The authors tested 2 mechanisms for the relation of movie smoking exposure with onset of cigarette smoking in adolescence. Longitudinal data with 8-month follow-up were obtained from a representative sample of 6,522 U.S. adolescents, ages 10-14 years. Structural modeling analysis based on initial nonsmokers, which controlled for 10 covariates associated with movie exposure, showed that viewing more smoking in movies was related to increases in positive expectancies about smoking and increases in affiliation with smoking peers, and these variables were both related to smoking onset. A direct effect of movie exposure on smoking onset was also noted. Mediation findings were replicated across cross-sectional and longitudinal analyses. Tests for gender differences indicated that girls showed larger effects of movie exposure for some variables. Implications for policy and prevention research are discussed. The spectral signatures of nitro attack of the aromatic portion of thermoplastic urethanes (TPU) were determined. Eight fragment molecules were synthesized that represent the nitrated and pristine methylenediphenyl section common to many TPUs. Infrared (IR) and Raman (785 nm illumination) spectra were collected and modeled using the B3LYP/6-31G(d)//B3LYP/6-31G(d) model chemistry. Normal mode animations were used to fully assign the vibrational spectra of each fragment. The vibrational assignment was used to develop a diagnostic method for aromatic nitro attack in thermoplastic urethanes. The symmetric NO(2) stretch coupled out of phase with the C-NO(2) stretch (1330 cm(-1)) was found to be free from spectral interferences. Spectral reference regions that enable correction for physical differences between samples were determined. The carbonyl stretch at 1700 cm(-1) was the best IR reference region, yielding a limit of quantitation (LOQ) of 0.66 +/- 0.02 g N/100 g Estane. Secondary IR reference regions were the N-H stretch at 3330 cm(-1) or the urethane nitrogen deformation at 1065 cm(-1). The reference region in the Raman was a ring stretching mode at 1590 cm(-1), giving an LOQ of 0.69 +/- 0.02 g N/100 g Estane. Raman spectroscopy displayed a larger calibration sensitivity (slope = 0.110 +/- 0.004) than IR spectroscopy (slope = 0.043 +/- 0.001) for nitration determination due to the large nitro Raman cross-section. Resilin is an elastic protein found in specialized regions of the cuticle of insects, which displays unique resilience and fatigue lifetime properties. As is the case with many elastomeric proteins, including elastin, gliadin and spider silks, resilin contains distinct repetitive domains that appear to confer elastic properties to the protein. Recent work within our laboratory has demonstrated that cloning and expression of exon 1 of the Drosophila melanogaster CG15920 gene, encoding a putative resilin-like protein, results in a recombinant protein that can be photochemically crosslinked to form a highly resilient, elastic biomaterial (Rec1 resilin). The current study describes a recursive cloning strategy for generating synthetic genes encoding multiple copies of consensus polypeptides, based on the repetitive domains within resilin-like genes from D. melanogaster and Anopheles gambiae. A simple non-chromatographic purification method that can be applied to these synthetic proteins and Rec1 is also reported. These methods for the design and purification of resilin-like periodic polypeptides will facilitate the future investigation of structural and functional properties of resilin, and the development of novel highly resilient biomaterials.  相似文献

Environmentally-mediated drug resistance in B-cell precursor acute lymphoblastic leukemia (BCP-ALL) significantly contributes to relapse. Stromal cells in the bone marrow environment protect leukemia cells by secretion of chemokines as cues for BCP-ALL migration towards, and adhesion to, stroma. Stromal cells and BCP-ALL cells communicate through stromal galectin-3. Here, we investigated the significance of stromal galectin-3 to BCP-ALL cells. We used CRISPR/Cas9 genome editing to ablate galectin-3 in stromal cells and found that galectin-3 is dispensable for steady-state BCP-ALL proliferation and viability. However, efficient leukemia migration and adhesion to stromal cells are significantly dependent on stromal galectin-3. Importantly, the loss of stromal galectin-3 production sensitized BCP-ALL cells to conventional chemotherapy. We therefore tested novel carbohydrate-based small molecule compounds (Cpd14 and Cpd17) with high specificity for galectin-3. Consistent with results obtained using galectin-3-knockout stromal cells, treatment of stromal-BCP-ALL co-cultures inhibited BCP-ALL migration and adhesion. Moreover, these compounds induced anti-leukemic responses in BCP-ALL cells, including a dose-dependent reduction of viability and proliferation, the induction of apoptosis and, importantly, the inhibition of drug resistance. Collectively, these findings indicate galectin-3 regulates BCP-ALL cell responses to chemotherapy through the interactions between leukemia cells and the stroma, and show that a combination of galectin-3 inhibition with conventional drugs can sensitize the leukemia cells to chemotherapy.  相似文献

