Solubilization and solid-state characterization of a poorly soluble 5-alpha reductase inhibitor

GI197111X is a 5-alpha reductase inhibitor for the treatment of androgenetic alopecia. Equilibrium solubilities of GI197111X were determined in multiple solvents or cosolvents. A polymorph screen was conducted using suspension equilibration and solution recrystallization methods. Single crystals were grown from pyridine/water and crystal structure was determined using a Bruker SMART diffractometer. Crystal structure data were imported into Cerius2 to provide visualization of the crystal structure and calculation of the simulated X-ray powder diffraction (XRPD) pattern. The solubility of GI19711IX was low at 25 degrees C in all vehicles suitable for animal and human dosing. The solubility of 6.4 mg/mL in Capmul MCM made it the only choice for a soft gel dosage form for phase I/II. Solution recrystallization and suspension equilibration of GI197111X have produced only one crystal form. Crystal structure data: orthorhombic P2(1) 2(1) 2(1); a= 10.8960(6) A, b=11.5683(6) A, c=20.9019(11) A; unit cell volume 2634.65(24) A3; Z=4; calculated density= 1.248 g/cc. The molecule has seven chiral centers, and single-crystal analysis eliminated all possible stereo-isomers except the expected conformation or its enantiomer. Hydrogen bonds occur from both carbonyl oxygens to an H-N group. Simulated vacuum-based crystal morphology (habit) calculated using the Bravais-Friedel-Donnay-Harker, Growth Morphology, and Hartman-Perdok modules in Cerius2 was a close match to the morphology observed by light microscopy.     

An extended Butterworth-Van Dyke model for quartz crystal microbalance applications in viscoelastic fluid media

An extended Butterworth-Van Dyke (EBVD) model with frequency-independent parameters for the characterization of a resonant compound formed by a quartz crystal in contact with a finite viscoelastic layer contacting a semi-infinite viscoelastic medium is extracted by analysis of the lumped element model. The formulation of the EBVD model is compared with the complete expression of the electrical admittance of the loaded quartz derived from the transmission line model (TLM). Relative deviations between them do not exceed 3% around 1% bandwidth near resonance. An extended Martin and Granstaff's model and an explicit expression for the frequency shift that supposes an extension of Kanazawa's model for viscoelastic media are obtained. An analysis of the errors associated with the extraction of shear parameters of the coating for different materials prove that, to obtain an error less than 5% in the shear parameters determination, the viscoelastic contribution, defined as the relative error in the thickness computed from the frequency shift by Sauerbrey equation, must be greater than a limit that depends on Q, which is defined as the ratio of the shear storage modulus (G') to shear loss modulus (G"). In the materials studied polymers in the transition range or in the rubbery state with Q=1 and 10, the viscoelastic contribution must be higher than 15% and 50%, respectively, for a 5% limit error in the shear parameters extraction. A criterion for a practical determination of the appropriate viscoelastic regimes is indicated     

Analysis of trans-resveratrol by laser desorption coupled with resonant ionization spectrometry. Application to trans-resveratrol content in vine leaves and grape skin.

A method has been developed for nonvolatile compound analysis in fruits, namely, trans-resveratrol in grapes and vine leaves. This has been accomplished by the combination of laser desorption with resonance-enhanced multiphoton ionization coupled to time-of-flight mass spectrometry detection. After the optimization of the experimental conditions and the finding of the resonant wave-length of the substance, the full validation of the technique (i.e., linearity, repeatability, reproducibility, accuracy, detection limit, and quantification limit) was carried out with satisfactory results. Essential features of the method are as follows: (1) an enhanced desorption yield due to the mixing of the analyte with metal powder and (2) a high resolution and sensitivity and a low detection limit due to laser resonant ionization and mass spectrometric detection. Application to the analysis of trans-resveratrol in vine leaves and grape skin demonstrated the capabilities of the analytical method reaching detection limits of only few ppb.     

Myriad: scalable VR via peer‐to‐peer connectivity,PC clustering,and transient inconsistency

Distributed scene graphs are important in virtual reality, both in collaborative virtual environments and in cluster rendering. Modern scalable visualization systems have high local throughput, but collaborative virtual environments (VEs) over a wide‐area network (WAN) share data at much lower rates. This complicates the use of one scene graph across the whole application. Myriad is an extension of the Syzygy VR toolkit in which individual scene graphs form a peer‐to‐peer network. Myriad connections filter scene graph updates and create flexible relationships between nodes of the scene graph. Myriad's sharing is fine‐grained: the properties of individual scene graph nodes to share are dynamically specified (in C++ or Python). Myriad permits transient inconsistency, relaxing resource requirements in collaborative VEs. A test application, WorldWideCrowd, demonstrates collaborative prototyping of a 300‐avatar crowd animation viewed on two PC‐cluster displays and edited on low‐powered laptops, desktops, and over a WAN. We have further used our framework to facilitate collaborative educational experiences and as a vehicle for undergraduates to experiment with shared virtual worlds. Water absorption and termite attack on a Kraft paper-based composite treated with recycled polystyrene and three commercial resins

Lignocellulosic–plastic composites are materials widely used in the construction industry; however, their mechanical performance and durability constitutes a serious issue due to the nature of the lignocellulosic component which makes it prone to biological degradation. This article gives account of the protective performance of a novel eco-friendly solvent based coating obtained from recycled expanded polystyrene dissolved with Limonene, and three different coating systems used generally for outside wood protection, applied on a Kraft paper for potential structural use. Three aspects were evaluated, namely coating interaction with the lignocellulosic substrate, water absorption and resistance to Nasutitermes corniger termite attack. Scanning electron microscopy revealed two types of interaction namely—adsorption and absorption—of the coatings systems in the fibrous structure. Coating absorption revealed better protection for water absorptiveness of the paper structure. After biotic exposure, sample weight loss and damaged structure were observed, highlighting the aggressiveness of this type of termites which attack any kind of material studied here, in order to reach their food source.     

Live cell luminescence imaging as a function of delivery mechanism

Mitochondria in live cells can be imaged with a ruthenium(II) complex that usually binds and images nuclear DNA. The cellular uptake mechanism of this probe was changed by using a biocompatible pH-sensitive polymersome vector. This change in delivery route, determines the final cellular location of the probe and thus modulates its imaging properties.     

Bifunctional trehalose anode incorporating two covalently linked enzymes acting in series

A bienzymatic electrode incorporating trehalase (Tre) and glucose oxidase (GOx) covalently bound to the surface of Pt through a functionalized thiol linker (Tre|GOx|Pt) has been designed and assembled and its catalytic properties examined by chemical and electrochemical methods in aqueous phosphate buffer solutions (PBS, pH 7.4). Exposure of Tre|GOx|Pt to PBS containing trehalose (Tr) and subsequent polarization at 0.6 V versus Ag/AgCl yielded after a few minutes well-defined steady-state currents ascribed to the oxidation of hydrogen peroxide generated by the GOx-mediated oxidation of glucose (Gl) produced by the Tre-mediated dissociation of Tr. Plots of the steady-state currents versus [Tr] over the range examined, i.e., 5-25 mM, were found to be linear. Implications of these results toward the development of an implantable biofuel cell as an autonomous energy conversion device for insects are discussed.
