A lectin gene homolog of Oryza sativa was successfully cloned and expressed in Escherichia coli. The deduced amino acid sequence of the protein product showed a significant similarity with known chitin‐binding lectins. Most of the recombinant lectin was found in an insoluble aggregated form as inclusion bodies and only a small part was in the culture medium in a soluble active form. Functional recombinant lectin was recovered from the inclusion bodies by solubilization with 8 M urea in Tris/HCl buffer, pH 7.0 and renaturation by 10‐fold dilution in the same buffer. The recombinant lectin with His‐tag was simply purified to homogeneity by the process of affinity chromatography and was obtained with a yield of 6–8 mg/L culture. The recombinant lectin was a homo‐dimer composed of 22 kDa. The hemagglutination activity of the recombinant lectin was optimal at pH 4.0–7.0 and it was very sensitive to inhibition by N‐acetylneuraminic acid and thyroglobulin.  相似文献

MnO-SiO2-Al2O3-MnS oxysulfide system has been investigated by experimental phase diagram and activity measurement coupled with thermodynamic modeling.Phase equilibria of the MnO-MnS,MnO-SiO2-MnS, MnO-Al2O3-MnS and MnO-SiO2-Al2O3-MnS systems under low oxygen partial pressure have been experimentally investigated for the temperature range of 1 185 to 1 500℃using equilibration and quenching techniques. Equilibrium phases were analyzed by scanning electron microscope,electron probe X-ray microanalysis(EPMA), and differential thermal analysis(DTA ).Phase diagrams were successfully constructed for the systems investigated.Two ternary compounds in the MnO-SiO2-MnS system were found.Activities of MnO and MnS in MnO-SiO2-Al2O3-MnS liquid oxysulfide solution from very low sulfur concentration to high sulfur concentration at solid MnS saturation were investigated employing gas/liquid/Pt - Mn alloy under controlled atmosphere at 1 500℃.As X(SiO2)/(X(MnO) + X(SiO2)) increases in liquid oxysulfide solution,activity coefficient of MnO decreases while that of MnS increases.As X(AlO1.5) increases,the activity coefficient of MnS increases while no remarkable change was observed for the activity coefficient of MnO.Quantitative analysis of the thermodynamic properties of the oxysulfide solution as well as phase diagram of the system was also earned out by employing the Modified Quasichemical Model in the quadruplet approximation.In view of inclusion utilization for free - cutting steel,it might have an advantage to decrease the Al2O3 content and increase the MnO/SiO2 ratio.  相似文献

Friction stir butt welding (FSW) between A5052-O aluminum alloy plates with a thickness of 2 mm was performed.The rotation speeds of the welding tool were 2000 and 3000 r/min,respectively.The traverse speed was ranged from 100 mm/min to 900 mm/min.The defect-free welds with the very smooth surface morphology were successfully obtained,except for at the welding condition of 3000 r/min and 100 mm/min.The onion ring structure was observed in the friction-stir-welded zone (SZ) at the condition of 2000 r/min and 100 mm/min.For all the welding conditions,the grain size of the SZ was smaller than that of the base metal,and was decreased with the decrease of the tool rotation speed and with the increase of the tool traverse speed.The stir zone exhibited higher average hardness than the base metal.The decrease of the tool rotation speed and the increase of the tool traverse speed resulted in the increase in the average hardness of the SZ.The tensile strength of the FSWed plates was similar to that of the base metal,except for at the welding condition of 3000 r/min and 100 mm/min.The total elongation of the FSWed plates was lower than that of the base metal.  相似文献

The phase behaviors of toluene/polycyclic aromatic hydrocarbon mixture systems were investigated with a continuous-flow type apparatus at 573.2,598.2.623.2 and 648.2K,while the pressure changed from 1 tp 5 MPa,The pseudo-binary phase behaviors were predicted with the Peng-Robinson equation of state with interaction parameters between toluene and pseudo-components considered.The phase diagrams of the system have been classified following the category of phase boundary diagram models.The extraction selectivity and efficiency of toluene as a solvent was discussed by comparing with that of hexane .The prediction model for selectivity was also suggested.  相似文献

