In this work, lipolysis, proteolysis and viscosity of ultra‐high temperature (UHT) milk containing different somatic cell counts (SCC) were investigated. UHT milks were analysed on days 8, 30, 60, 90 and 120 of storage. Lipolysis as measured by free fatty acids increase, casein degradation and viscosity of UHT milk were not affected by SCC but increased during storage. A negative relationship was observed between SCC and casein as a percentage of true protein on the 120th day of storage, hence indicating that high SCC increases the proteolysis of UHT milk by the end of its shelf life.  相似文献

Inulin is a storage polysaccharide present in more than 30,000 vegetable products, including chicory roots, that are considered suitable for industrial application. The objective of this work was to evaluate the influence of temperature and the soluble solids concentration on the rheological behavior of a concentrated inulin solution obtained from a centrifugation process from chicory roots, after freezing at − 24C. For all the evaluated soluble solids concentrations, inulin solutions showed a rheological behavior of a highly pseudoplastic fluid, with high resistance to flow at low strain rates followed by a breakdown of the structure when the shear rate increased. The effect of temperature on the apparent viscosity of inulin solutions can be represented by the Arrhenius equation. The rheological behavior of inulin solutions can be represented by the Herschel–Bulkley, Casson, Cross and Power Law equations, where the consistency index increases as temperature rises and the soluble solids concentration as well.

PRACTICAL APPLICATIONS


The main objective of this work was to analyze the rheological behavior of a concentrated inulin solution obtained from chicory roots, to check the possibility of its application to obtain powder inulin.
The concentrated inulin solution was obtained by diffusion in hot water, followed by concentration by evaporation and phase separation conducted by lowering the temperature (−24C) and centrifugation at a velocity of 10,000 rpm for a time interval of 15 min. However, this solution is still going to be processed. Therefore, it is important to know its rheological behavior.
The influence of temperature and the soluble solids concentration on the rheological behavior of the concentrated inulin solution was also studied.  相似文献

Sugar is a determinant for the quality of mangoes, but information about its accumulation is scarce. Although starch can contribute to sugar production during ripening, not much is known about the enzymes involved. This work presents the changes in carbohydrate and enzymes during the development and ripening of Keitt mangoes. Starch disappearance was concomitant to a fivefold increase of sucrose, the most abundant sugar of the ripe fruits. The activities of α-amylase, β-amylase, phosphorylase and isoamylase were detected in the pulp, and while α-amylase increased parallel to the starch content, β-amylase presented a 20-fold increase during ripening. On the other hand, high phosphorylase activity was observed when fruits were still accumulating starch, and lowered during ripening. Isoamylase was detected during development and increased slightly during ripening, which would be in agreement to the expected role for isoamylases as acting on both subproduct of starch synthesis and degradation.

PRACTICAL APPLICATIONS


The present work reinforces our previous works that Keitt mangoes do not ripen when attached to the tree. This fact allowed us to study all the starch degradation after mango harvesting which does not occur with the other cultivars. Data obtained in this work reinforce the role of α-amylase, β-amylase and isoamylase rather than the starch phosphorylases on starch granule degradation in mangoes, and the subsequent soluble sugar accumulation.  相似文献

This research aimed to identify the drivers of acceptance and purchase intent of a probiotic (Bifidobacterium longum BL05) nonflavoured yoghurt supplemented with glucose oxidase, and to model the consumers’ acceptability using sensometrics and artificial neural networks (ANN). Consumers (n = 100) evaluated the degree of liking of yoghurt assays in respect of appearance, aroma, taste, texture and overall linking. Sensometric techniques – multiple linear regression (MLR), partial least squares regression (PLS), principal component regression (PCR) – and ANN were used to model the overall liking. Sensory drivers of global acceptance and purchase intent were also determined using logistic regression (LR). Hierarchical cluster analysis (HCA) identified three consumer segments that presented differences in all sensory attributes evaluated (P < 0.05). The ANN model showed the best performance to predict overall liking, followed by the MLR, PLS and PCR, indicating that taste and texture were the most significant attributes impacting the yoghurts overall liking. In accordance with the logistic models, overall acceptance and purchase intent could be predicted with 81.94 and 85.49% accuracy, respectively. The logistic regression indicated that taste was the attribute that contributed significantly (P < 0.0001) to higher scores for purchase intent and was considered the driver of acceptance.  相似文献

