The temporal dynamics of microphytoplankton, microzooplankton and mesozooplankton were monitored over 37 months in the Adriatic Sea in order to identify alterations in the plankton structures, which can lead to, or enhance the production of macro-aggregates, that affected the entire northern basin in summers 2000 and 2002, and to assess any negative effects of mucilage on plankton temporal patterns. Samples were collected monthly, from June 1999 to July 2002, on three transects at 9 stations across the northern and central Adriatic Sea. Besides the high year-to-year variations in abundances and taxonomical composition, plankton communities only showed a clear seasonal succession during 2001, when since April a grazing food web developed and was able to control large sized phytoplankton increase. In spring-summer 2000 and 2002 consumer abundances remained quite low and the dominant mesozooplankton summer species (Penilia avirostris) did not reach its usual summer maximum. The lack of an efficient top control was more evident on the northernmost transect, where generally grazing food web prevails over the microbial one. A large part of the microphytoplankton blooms, although not particularly intense, was exported to the bottom in the particulate phase, where it was processed by bacteria, enhancing the production of refractory dissolved material.  相似文献

In this work we present the fabrication and characterization of immunosensors based on polystyrene (PS)-multiwalled carbon nanotube (MWCNT) composites. The electrochemical properties of the sensors have been investigated and show that the surface area is increased upon addition of the MWCNT-PS layer. Furthermore, a plasma activation process is used to partially remove the PS and expose the MWCNTs. This results in a huge increase in the electrochemical area and opens up the possibility of binding biomolecules to the MWCNT wall. The MWCNTs have been functionalized covalently with a model antibody (rabbit IgG). The biosensors have been tested using amperometric techniques and show detection limits comparable to standard techniques such as ELISA.  相似文献

The miscibility, the thermal degradation and biocompatibility of the blends of two biodegradable and bioreabsorbable polymers, brittle polyhydroxybutyric acid (PHB) and flexible poly (p-dioxanone) (PPD) are reported. The blends were prepared by casting from chloroform solutions and analyzed by differential scanning calorimetry, dynamic mechanical analysis, scanning electron microscopy and thermal gravimetric analysis. The blends are immiscible and present promising morphology of a dense phase and a microporous phase, one or the other being the matrix, depending on the composition. Despite the immiscibility, the thermal stability under an inert atmosphere is improved for both polymers. The results obtained from toxicity tests showed that PHB/PPD blends do not present indirect or direct cytotoxicity as a substrate for cellular growth.  相似文献

The construction and characteristics of the stable and well-structured alpha(4)W protein are described. The 117-residue, single-chain protein has a molecular weight of 13.1 kDa and is designed to fold into a four-helix bundle. Experimental characterization of the expressed and purified protein shows a 69.8 +/- 0.8% helical content over a 5.5-10.0 pH range. The protein is thermostable with a T(M) > 355 K and has a free energy of unfolding as measured by chemical denaturation of -4.7 kcal mol(-1) at 25 degrees C and neutral pH. One-dimensional (1D) proton and 2D (15)N-HSQC spectra show narrow, well-dispersed spectral lines consistent with a uniquely structured alpha-helical protein. Analytical ultracentrifugation and NMR data show that the protein is monomeric over a broad protein concentration range. The 324 nm emission maximum of the unique Trp-106 is consistent with a sequestered position of the aromatic residue. Additionally, differential pulse voltammetry characterization indicates an elevated peak potential for Trp-106 when the protein is folded (pH range 7.0-8.5) relative to partly unfolded (pH range 11.4-13.2). The oxidation of Trp-106 is coupled to proton release as shown by a 53 +/- 3 mV/pH unit dependence of the peak potential over the 7.0-8.5 pH range.  相似文献

Anticipated emission legislation and reduced fuel consumption are the main driving forces when developing new engines. Optimization of the active surfaces in the piston system is one possible way to meet the above demands. In this study the effects of surface topography and texture direction of the ring/liner contact on oil film thickness and friction were simulated and experimentally tested. “Low wear” results from the experimental wear tests with “glide honed” smooth liner surfaces supported the “low friction” simulation results. In addition a new wear volume sensitive surface roughness parameter, Rktot, based on the Abbot–Firestone bearing area curve was introduced.  相似文献

