycoplasma pneumoniae infection in a 65 year-old woman is reported in order to stress some clinical features and therapeutic problems; during a 5-year follow-up. On the basis of this experience it is possible to state that in the pathogenesis an autoimmune mechanism probably plays an important role, whereas in therapy specific antibiotics are not effective and a long-term treatment with anti-inflammatory drugs is necessary.

Low-resolution structures of proteins in solution retrieved from X-ray scattering with a genetic algorithm

Small-angle x-ray solution scattering (SAXS) is analyzed with a new method to retrieve convergent model structures that fit the scattering profiles. An arbitrary hexagonal packing of several hundred beads containing the problem object is defined. Instead of attempting to compute the Debye formula for all of the possible mass distributions, a genetic algorithm is employed that efficiently searches the configurational space and evolves best-fit bead models. Models from different runs of the algorithm have similar or identical structures. The modeling resolution is increased by reducing the bead radius together with the search space in successive cycles of refinement. The method has been tested with protein SAXS (0.001 < S < 0.06 A(-1)) calculated from x-ray crystal structures, adding noise to the profiles. The models obtained closely approach the volumes and radii of gyration of the known structures, and faithfully reproduce the dimensions and shape of each of them. This includes finding the active site cavity of lysozyme, the bilobed structure of gamma-crystallin, two domains connected by a stalk in betab2-crystallin, and the horseshoe shape of pancreatic ribonuclease inhibitor. The low-resolution solution structure of lysozyme has been directly modeled from its experimental SAXS profile (0.003 < S < 0.03 A(-1)). The model describes lysozyme size and shape to the resolution of the measurement. The method may be applied to other proteins, to the analysis of domain movements, to the comparison of solution and crystal structures, as well as to large macromolecular assemblies.     

Human CD5 signaling and constitutive phosphorylation of C-terminal serine residues by casein kinase II

CD5 is a lymphocyte surface glycoprotein with a long cytoplasmic domain suitable for phosphorylation and signal transduction, which is involved in the modulation of Ag-specific receptor-mediated activation and differentiation signals. In this study, we use Jurkat T cell transfectants of CD5 cytoplasmic tail mutants to reveal phosphorylation sites relevant to signal transduction. Our results show that casein kinase II (CKII) is responsible for the constitutive phosphorylation of CD5 molecules at a cluster of three serine residues located at the extreme C terminus (S458, S459, and S461). Furthermore, the yeast two-hybrid system demonstrates the specific association between the C-terminal regions of the CD5 cytoplasmic tail and the regulatory beta subunit of CKII. We demonstrate that CKII associates with and phosphorylates the C-terminal region of CD5, a conserved domain known to be relevant for the generation of second lipid messengers, and thereby enables at least one component of its signaling function.
