This paper reports on the design and successful field testing of a photovoltaic (PV)-powered desalination system. The system described here is intended for use in remote areas of the Australian outback, where fresh water is extremely limited and it is often necessary to drink high salinity bore water. A hybrid membrane configuration is implemented, whereby an ultrafiltration (UF) module is used for removing particulates, bacteria and viruses, while a reverse osmosis (RO) or nanofiltration (NF) membrane retains the salts. The concepts of water and energy recovery are implemented in the design. Field trials, performed in White Cliffs (New South Wales), demonstrated that clean drinking water was able to be produced from a variety of feed waters, including high salinity (3500 mg/l) bore water and high turbidity (200 NTU) dam water. The specific energy consumption ranged from 2 to 8 kW h/m3 of disinfected and desalinated drinking water, depending on the salinity of the feed water and the system operating conditions. The optimum operating pressure when filtering bore water was determined to be in the range 6–7 bar.  相似文献

The Java? 2 Platform, Enterprise Edition (J2EE?), is established as the standard platform for hosting enterprise applications written in the Java programming language. Similar to an operating system, a J2EE server can host multiple applications, but this is problematic due to limitations on scalability, weak inter‐application isolation and inadequate resource management facilities in the underlying Java platform. These limitations lead to a proliferation of server instances with a consequent dramatic increase in the total memory footprint and more complex system administration. The Multi‐tasking Virtual Machine (MVM) solves this problem by providing an efficient and scalable implementation of the isolate API for multiple, isolated tasks, enabling the co‐location of multiple server instances in a single MVM process. Isolates also enable the restructuring of a J2EE server implementation as a collection of isolated components, offering increased flexibility and reliability. Sodium-glucose co-transporters (SGLTs) serve to reabsorb glucose in the kidney. Recently, these transporters, mainly SGLT2, have emerged as new therapeutic targets for patients with diabetes and kidney disease; by inhibiting glucose reabsorption, they promote glycosuria, weight loss, and improve glucose tolerance. They have also been linked to cardiac protection and mitigation of liver injury. However, to date, the mechanism(s) by which SGLT2 inhibition promotes systemic improvements is not fully appreciated. Using an obese TallyHo mouse model which recapitulates the human condition of diabetes and nonalcoholic fatty liver disease (NAFLD), we sought to determine how modulation of renal glucose handling impacts liver structure and function. Apart from an attenuation of hyperglycemia, Empagliflozin was found to decrease circulating triglycerides and lipid accumulation in the liver in male TallyHo mice. This correlated with lowered hepatic cholesterol esters. Using in vivo MRI analysis, we further determined that the reduction in hepatic steatosis in male TallyHo mice was associated with an increase in nuchal white fat indicative of “healthy adipose expansion”. Notably, this whitening of the adipose came at the expense of brown adipose tissue. Collectively, these data indicate that the modulation of renal glucose handling has systemic effects and may be useful as a treatment option for NAFLD and steatohepatitis.  相似文献

Gold nanoparticles (AuNP) can increase the efficacy of radiation therapy by sensitising tumor cells to radiation damage. When used in combination with radiation, AuNPs enhance the rate of cell killing; hence, they may be of great value in radiotherapy. This study assessed the effects of radiation and AuNPs on mitochondrial reactive oxygen species (ROS) generation in cancer cells as an adjunct therapeutic target in addition to the DNA of the cell. Mitochondria are considered one of the primary sources of cellular ROS. High levels of ROS can result in an intracellular state of oxidative stress, leading to permanent cell damage. In this study, human melanoma and prostate cancer cell lines, with and without AuNPs, were irradiated with 6-Megavolt X-rays at doses of 0–8 Gy. Indicators of mitochondrial stress were quantified using two techniques, and were found to be significantly increased by the inclusion of AuNPs in both cell lines. Radiobiological damage to mitochondria was quantified via increased ROS activity. The ROS production by mitochondria in cells was enhanced by the inclusion of AuNPs, peaking at ~4 Gy and then decreasing at higher doses. This increased mitochondrial stress may lead to more effectively kill of AuNP-treated cells, further enhancing the applicability of functionally-guided nanoparticles.  相似文献

Thermal unfolding methods are commonly used as a predictive technique by tracking the protein’s physical properties. Inherent protein thermal stability and unfolding profiles of biotherapeutics can help to screen or study potential drugs and to find stabilizing or destabilizing conditions. Differential scanning calorimetry (DSC) is a ‘Gold Standard’ for thermal stability assays (TSA), but there are also a multitude of other methodologies, such as differential scanning fluorimetry (DSF). The use of an external probe increases the assay throughput, making it more suitable for screening studies, but the current methodologies suffer from relatively low sensitivity. While DSF is an effective tool for screening, interpretation and comparison of the results is often complicated. To overcome these challenges, we compared three thermal stability probes in small GTPase stability studies: SYPRO Orange, 8-anilino-1-naphthalenesulfonic acid (ANS), and the Protein-Probe. We studied mainly KRAS, as a proof of principle to obtain biochemical knowledge through TSA profiles. We showed that the Protein-Probe can work at lower concentration than the other dyes, and its sensitivity enables effective studies with non-covalent and covalent drugs at the nanomolar level. Using examples, we describe the parameters, which must be taken into account when characterizing the effect of drug candidates, of both small molecules and Designed Ankyrin Repeat Proteins.  相似文献