Point mutations in beta-glucocerebrosidase (GCase) can result in a deficiency of both GCase activity and protein in lysosomes thereby causing Gaucher Disease (GD). Enzyme inhibitors such as isofagomine, acting as pharmacological chaperones (PCs), increase these levels by binding and stabilizing the native form of the enzyme in the endoplasmic reticulum (ER), and allow increased lysosomal transport of the enzyme. A high-throughput screen of the 50,000-compound Maybridge library identified two, non-carbohydrate-based inhibitory molecules, a 2,4-diamino-5-substituted quinazoline (IC(50) 5 microM) and a 5-substituted pyridinyl-2-furamide (IC(50) 8 microM). They raised the levels of functional GCase 1.5-2.5-fold in N370S or F213I GD fibroblasts. Immunofluorescence confirmed that treated GD fibroblasts had decreased levels of GCase in their ER and increased levels in lysosomes. Changes in protein dynamics, monitored by hydrogen/deuterium-exchange mass spectrometry, identified a domain III active-site loop (residues 243-249) as being significantly stabilized upon binding of isofagomine or either of these two new compounds; this suggests a common mechanism for PC enhancement of intracellular transport.  相似文献

CuO supported on CeO2, Ce0.8Zr0.2O2 and Ce0.8Al0.2O2 based catalysts (6%wt Cu) were synthesized and tested in the preferential oxidation of CO in a H2-rich stream (CO-PROX). Nanocrystalline supports, CeO2 and solid solutions of modified CeO2 with zirconium and aluminum were prepared by a freeze-drying method. CuO was supported by incipient wetness impregnation and calcination at 400 °C. All catalysts exhibit high activity in the CO-PROX reaction and selectivity to CO2 at low reaction temperature, being the catalyst supported on CeO2 the more active and stable. The influence of the presence of CO2 and H2O was also studied.  相似文献

Origanum × majoricum Cambess. is the most commonly grown oregano in Argentina. The aim of this work was to explore and describe its polyphenolic profile by high-performance liquid chromatography with diode array detection (HPLC-DAD) to support identification by classical microscopy. Remarkable homogeneity was found when defining the HPLC-DAD fingerprint with 17 characteristic common peaks. Eleven spectroscopically identified compounds were located. The method was successfully applied for the characterization of 22 samples from different productive regions of Argentina. The genetic inheritance from both progenitors of this hybrid oregano was evidenced by the presence of compounds coming from Origanum majorana L. and Origanum vulgare L. This method, combined with statistical multivariate analysis, was also useful to recognize two groups within the samples. Although O. × majoricum Cambess. must be considered the only hybrid between two species, two origins considering the subspecies level of O. vulgare L. have been morphologically differentiated for the hybrid, both with O. majorana L. These formulas were differentiated by their HPLC-DAD fingerprints. The Argentine hybrids studied appeared consistently grouped under these two profiles.  相似文献

Zucchini is a non-climacteric fruit which is harvested immature on reaching about 18 cm in length. In the present paper we compare fruit quality parameters in different zucchini cultivars with the production of ethylene and the expression of two ethylene biosynthesis genes (CpACS1 and CpACO1) throughout postharvest storage at 4, 12 and 20 °C. In fruit stored at 12 or 20° ethylene production and expression of CpACS1 and CpACO1 genes remained very low throughout the whole storage period. Ethylene production was also low at 4 °C, but was induced rapidly upon transfer to 20 °C for a minimum of 4 h. The expression of both CpACS1 and CpACO1 genes was also induced by storage at 4 °C. This cold-induced ethylene is not necessary for triggering chilling injury (CI) symptoms in zucchini, since they were already evident in the cold chambers before rewarming of the fruit and, therefore, before the burst of ethylene. However, the level of the cold-induced ethylene was lower in those cultivars that were more tolerant to CI, and it fell in response to temperature conditioning treatments that alleviate CI symptoms.  相似文献