Glial cells are crucial for maintaining central nervous system (CNS) homeostasis. They actively participate in immune responses, as well as form functional barriers, such as blood-brain barrier (BBB), which restrict the entry of pathogens and inflammatory mediators into the CNS. In general, viral infections during the gestational period can alter the embryonic and fetal environment, and the related inflammatory response may affect neurodevelopment and lead to behavioral dysfunction during later stage of life, as highlighted by our group for Zika virus infection. Severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) induces a cytokine storm and, during pregnancy, may be related to a more severe form of the coronavirus disease-19 (COVID-19) and also to higher preterm birth rates. SARS-CoV-2 can also affect the CNS by inducing neurochemical remodeling in neural cells, which can compromise neuronal plasticity and synaptic function. However, the impact of SARS-CoV-2 infection during pregnancy on postnatal CNS, including brain development during childhood and adulthood, remains undetermined. Our group has recently highlighted the impact of COVID-19 on the expression of molecular markers associated with neuropsychiatric disorders, which are strongly related to the inflammatory response. Thus, based on these relationships, we discussed the impact of SARS-CoV-2 infection either during pregnancy or in critical periods of neurodevelopment as a risk factor for neurological consequences in the offspring later in life, focusing on the potential role of glial cells. Thus, it is important to consider future and long-term public health concerns associated with SARS-CoV-2 infection during pregnancy.  相似文献

A method for reflectance in situ hybridization (RISH) is presented. The importance of the method is demonstrated by results obtained on cytological and histological breast cancer specimens. Scattering reflectance signals from 1-nm colloidal-gold particles after RNA/RNA in situ hybridization, using digoxigenin-labelled riboprobes, were detected by confocal scanning laser microscopy. The mRNA expression of two ras-related genes, rho B and rho C, was analysed in human histological breast cancer specimens and in human breast cancer cell lines. Horizontal (x, y) and vertical (z) optical sections after three-dimensional imaging were used for visualization. A marked heterogeneity (between individual cells and between specimens) was noted for the expression of the rho B gene, both in cytological and in histological samples. On the other hand, rho C was always expressed and showed no heterogeneity. This method allows the identification of several cellular constituents in an heterogeneous tissue structure, as demonstrated by the simultaneous detection of rho B (or rho C) by reflectance and of DNA, cytokeratin and/or vimentin by fluorescence.  相似文献

SUMMARY-The development of food mixtures containing vegetable protein concentrates, minerals, and vitamins represents an important approach to the problem of protein-calorie malnutrition of preschool children in many parts of the world. In Brazil, soy flour is the most readily available good-quality protein for this purpose. Protein efficiency-ratio studies in rats demonstrated that soy flour with a PER value of 1.99 was available in large quantities. Accordingly, a formula containing 19.50% soy flour, 4.50% nonfat milk powder, 32.28% corn starch, 39.00% sucrose, and 4.72% vitamins and minerals was thoroughly tested as an infant food. The PER value of the protein in rats was 2.31. Amino acid assay demonstrated that the protein compared favorably with the essential amino acid pattern of the FAO/WHO reference protein (FAO/WHO, 196.5) except for methionine-cystine, The formula was well accepted and tolerated by children and adults. Average nitrogen absorption and retention of the formula as the sole food of malnourished children were 72.68 and 23.82%; values for milk protein were 80.17 and 30.13%. Average nitrogen absorption and retention were 63.4 and 22.0% when the formula provided one-half (15 g) of the daily protein for normal children consuming a typical diet of the region, comparing favorably with values of 69.3 and 21.3% for milk protein.  相似文献

The effects of pH, temperature, inhibitors, and substrates on proteolytic activities of crude extracts from the stomach, liver, pyloric caeca, and intestines of Colossoma macropomum were investigated. The highest acid and alkaline proteolytic activities were found in stomach and pyloric caeca, respectively. The optimum pH for the acid and alkaline proteases were 1.8 and 7.0–9.0, respectively, while the optimum temperatures were 35C and 65C. This alkaline protease thermal stability remained unaltered after 90 min incubation at 55C. A pepsin‐like protease was responsible for most of the acidic proteolytic activity (Pepstatin A inhibited approximately 90%), whereas PMSF inhibited about 40% of the alkaline protease. The alkaline proteolytic activity has attractive thermal properties for industrial applications.  相似文献

Crack closure delays the intrinsic mechanisms responsible for crack growth, therefore, it must be considered in fatigue crack growth modelling. The objective of this work is to develop a numerical procedure to predict crack closure induced by plasticity. First the crack closure was experimentally measured on M(T) 6082‐T6 aluminium alloy specimens of 3 mm thickness. A pin microgauge was used with the compliance technique. Then different parameters of the numerical procedure were analysed, namely the finite element mesh and the crack propagation scheme. The size of crack‐tip elements has an important influence and it is recommended to be of the same order of cyclic plastic zone. Crack‐opening levels only 10% lower than experimental results were obtained considering kinematic hardening and two load cycles in each increment.  相似文献