Compatible phases in the system Li2O-Al2O3-TiO2 at various temperature levels were determined mainly by solid-state reactions for the portion of the ternary system bounded by Li2O Al2O2, Li2O.TiO2, Al2O, and TiO2. The existence of a ternary compound, Li2O.Al2O3.4TiO2, and nine joins was established. The ternary compound has a lower limit of stability at 1090°± 15°C. and dissociates and recombines rapidly at 1380°± 15°C.  相似文献

Cr-Al-Si-N coatings were deposited on SUS 304 substrate by a hybrid coating system. A Cr interlayer was introduced between Cr-Al-Si-N coating and SUS 304 substrate to improve the coating adherence. The effects of Cr interlayer on the microhardness, adhesion, and tribological behavior of Cr-Al-Si-N coatings were systematically investigated. The results indicate that the microhardness of the Cr-Al-Si-N coatings gradually deceases with increasing thickness of Cr interlayers. The adhesion between Cr-Al-Si-N and SUS 304 substrate is improved by addition of the Cr interlayers. A peak critical load of ~50 N is observed for the coating containing Cr interlayer of 60 nm as compared ~ 20 N for the coating without Cr interlayer. The thicker Cr interlayers result in reduced critical load values. Moreover, the wear resistance of the Cr-Al-Si-N coatings is greatly enhanced by introducing the Cr interlayer with thickness of 60 nm in spite of the decreased microhardness. The friction coefficient of the coating system is also moderately reduced.  相似文献

Laser transformation hardening(LTH) is one of the laser surface modification processes. The surface hardening of rod-shaped carbon steel (SM45C) was performed by lathe-based laser composite processor with Gaussian-beam optical head. The LTH characteristics by dominant processes, longitudinal and depth directional hardness distributions and behaviors of phase transformation in hardened zones were examined. Especially, two concepts of circumferential speed and theoretical overlap rate were applied. When laser power increased or circumferential speed decreased, the surface hardening depth gradually increases due to the increased heat input. Moreover, the longitudinal hardness distribution particularly shows periodicity of repetitive increase and decrease, which results from tempering effect by overlap. Finally, the feasibility of laser transformation hardening is verified by using the beam with Gaussian intensity distribution.  相似文献

Aluminum alloys are used as substitutes for fiber reinforced polymer(FRP) in ships and boats. However, ships constructed with 5000-series Al alloy suffer a little corrosion in the marine environment, when they run in high speed, and high flow rate, etc. Therefore, solution heat treatment was carried out to prevent corrosion. The optimal heat treatment involved heating specimens for 120 min at 420 °C and then cooling them in water. In addition, the optimal ageing condition involved ageing specimens for 240 min at 180 °C. The slow strain rate test(SSRT) in a seawater revealed that heat treatment under optimal conditions produced improved elongation, time-to-fracture and amount of dimples compared with the as-received specimen.  相似文献

以大豆7S和11S球蛋白为研究对象,采用纳米二氧化硅(SiO2)对其进行分子修饰,添加量分别为蛋白基料的0.5%、1.0%、1.5%,然后用1、3、5 mol/L尿素控制变性相结合的方法来提高7S与11S球蛋白在3种木材(水曲柳、樱桃木、松木)上的胶黏强度。结果表明,经纳米SiO2修饰后,大豆7S和11S球蛋白的胶黏强度明显增大,最佳添加量为1%;当浓度为1 mol/L的尿素与1%的纳米SiO2共同修饰7S和11S球蛋白后,其胶黏强度最大。同时采用差示扫描量热仪测定了大豆球蛋白修饰前后的焓变,并探讨了胶黏作用增强的可能机理。  相似文献

Selected mixes consisting of Bf [high-beef (29%) low-fat (2.96%)], bf [low-beef (20%) low-fat] and BF [high-beef high-fat (5%)] which incorporated raw beef, defatted soy flour, and corn starch were extruded in a single-screw extruder. The products had no flavor additives and trained sensory panelists detected hay-like, beany or grainy flavors. Bf extrudate was more expanded than BF extrudate and rated least hard by the sensory panel, whereas bf extrudate was least susceptible to lipid oxidation. Bf and bf extrudates showed microstruc-tures with large air cells. All three showed advantages of beef and soy flour in amino acid and fatty acid compositions, respectively, and were microbiologically safe during prolonged storage at 37°C.  相似文献
