Free-standing polyelectrolyte multilayer membranes have been formed by the layer-by-layer technique using a dip-coating apparatus. The polymer-chain mobility has been studied by 1H relaxation in the rotating frame T 1rho NMR with 13C chemical shift resolution. For each of the individual polymers a single relaxation component has been observed for all resolved signals. In the multilayer a significantly different relaxation time T 1rho has been observed with a minor second component. The interaction between the oppositely charged polyelectrolytes influences the molecular mobility.  相似文献

Assessing changes in the isotopic signature of contaminants is a promising new tool to monitor microbial degradation processes. In this study, chloroethene degradation was proven by depletion of chloroethenes, formation of chloride, increase in protein content and stable carbon isotope fractionation. Aerobic degradation of vinyl chloride (VC) was found to proceed metabolically, with degradation rates of 0.48 and 0.29 d(-1); and growth yields of 9.7 and 6.4 g of protein/mol of VC at room and groundwater temperature, respectively. Cis-1,2-dichloroethene (cDCE) was degraded cometabolically under aerobic conditions when VC was provided as growth substrate. Aerobic degradation was associated with significant stable carbon isotope fractionation, with enrichment factors ranging from -5.4+/-0.4 per thousand for metabolic degradation of VC to -9.8+/-1.7 per thousand for cometabolic degradation of cDCE. Thus, it was demonstrated that stable carbon isotope fractionation is suitable for assessing aerobic chloroethene degradation, which can contribute significantly to site remediation.  相似文献

In the paraxial Debye regime, simple and power-efficient pupil filters are designed to break the diffraction limit along a large depth of focus (DOF) for the Gaussian beam. Dependences of the superresolution factor, DOF gain, Strehl ratio, sidelobe strength, and axial intensity nonuniformity on the Gaussian profile in the pupil plane are characterized using the numerical method. Optimal filter designs are proposed for either high-resolution or ultra-large-DOF applications followed by experimental verifications.  相似文献

Targeted drug delivery systems that combine imaging and therapeutic modalities in a single macromolecular construct may offer advantages in the development and application of nanomedicines. To incorporate the unique optical properties of luminescent quantum dots (QDs) into immunoliposomes for cancer diagnosis and treatment, we describe the synthesis, biophysical characterization, tumor cell-selective internalization, and anticancer drug delivery of QD-conjugated immunoliposome-based nanoparticles (QD-ILs). Pharmacokinetic properties and in vivo imaging capability of QD-ILs were also investigated. Freeze-fracture electron microscopy was used to visualize naked QDs, liposome controls, nontargeted QD-conjugated liposomes (QD-Ls), and QD-ILs. QD-ILs prepared by insertion of anti-HER2 scFv exhibited efficient receptor-mediated endocytosis in HER2-overexpressing SK-BR-3 and MCF-7/HER2 cells but not in control MCF-7 cells as analyzed by flow cytometry and confocal microscopy. In contrast, nontargeted QD-Ls showed minimal binding and uptake in these cells. Doxorubicin-loaded QD-ILs showed efficient anticancer activity, while no cytotoxicity was observed for QD-ILs without chemotherapeutic payload. In athymic mice, QD-ILs significantly prolonged circulation of QDs, exhibiting a plasma terminal half-life ( t 1/2) of approximately 2.9 h as compared to free QDs with t 1/2 < 10 min. In MCF-7/HER2 xenograft models, localization of QD-ILs at tumor sites was confirmed by in vivo fluorescence imaging.  相似文献