A series of eight thermoplastic polyurethane elastomers were synthesized from 4,4′-methylene diphenyl diisocyanate (MDI) and 1,4-butanediol (BDO) chain extender, with poly(hexamethylene oxide) (PHMO) macrodiol soft segments. The soft segment molecular weights employed ranged from 433 g/mol to 1180 g/mol. All materials contained 60% (w/w) of the soft segment macrodiol. Differential scanning calorimetry (DSC), dynamic mechanical thermal analysis (DMTA), wide angle x-ray diffraction (WAXD), and small angle x-ray scattering (SAXS) techniques were employed to characterize morphology. Tensile and Shore hardness tests were also performed. Materials were tested in the annealed state. It was found that an increase in segment length was accompanied by an increase in the degree of microphase separation, average interdomain spacing, hard domain order, hardness, stiffness, and opacity. DSC experiments showed the existence of several hard segment melting regions that were postulated to result from the disordering or melting of various hard segment length populations. For the system and composition ratio employed, it was found that optimum tensile properties (UTS and breaking strain) were achieved when a PHMO molecular weight of between 650 and 850 was utilized. © 1996 John Wiley & Sons, Inc.  相似文献

The electrochemical behavior of electrodes made by sealing carbon nanofibers in glass or with electrophoretic paint has been studied by scanning electrochemical microscopy (SECM). Because of their small electroactive surface area, conical geometry with a low aspect ratio and high overpotential for proton and oxygen reduction, carbon nanofiber (CNF) electrodes are promising candidates for producing electrode nanogaps, imaging with high spatial resolution and for the electrodeposition of single metal nanoparticles (e.g., Pt, Pd) for studies as electrocatalysts. By using the feedback mode of the SECM, a CNF tip can produce a gap that is smaller than 20 nm from a platinum disk. Similarly, the SECM used in a tip-collection substrate-generation mode, which subsequently shows a feedback interaction at short distances, makes it possible to detect a single CNF by another CNF and then to form a nanometer gap between the two electrodes. This approach was used to image vertically aligned CNF arrays. This method is useful in the detection in a homogeneous solution of short-lifetime intermediates, which can be electrochemically generated at one electrode and collected at the second at distances that are equivalent to a nanosecond time scale.  相似文献

We describe a simple, inexpensive coating method to produce thin silica and titania films with surfactant templated, orthogonally tilted cylindrical nanopore arrays. These films can be deposited onto any substrate because orientation of the 2D hexagonally close packed (HCP) mesophases out of the plane of the film is directed by a chemically neutral sacrificial copolymer layer. Orientation of the HCP mesophases through the entire thickness of films cured in open air is achieved by limiting the coating thickness. This generalizes the coating method by making it possible to deposit oriented films on substrates of any curvature and size. We find a critical thickness between 70 and 100 nm, below which the triblock copolymer surfactant-templated HCP phase aligns completely out of the plane of glass and silicon wafer substrates. Above this thickness, the effect of the chemically neutral bottom layer does not propagate across the entire film, and alignment of the HCP mesophases parallel to the (nonpolar) air interface produces a mixed orientation.  相似文献

Innovative TiO2/SnO2 nanofibers were fabricated via electrospinning an innovated precursor solution and used for photocatalytic H2 generation. The nanofibers exhibited greatly enhanced H2 evolution rate compared to bare TiO2 nanofiber and P25. The enhanced efficiency of the TiO2/SnO2 nanofibers was attributed to its excellent synergistic properties: (1) its good mesoporosity; (2) the red-shift of absorbance spectra to enhance light absorbance capability; (3) its long nanofibrous structure and (4) anatase TiO2 – rutile TiO2 – rutile SnO2 ternary junctions favorable for the separation of electrons and holes. Based on our experimental results, the optimum ratio of TiO2/SnO2 nanofibers with 3% Sn demonstrated the highest efficiency in H2 generation.  相似文献

A novel photocatalyst was designed from the inspiration of natural forest's high efficient on light harvesting and energy conversion. This novel “forest-like” photocatalyst was successfully synthesized by a facile continuously-conducted three steps methods: electrospinning TiO2 nanofiber acts as the trunks, hydrothermal growth ZnO nanorods on the surface of TiO2 nanofiber acts as the branches, while photodeposition of Cu nanoparticles on the surface of TiO2 nanofiber and ZnO nanorods act as the leaves. This novel photocatalyst demonstrated higher photocatalytic hydrogen generation rate than most of semiconductor catalysts and many newly developed catalysts such as Pt/TiO2 catalyst and artificial leaves Pt/N–TiO2 catalyst in a water/methanol sacrificial reagent system under the light irradiation as a result of its enhanced light absorption ability, enlarged specific surface area promoting mass transfer and providing more reaction sites and its potential on anti-recombination of electrons and holes. Meanwhile, it is interesting to note that the photocatalytic hydrogen generation activity has a liner relationship with the hierarchy of materials, which means higher hierarchy materials display higher photocatalytic hydrogen generation activity. It is reasonable to believe that this natural mimic photocatalyst without noble metals will benefit the energy generation and novel materials development.  相似文献

Conventional intensity imaging through turbid media suffers from rapid loss of image contrast due to light scattering from particles or random variations of refractive index. This paper features the development of an active imaging, snapshot, system design and postprocessing algorithms that differentiate between radiation that scatters or reflects from remote, obscured objects and the radiation from the scattering media itself through a combination of polarization difference imaging, channel blurring, and Fourier spatial filtering. The produced sensor acquires and processes image data in real time, yielding improved image contrasts by factors of 10 or greater for dense water vapor obscurants.  相似文献
