Small amount of TiB2 (<5 wt%) was added into B4C through a novel method that combines the use of sputter deposition and hot pressing. Sputter deposition provided more uniform dispersion of TiB2 grains with smaller grain sizes as compared to the conventional particulate mixing. Small amount TiB2 addition demonstrated to be an effective way for improving the fracture behavior and toughness of B4C while not sacrificing its outstanding lightweight property to a large extent: 2.3 wt% TiB2 addition brought 15% improvement in indentation fracture toughness while resulting in less than 2% increase in density. The improvement can be attributed to the combination of crack impeding by TiB2 grains and crack deflection at the B4C–TiB2 interfaces. TiB2 also played as grain growth inhibitor resulting in a slight increase (2%) in Vickers hardness. Another intention of employing sputter deposition was to modify the grain boundary of B4C; however, neither formation of Ti-containing phase nor Ti segregation has been observed at grain boundaries likely due to the poor wettability of B4C. 相似文献

Anycast is a one‐to‐any bidirectional transmission scheme that can be used to provide various service‐oriented applications. However, traditional anycast cannot support some distributed applications that need to locate multiple servers such as network time protocols and threshold cryptography. Although there are some extended anycast schemes that can support the discovery of k services, they may cause high control overhead, low satisfaction ratio, or high searching latency when there are more service requesters and providers. The main reasons for these shortcomings are unknown service information and limited hierarchical structure. This work proposes an efficient anycast scheme, called AnyKast, for discovering k services in cluster‐based mobile ad hoc networks. In the AnyKast scheme, an anycast tree based on the clustering and virtual backbone is established to reduce unnecessary message transmission. In the anycast tree, anycast clusterheads disseminate their information to certain specific nodes that take the service information and perform the service selection task instead of blindly searching. Furthermore, to decrease the cost of service information collection and maintenance, scope flooding is used to limit the information transmission, and information piggybacking and periodical inquiry mechanism are used to increase the service information accuracy. The experimental results demonstrate that our scheme can effectively discover services, reduce request and reply message control overhead, and lower the searching latency. Obesity has become a pandemic that threatens the quality of life and discovering novel therapeutic agents that can reverse obesity and obesity-related metabolic disorders are necessary. Here, we aimed to identify new anti-obesity agents using a phenotype-based approach. We performed image-based high-content screening with a fluorogenic bioprobe (SF44), which visualizes cellular lipid droplets (LDs), to identify initial hit compounds. A structure-activity relationship study led us to yield a bioactive compound SB1501, which reduces cellular LDs in 3T3-L1 adipocytes without cytotoxicity. SB1501 induced the expression of gene products that regulate mitochondrial biogenesis and fatty acid oxidation in 3T3-L1 adipocytes. Daily treatment with SB1501 improved the metabolic states of db/db mice by reducing body fat mass, adipose tissue mass, food intake, and increasing glucose tolerance. The anti-obesity effect of SB1501 may result from perturbation of the PGC-1α–UCP1 regulatory axis in inguinal white adipose tissue and brown adipose tissue. These data suggest the therapeutic potential of SB1501 as an anti-obesity agent via modulating mitochondrial activities. 相似文献

Phorbol 12-myristate 13-acetate (PMA) is a potent tumor promoter and highly inflammatory in nature. Here, we investigated the toxic effects of PMA on different model system. PMA (10 μg) caused chromosomal aberrations on the Allium cepa root tip and induced mitotic dysfunction. Similarly, PMA caused embryonic and larval deformities and a plummeted survivability rate on zebrafish embryo in a dose-dependent manner. Persistently, PMA treatment on immortalized human keratinocyte human keratinocyte (HaCaT) cells caused massive inflammatory rush at 4 h and a drop in cell survivability at 24 h. Concomitantly, we replicated a cutaneous inflammation similar to human psoriasis induced by PMA. Herein, we used tangeretin (TAN), as an antagonist to counteract the inflammatory response. Results from an in vivo experiment indicated that TAN (10 and 30 mg/kg) significantly inhibited PMA stimulated epidermal hyperplasia and intra-epidermal neutrophilic abscesses. In addition, its treatment effectively neutralized PMA induced elevated reactive oxygen species (ROS) generation on in vitro and in vivo systems, promoting antioxidant response. The association of hypoxia-inducible factor 1-alpha (HIF-1α)-nuclear factor kappa-light-chain-enhancer of activated b cells (NF-κB) crosstalk triggered by PMA enhanced PKCα-ERK1/2-NF-κB pathway; its activation was also significantly counteracted after TAN treatment. Conclusively, we demonstrated TAN inhibited the nuclear translocation of HIF-1α and NF-κB p65. Collectively, TAN treatment ameliorated PMA incited malignant inflammatory response by remodeling the cutaneous microenvironment. 相似文献
