We have previously identified and cloned a human gene, D40, that is preferentially expressed in testis among normal organs, while it is widely expressed in various human tumor cell lines and primary tumors derived from different organs. In this report, we have examined the expression and localization of this protein in human testis with an antibody specific to D40 protein. In Western analyses, the anti-D40 antibody recognized a major band with a molecular mass of 300 kDa and a minor band of 250 kDa. These bands were not observed in the testis lysates from patients with Sertoli-cell-only syndrome and with Kleinfelter syndrome, who lack germ cells of the testis, indicating that D40 protein is expressed in the germ cells of normal testis. Immunohistochemical studies have revealed that D40 protein is highly expressed in spermatocytes and in the pre-acrosome of round spermatids. In the acrosome, D40 protein expression is observed not inside but outside the acrosome membrane. This is consistent with the finding that the amino-acid sequence at the amino terminal of the D40 protein lacks a hydrophobic signal peptide that is required for proteins to translocate to the membrane. Expression of D40 protein is observed in the acrosome of ejaculated spermatozoa as well, although the level is low compared with that in the pre-acrosome of spermatids. These results suggest that D40 protein plays important roles in spermatogenesis, especially in the formation and maintenance of the acrosome.  相似文献

We have developed a novel pulse-coupled neural network (PCNN) for speech recognition. One of the advantages of the PCNN is in its biologically based neural dynamic structure using feedback connections. To recall the memorized pattern, a radial basis function (RBF) is incorporated into the proposed PCNN. Simulation results show that the PCNN with a RBF can be useful for phoneme recognition. A novel cross-correlation measurement system without mechanical action has been demonstrated in which the delay time between the reference and signal pulse can be electronically scanned by utilising the soliton self-frequency shift and the chromatic dispersion in the optical fibre. In this system, the reference pulse is the ideal femtosecond soliton pulse. The degree of fatigue in ferritic wrought steel during cyclic bending was determined by analysis of diffracted SH ultrasonic waves with the aid of multiple regression analysis. As the degree of fatigue increases, accompanied by residual stress, the incident waves curve to the interior region with positive stress field owing to the acoustoelastic effect. As a result, the propagation time of the launched waves lengthens, leading to a modulation of the received waveform. Multiple regression analysis for the waveform modulation produces a reliable estimation, with correlation coefficient of 0.948, for the degree of fatigue.  相似文献

Phase equilibria in the bcc Fe-Ge system have been investigated by means of electron probe microanalysis and transmission electron microscopy. Two kinds of ordered phases, B2 and DO3 types, have been detected, the B2 phase being formed from the disordered A2 phase by a 2nd order transition. The DO3 phase is also formed from the B2 phase by a 2nd or higher order transition at higher temperatures. Below about 820 °C, however, the transition becomes a 1st order, and a miscibility gap between these ordered phases occurs in the composition range from 13 to 16 at. pct Ge. It has been also confirmed that the solubility of Ge in the bec phase shows a retrogression due to the formation of DO3 phase. On the basis of these experimental results, the phase diagram for the Fe-Ge system has been partly revised.  相似文献

An enzyme-based glucose/O2 biofuel cell was constructed within a microfluidic channel to study the influence of electrode configuration and fluidic channel height on cell performance. The cell was composed of a bilirubin oxidase (BOD)-adsorbed O2 cathode and a glucose anode prepared by co-immobilization of glucose dehydrogenase (GDH), diaphorase (Dp) and VK3-pendant poly-l-lysine. The consumption of O2 at the upstream cathode protected the downstream anode from interfering O2 molecules, and consequently improved the cell performance (maximum cell current) ca. 10% for the present cell. The cell performance was also affected by the channel height. The output current and power of a 0.1 mm-height cell was significantly less than those of a 1 mm-height cell because of the depletion of O2, as determined by the shape of the EI curve at the cathode. On the other hand, the volume density of current and power was several times higher for the narrower cell.  相似文献

The solid-state synthesis of a tetrathiafulvalene–chloranil (TTF–CA) complex was demonstrated, and progress in the formation of the complex was monitored using a GaP Raman terahertz (THz) spectrometer. Clear transitions in the THz absorption spectra indicated that the mixture of TTF and CA became the black phase TTF–CA complex. The results of X-ray diffraction, Raman scattering, and FTIR spectra are presented as supporting evidence.  相似文献