The effects of the dopamine receptor antagonist flupenthixol were examined on the development of conditioned partner preference induced by paced copulation in female rats. In Experiment 1, ovariectomized, hormone-primed rats were conditioned to associate scented and unscented males with paced and nonpaced copulation, respectively. Females in Experiment 2 associated albino or pigmented males with paced or nonpaced copulation. Flupenthixol or saline was administered before each conditioning trial. During a final drug-free preference test, females could choose to copulate with either a pacing-related or nonpacing-related male. Saline-trained females copulated preferentially with the pacing-related male, whereas flupenthixol disrupted odor but not strain conditioning. We investigate the greenfield deployment of hybrid time division multiplexing/wavelength division multiplexing passive optical networks, i.e., the optimized covering of a given geographical area by a set of cost-effective and properly dimensioned hybrid PONs. We first propose a three-phase optimization scheme. In the first phase, a p-center model is proposed to determine the best ONU clusterings and the lower-level passive distribution node equipment locations. The second phase takes care of the placement of the upper-level passive distribution node equipment and the third one, using the traffic demands, a mix of unicast and multicast requests, selects the most appropriate passive distribution node equipment, i.e., either an arrayed waveguide gratings or a splitter. We next merge the first two phases, leading then to a two-phase scheme. Computational experiments are conducted on two sets of ONUs, one with 128 and one with 512 ONUs. We compare the two- and three-phase schemes in terms of PON design costs. We observe that the two-phase scheme provides more cost-effective solutions. We also provide a sensitivity analysis with respect to several parameters in order to identify the number of ONU clusters that minimizes the deployment cost.  相似文献

The chemical composition of plant surfaces plays a role in selection of host plants by herbivorous insects. Once the insect reaches the plant, these cues determine host acceptance. Laboratory studies have shown that the stem borer Busseola fusca (Lepidoptera: Noctuidae), an important pest of sorghum and maize in sub-Saharan Africa, is able to differentiate between host and non-host plant species. However, no information is available on the cues used by this insect to seek and accept the host plant. Thus, the role of surface phytochemical stimuli on host selection and oviposition by B. fusca was studied in the laboratory using two host plants, sorghum, Sorghum bicolor, and maize, Zea mays, and one non-host plant, Napier grass, Pennisetum purpureum. The numbers of eggs and egg masses deposited on the three plant species were compared first under no-choice and choice conditions. In both cases, more eggs and egg masses were laid on maize and sorghum than on the non-host. Artificial surrogate stems treated with a water or chloroform surface extract of each plant were then compared with surrogate stems treated with, respectively, water or chloroform as controls, under similar conditions. Surrogate stems treated with plant water extracts did not show an increase in oviposition when compared to controls, indicating that the major compounds in these extracts, i.e., simple sugars and free amino acids, are not significantly responsible for the oviposition preference. By contrast, a chloroform extract of sorghum enhanced oviposition on the surrogate stems compared to the control, while those of maize and Napier grass showed no significant effects. Analysis of the chloroform extract of sorghum showed higher amounts of α-amyrin, ß-amyrin, and n-nonacosane compared to those of maize and Napier grass. A blend of the three chemicals significantly increased oviposition compared to the chloroform-treated control, indicating that these compounds are part of the surface chemical signature of the plant responsible for host recognition and oviposition by B. fusca.  相似文献

One hundred isolates of the commercial Saccharomyces cerevisiae strain Zymaflore VL1 were recovered from spontaneous fermentations carried out with grapes collected from vineyards located close to wineries in the Vinho Verde wine region of Portugal. Isolates were differentiated based on their mitochondrial DNA restriction patterns and the evaluation of genetic polymorphisms was carried out by microsatellite analysis, interdelta sequence typing and pulsed-field gel electrophoresis (PFGE). Genetic patterns were compared to those obtained for 30 isolates of the original commercialized Zymaflore VL1 strain. Among the 100 recovered isolates we found a high percentage of chromosomal size variations, most evident for the smaller chromosomes III and VI. Complete loss of heterozygosity was observed for two isolates that had also lost chromosomal heteromorphism; their growth and fermentative capacity in a synthetic must medium was also affected. A considerably higher number of variant patterns for interdelta sequence amplifications was obtained for grape-derived strains compared to the original VL1 isolates. Our data show that the long-term presence of strain VL1 in natural grapevine environments induced genetic changes that can be detected using different fingerprinting methods. The observed genetic changes may reflect adaptive mechanisms to changed environmental conditions that yeast cells encounter during their existence in nature.  相似文献

A new hybrid method for the analysis of diffractive optical elements, which combines fully vectorial and scalar theories, is presented. It is suitable for use with elements of arbitrary large zone, even when the local feature size is of the order of the wavelength. To assess its applicability, we have performed cross-checking tests. The model is shown to accurately predict many optical properties of diffractive optical elements based on two-dimensional artificial dielectrics, like the useful energy diffracted into the order of interest or the deterministic loss into high diffraction orders for an illumination with a wavelength different from the design wavelength or for highly oblique incidence.  相似文献
