The 87-membered polypeptide with the sequence of the gamma subunit of cGMP phosphodiesterase from bovine retina rods (PDE gamma) was synthesized by the solid phase method. Two synthetic approaches, which were based on the Boc/Bzl-strategy, were used; both syntheses were carried out in a continuous-flow reactor with swellographic monitoring. In the first approach, five Arg residues were coupled in the form of Boc-Arg(Z)2-OH and the final cleavage of the peptide from the support was effected by the mixture of CF3SO2SiMe3 and thionisole in trifluoroacetic acid. There resulted a heterogeneous, ornitine-rich, and absolutely inactive peptide material which was insoluble in aqueous alkali. In the second approach, Arg(Tos) and the HF low-high cleavage procedure were used, which resulted in a homogeneous polypeptide (according to HPLC and capillary electrophoresis) that manifested correct molecular mass under ion-spray mass spectrometry and the full functional activity characteristic of the native protein. The effect of zinc salts on the PDE gamma fluorescence in solutions and on its solubility was established. This demonstrated a significant PDE gamma affinity with Zn2+ ions and appeared to be connected with the functioning of the protein in the retina cells. For the first time, the dynamics of the peptidylpolymer swelling in different solvents was studied during the synthesis of peptides with very long sequences. 相似文献

The resistance of carbon and low-alloyed steels to local corrosion is shown to be directly determined by their phase-structure, rather than chemical, composition. A correlation between the presence of various nonmetal inclusions in steels and the local corrosion rate is revealed. Inclusions containing Ca and S are shown to be the most corrosive, manganese sulfide inclusions act less corrosively, and silicates are the least active. The formation of lamellar pearlite in the steel structure also impairs its resistance to local corrosion. By providing metal pure of corrosive nonmetallic inclusions at a melting stage and ensuring the pearlite spheroidizing during the stages of thermal treatment and processing, it is possible to enhance its resistance to total and local corrosion. 相似文献

This paper presents the results of a laser interference microscopy study of the morphology and dynamical properties of myelinated nerve fibres. We describe the principles of operation of the phase-modulated laser interference microscope and show how this novel technique allows us to obtain information non-invasively about the internal structure of different regions of a nerve fibre. We also analyse the temporal variations in the internal optical properties in order to detect the rhythmic activity in the nerve fibre at different time scales and to shed light on the underlying biological processes. We observe pronounced frequencies in the dynamics of the optical properties and suggest that the oscillatory modes have similar origin in different regions, but different strengths and mutual modulation properties. 相似文献
