It has previously been shown that the Copenhagen (COP) rat contains several genetic loci that contribute to its mammary tumor-resistant phenotype after 7,12-dimethylbenz(a)anthracene (DMBA) administration. One of these loci, mammary carcinoma susceptibility 1 (Mcs1), is located on the centromeric end of chromosome 2 and appears to act in a semidominant fashion. To confirm the existence and independent action of this locus and also aid in the identification of the physical location of the Mcs1 gene, congenic lines were generated by transferring the Mcs1 COP allele onto a Wistar Furth (WF) genetic background. Male carriers were genotyped using microsatellite markers spanning 20-30 cM of the Mcs1 locus. One of the congenic lines minimally retained the COP allele at D2Mit29 on the centromeric end of chromosome 2 and extended distally to D2Rat201. Heterozygous Mcs1 carrier rats were interbred, and the female offspring were treated with DMBA. The female rats from the Mcs1 congenic line that carried one or two COP alleles of the Mcs1 region had a significantly reduced (65 and 85%, respectively) tumor development (P < 0.001) compared with rats carrying zero COP alleles at this locus. A WF.COP-D2Mit29/D2Rat201 homozygous congenic strain derived at the N10 generation was treated with DMBA, and the COP homozygous rats developed 1.5 +/- 0.3 carcinomas/rat versus 6.3 +/- 0.5 in WF control rats (P < 0.0001). Fine mapping of this congenic interval using several recombinant lines identified three genetic loci within the Mcs1 congenic region that independently supported a tumor resistance phenotype. These genetic loci have been termed Mcs1a, Mcs1b, and Mcs1c. In rats for which each locus was homozygous for the COP allele, tumor development was reduced by approximately 60% compared with littermate controls. The identification of these independent loci within the Mcs1 COP allele provide a model of the genetic complexity of cancer.  相似文献

The ability to temporarily maintain information in order to successfully perform a task is important in many daily activities. However, the ability to quickly and accurately update existing mental representations in distracting situations is also imperative in many of these same circumstances. In the current studies, individuals varying in working memory capacity (WMC) performed different varieties of go/no-go tasks that have been hypothesized to measure inhibitory ability. The results indicated that low-WMC individuals relative to high-WMC individuals showed worse performance specifically in certain conditions of the conditional go/no-go task. Further analyses showed that increasing the temporal lag/number of intervening items between the previous target and the current lure had a deleterious effect on the performance of the low-WMC group only. The results indicate a relationship between WMC and the ability to selectively update, maintain, and retrieve information, especially in interference-rich conditions. Aggressive reimbursement reform has been an imposing directive for care providers of ICU medicine. Timely knowledge of actual care routines obtained from a large sample of actively practicing physicians should be mandatory when developing any guidelines or practice standards. A questionnaire was therefore designed by the steering committee of the ACCP Council on Critical Care and sent to its members. The 1,294 responses were analyzed for demographics of the individual practitioner, local aspects of ICU staffing and policies, reimbursement, and a specific practice issue, nutrition. The typical respondent was aged 41 to 50 (41 percent), was a pulmonary subspecialist (68 percent), was not critical care certified (55 percent), worked 25 to 50 percent of his or her total time in the ICU (40 percent), and would continue ICU practice despite poor reimbursement (82 percent). Physicians practiced within a group (53 percent), in a 100- to 500-bed hospital (69 percent), with house staff available (60 percent), and predominantly cared for Medicare patients (55 percent). The following data may allow better judgments to be made pertaining to the implementation of care policies in the current ICU environment.  相似文献

Acute intestinal inflammation was established in rats by intraluminal administration of acetic acid into loops of distal ileum, proximal jejunum or ascending colon. The study included two control groups of intact (untreated) rats and sham-operated (saline-treated) rats for each intestinal segment. A third group of rats received acetic acid. Histological evaluation demonstrated that acetic acid treatment induced a mild inflammatory response. Two days after treatment, zinc absorption was measured using ligated 10-cm loops of each segment in which 65Zn was injected intraluminally. 65Zn absorption by the ileum, jejunum and colon was markedly reduced in those rats in which inflammation was induced by acetic acid. The liver showed the highest uptake of radioisotope, but the relative tissue distribution generally followed the amount of absorption. The surgical procedure itself seemed to reduce zinc absorption. No changes in [3H]leucine absorption were observed between sham-operated and acetic acid-treated controls. There was no significant serosal-->luminal secretion of intramuscularly injected 65Zn in any of the studied segments. Therefore, based upon the data obtained, we conclude that acetic acid-induced intestinal inflammation reduces absorption of zinc by the small and large intestine, and that a surgical procedure (laparotomy) also reduces zinc absorption. The mechanism of this inflammation is such that malabsorption shows some specificity.  相似文献

