The major obstacle in applying peptides to intracellular targets is their low inherent cell permeability. Standard approaches to attach a fluorophore (e. g. FITC, TAMRA) can change the physicochemical properties of the parent peptide and influence their ability to penetrate and localize in cells. We report a label-free strategy for evaluating the cell permeability of cyclic peptide leads. Fluorescent tryptophan analogues 4-cyanotryptophan (4CNW) and β-(1-azulenyl)-L-alanine (AzAla) were incorporated into in vitro translated macrocyclic peptides by initiator reprogramming. We then demonstrate these efficient blue fluorescent emitters are good tools for monitoring peptide penetration into cells. 相似文献

This paper studies a problem for seamless migration of legacy networks of Internet service providers to a software-defined networking (SDN)-based architecture along with the transition to the full adoption of the Internet protocol version 6 (IPv6) connectivity. Migration of currently running legacy IPv4 networks into such new approaches requires either upgrades or replacement of existing networking devices and technologies that are actively operating. The joint migration to SDN and IPv6 network is considered to be vital in terms of migration cost optimization, skilled human resource management, and other critical factors. In this work, we first present the approaches of SDN and IPv6 migration in service providers' networks. Then, we present the common concerns of IPv6 and SDN migration with joint transition strategies so that the cost associated with joint migration is minimized to lower than that of the individual migration. For the incremental adoption of software-defined IPv6 (SoDIP6) network with optimum migration cost, a greedy algorithm is proposed based on optimal path and the customer priority. Simulation and empirical analysis show that a unified transition planning to SoDIP6 network results in lower migration cost. 相似文献

Here we report a successful preparation of nanostructured calcium silicate by wet chemical approach. The synthesized sample was characterized by various physico-chemical methods. Thermal stability was investigated using thermo-gravimetric and differential thermal analysis (TG-DTA). Structural characterization of the sample was carried out by the X-ray diffraction technique (XRD) which confirmed its single phase hexagonal structure. Transmission electron microscopy (TEM) was used to study the nanostructure of the ceramics while homogeneous grain distribution was revealed by scanning electron microscopy studies (SEM). The elemental analysis data obtained from energy dispersive X-ray spectroscopy (EDAX) were in close agreement with the starting composition used for the synthesis. Superhydrophilic nature of CaSiO3 was investigated at room temperature by sessile drop technique. Effect of porous nanosized CaSiO3 on early adhesion and proliferation of human bone marrow mesenchymal stem cells (BMMSCs) and cord blood mesenchymal stem (CBMSCs) cells was measured in vitro. MTT cytotoxicity test and cell adhesion test showed that the material had good biocompatibility and promoted cell viability and cell proliferation. It has been stated that the cell viability and proliferation are significantly affected by time and concentration of CaSiO3. These findings indicate that the CaSiO3 ceramics has good biocompatibility and that it is promising as a biomaterial. 相似文献

The cold-spray technique is of significant interest to deposit refractory metals with relatively high melting point for a variety of demanding applications. In the present study, mechanical properties of cold-sprayed tantalum coatings heat treated at different temperatures were investigated using microtensile testing, scratch testing, and nanoindentation. The corrosion performance of heat-treated coatings was also evaluated in 1 M KOH solution, and potentiodynamic polarization as well as impedance spectroscopy studies were carried out. Assessment of structure–property correlations was attempted based on microstructure, porosity, and intersplat bonding state, together with mechanical and corrosion properties of the heat-treated cold-sprayed tantalum coatings. Coatings annealed at 1500 °C, which is very close to the recrystallization temperature of tantalum, were found to perform almost as bulk tantalum, with exciting implications for various applications. 相似文献

We report investigations on polycrystalline LaCo1?x Al x O3 (x = 0–0.9) bulk samples. The solid state synthesized samples showed a coexistence of rhombohedral and monoclinic phases in the intermediate concentrations (0.2 ≤ x ≤ 0.5) and pure rhombohedral phase otherwise. The observed effect of Al doping on dc transport has been analysed on the basis of small polaron hopping mechanism. The magnetisation results presented give evidence of weak ferromagnetism and anomalous temperature dependence of coercivity which we associate to the canting of the localised high-spin Co(III) and anti-symmetric exchange interactions at low temperatures. 相似文献
