Characterization of EspC, a 110-kilodalton protein secreted by enteropathogenic Escherichia coli which is homologous to members of the immunoglobulin A protease-like family of secreted proteins

Enteropathogenic Escherichia coli (EPEC) secretes at least five proteins. Two of these proteins, EspA and EspB (previously called EaeB), activate signal transduction pathways in host epithelial cells. While the role of the other three proteins (39, 40, and 110 kDa) remains undetermined, secretion of all five proteins is under the control of perA, a known positive regulator of several EPEC virulence factors. On the basis of amino-terminal protein sequence data, we cloned and sequenced the gene which encodes the 110-kDa secreted protein and examined its possible role in EPEC signaling and interaction with epithelial cells. In accordance with the terminology used for espA and espB, we called this gene espC, for EPEC-secreted protein C. We found significant homology between the predicted EspC protein sequence and a family of immunoglobulin A (IgA) protease-like proteins which are widespread among pathogenic bacteria. Members of this protein family are found in avian pathogenic Escherichia coli (Tsh), Haemophilus influenzae (Hap), and Shigella flexneri (SepA). Although these proteins and EspC do not encode IgA protease activity, they have considerable homology with IgA protease from Neisseria gonorrhoeae and H. influenzae and appear to use a export system for secretion. We found that genes homologous to espC also exist in other pathogenic bacteria which cause attaching and effacing lesions, including Hafnia alvei biotype 19982, Citrobacter freundii biotype 4280, and rabbit diarrheagenic E. coli (RDEC-1). Although these strains secrete various proteins similar in molecular size to the proteins secreted by EPEC, we did not detect secretion of a 110-kDa protein by these strains. To examine the possible role of EspC in EPEC interactions with epithelial cells, we constructed a deletion mutant in espC by allelic exchange and characterized the mutant by standard tissue culture assays. We found that EspC is not necessary for mediating EPEC-induced signal transduction in HeLa epithelial cells and does not play a role in adherence or invasion of tissue culture cells.

On modeling the design of cellular manufacturing systems

A new optimization model is discussed for the design of cellular manufacturing systems. It is based on an integer programming formulation that updates some other models by eliminating redundant machine assignment and cost coefficients dependent on cell configuration. To reduce computational burdens, a simplified integer programming model and a decomposition algorithm are proposed. Several computer solutions were performed to evaluate the performance of the new model. The computational results are discussed.     

Minimizing the number of tool switches on a flexible machine

This article analyzes a tool switching problem arising in certain flexible manufacturing environments. A batch of jobs have to be successively processed on a single flexible machine. Each job requires a subset of tools, which have to be placed in the tool magazine of the machine before the job can be processed. The tool magazine has a limited capacity, and, in general, the number of tools needed to produce all the jobs exceeds this capacity. Hence, it is sometimes necessary to change tools between two jobs in a sequence. The problem is then to determine a job sequence and an associated sequence of loadings for the tool magazine, such that the total number of tool switches is minimized. This problem has been previously considered by several authors; it is here revisited, both from a theoretical and from a computational viewpoint. Basic results concerning the computational complexity of the problem are established. Several heuristics are proposed for its solution, and their performance is computationally assessed.

Genetic transformation of Vitreoscilla sp

Of all the methods customarily used to transform E. coli we found only electroporation to be effective for transformation of the Gram-negative bacterium Vitreoscilla, yielding 5.10(5) transformants/microgram of plasmid DNA. The conditions used were close to those described for E. coli E. coli plasmids are stably maintained in Vitreoscilla. This is the first report of exogenous DNA transfer in Vitreoscilla which opens the way for the application of recombinant-DNA techniques to study this unique group of organisms.     

Rates of reinfection with Echinococcus granulosus, Taenia hydatigena, taenia ovis and other cestodes in a rural dog population in Uruguay

Forced thinking is an incompletely understood and rarely described epileptic aura. We studied three patients with forced thinking from left frontal lesions, two neoplastic and one vascular. All three experienced repetitive, intrusive thoughts at the onset of seizures. Their forced thinking was associated with the desire to vocalize, orobuccal movements, and speech arrest. The episodes occurred with other ictal manifestations and responded to antiseizure therapy. These patients suggest that epileptic forced thinking is a heterogeneous phenomenon; forced thinking from left frontal lesions is a manifestation of expressive language and is distinct from experiential thoughts arising from temporal limbic foci.     

Catalysis, the turntable for a clean future

Upgrading the bottom of the barrel for clean fuel production, developing new engines with optimized catalytic posttreatments, implementing clean catalytic combustion technologies, improving the overall CO₂ balance via catalytic processes and, lastly, developing new water purification techniques represent major upheavals in catalysis linked to refining and environmental protection. This paper, thus, will focus on the determining contribution of catalysis to the most recent achievements in these domains. A technico-economic approach will follow, underlining their main incentives and drawbacks within the framework of future environmental constraints.     

The schnittaxiom and unions of fuzzy subgroups

A necessary and sufficient condition for the union of an arbitrary family of fuzzy subgroups of a group to be a fuzzy group has been proposed. The criterion which gives a cut of the unit interval is called the Schnittaxiom. It has been applied successfully to investigate fuzzy subgroupness of arbitrary unions of homomorphic images and preimages.