The first of two years of training in case study activities, adapted from the program originally developed by Fritz Redl, Daniel A. Prescott, and others at the Universities of Chicago and Maryland in the 1940s, is described. Excerpts from the case study of a 16-year-old female provide examples of activities, such as: (1) identifying recurring patterns of behavior, (2) selecting a pattern of behavior for analysis by the multiple hypotheses method, and (3) summarizing the case study record. The formation of nanostructures with controlled size and morphology has been the focus of intensive research in recent years. Such nanostructures are important in the development of nanoscale devices and in the exploitation of the properties of nanomaterials. Here we show how tree-like nanostructures ('nanotrees') can be formed in a highly controlled way. The process involves the self-assembled growth of semiconductor nanowires via the vapour-liquid-solid growth mode. This bottom-up method uses initial seeding by catalytic nanoparticles to form the trunk, followed by the sequential seeding of branching structures. Each level of branching is controlled in terms of branch length, diameter and number, as well as chemical composition. We show, by high-resolution transmission electron microscopy, that the branching mechanism gives continuous crystalline (monolithic) structures throughout the extended and complex tree-like structures. The controlled seeding method that we report here has potential as a generic means of forming complex branching structures, and may also offer opportunities for applications, such as the mimicking of photosynthesis in nanotrees.  相似文献

A review and expansion of the fundamental processes of the vapor–liquid–solid (VLS) growth mechanism for nanowires is presented. Although the focus is on nanowires, most of the concepts may be applicable to whiskers, nanotubes, and other unidirectional growth. Important concepts in the VLS mechanism such as preferred deposition, supersaturation, and nucleation are examined. Nanowire growth is feasible using a wide range of apparatuses, material systems, and growth conditions. For nanowire growth the unidirectional growth rate must be much higher than growth rates of other surfaces and interfaces. It is concluded that a general, system independent mechanism should describe why nanowires grow faster than the surrounding surfaces. This mechanism is based on preferential nucleation at the interface between a mediating material called the collector and a crystalline solid. The growth conditions used mean the probability of nucleation is low on most of the surfaces and interfaces. Nucleation at the collector‐crystal interface is however different and of special significance is the edge of the collector‐crystal interface where all three phases meet. Differences in nucleation due to different crystallographic interfaces can occur even in two phase systems. We briefly describe how these differences in nucleation may account for nanowire growth without a collector. Identifying the mechanism of nanowire growth by naming the three phases involved began with the naming of the VLS mechanism. Unfortunately this trend does not emphasize the important concepts of the mechanism and is only relevant to one three phase system. We therefore suggest the generally applicable term preferential interface nucleation as a replacement for these different names focusing on a unifying mechanism in nanowire growth.  相似文献

Linear spray atomization and deposition is an attractive technique to produce near-net-shape deposits, such as aluminum sheet and strip. In the present study, phase Doppler interferometry (PDI) was used in a backscatter mode to characterize, in situ, the droplet size and velocity distributions during linear spray atomization and deposition of a 3003 aluminum alloy. The PDI measurements were obtained along axes corresponding to the direction parallel to the nozzle slit and to the direction perpendicular to the slit. The PDI results delineate the temporal and spatial distribution of the droplet size and velocity during the metal spray. Both point and “line” measurements were obtained and are reported. The line measurements resulted from the integration of measurement made along a line scan obtained in real time (i.e., not ensemble averaged). Postrun analysis of the droplet size distribution using laser diffraction and sieving techniques is also reported. The PDI point measurements revealed that droplet size and velocity distribution were relatively invariant with time. The line measurements of droplet velocity showed that the droplet axial velocity exhibits a bimodal behavior, which becomes more apparent with increasing atomizing gas pressure, a result of droplet recirculation inside the spray chamber. In addition, the peak in the droplet axial velocity distribution increased as atomizing gas pressure increased. The line characterization also showed that the droplet size distribution became more homogeneous with increasing gas pressure, and that the distribution characteristic diameters of droplets decreased consistently with increasing gas pressure. Postrun characterization of the droplet size distribution of the entire metal spray using diffraction and sieving methods indicated that the mass (volume) median diameter D 50 and the Sauter mean diameter (SMD) D 32 decreased with increasing gas pressure in a manner consistent with PDI results.  相似文献
