The historical use of enzymes for food processing is briefly reviewed. The manufacturing process of microbial food enzymes is described including the fundamental purity criteria recommended by Joint FAO/WHO Expert Committee on Food Additives (JECFA) and Food Chemicals Codex. Principles for safety evaluation focus on the possible presence of toxic contaminants in the commercial enzyme preparation. Possible sources of any undesirable contaminants include the raw materials used, the properties of the production strain, contamination with foreign micro‐organisms, use of additives and processing aids in recovery of the enzyme. Systems for design of toxicology programmes are reviewed. A case study is given summarizing the safety evaluation of an immobilized lipase used for the interesterification of fats and oils. Based upon a no‐effect level derived from in vitro and animal feeding studies it is concluded that the enzyme is safe for the intended use.

The safety of the enzyme part of the immobilized preparation is documented by a thorough toxicology programme. The safety of the carrier, a phenol formaldehyde based ion‐exchange resin, is documented by leakage analysis of extractable materials under forced conditions, as well as by a feeding study employing a fat treated with the immobilized enzyme.  相似文献

In this study the combinatorial action of the main phenolic acids (ferulic, caffeic, p-coumaric and sinapic acids) found in extracts of free (ME) and bound phenolic acids (HE) from oat, barley and wheat flour on the modulation of NF-κB activity was investigated. The results show that combination of phenolic acids in low concentrations (<0.1 μg/ml) has a significant synergistic, or enhanced effect, on NF-κB activity, while their combination in high concentrations (0.3–70 μg/ml) helps to suppress the strong effect obtained by individual solutions of ferulic and p-coumaric acids. The modulation of NF-κB activity observed by HE extracts is the effect of combinatorial action of the phenolic acids present therein, while the modulation of NF-κB activity observed with ME extracts is the result of combinatorial action of phenolic acids together with other phenolic compounds present in the extracts. These results increase the knowledge about the health promoting effect from cereal consumption and dietary phenolic acids.  相似文献

This study investigated the efficacy of high voltage atmospheric cold plasma (HVACP) treatment on degradation of aflatoxin in corn. Gas type (Air, MA65), relative humidity (5, 40, 80% RH), treatment time (1, 2, 5, 10, 20,and 30 min), mode of reaction, post-treatment storage, stirring of corn material were parameters investigated on degradation of aflatoxin by HVACP treatment. Generation of reactive gas species was characterized with optical emission spectroscopy and measured with dragger tubes. Generation of reactive gas species are influence by gas type and relative humidity. Higher concentration of ozone and NOx were generated during HVACP treatment in MA65 than in air and with lower relative humidity. Aflatoxin in corn could be rapidly degraded by HVACP treatment. Aflatoxin in corn was degraded by 62% and 82% by 1 and 10 min HVACP treatment in RH 40% air, respectively. The degradation kinetics of aflatoxin by HVACP treatment follows a logistic model. Higher degradation of aflatoxin was achieved in gas MA65, at higher relative humidities (40%, 80%). Direct or indirect HVACP treatment was equally effective in degrading aflatoxin in corn. Stirring the corn sample during HVACP treatment and post-treatment storage increased aflatoxin degradation in corn by HVACP treatment.  相似文献

Dairy cows are typically gregarious, but isolate themselves in the hours before calving when kept on pasture. Self-isolation is also a common behavior of ill animals. The objectives of this study were to determine if dairy cows would (1) isolate to calve when housed indoors in an individual maternity pen and (2) continue to isolate when ill after calving. We selected individuals from a pool of 79 multiparous Holstein dairy cows based on inclusion criteria created to address each objective. Cows were moved from a group pen to 1 of 10 adjacent maternity pens. Half of these individual pens were partially covered with plywood, creating a secluded corner as well as a window that provided visual access to the group pen. The other individual pens were uncovered on all sides. For our first objective, we selected 39 cows that were moved into the maternity pens >8 h before calving (partially covered: n = 19; uncovered: n = 20). For our second objective, we selected 18 cows housed in the partially covered pens: 9 cows with high rectal temperature after calving and signs of an infectious disease (mastitis, metritis, pneumonia, or some combination), and 9 healthy cows paired with ill cows based on the amount of time they spent in the maternity pen before calving. Ten-minute scan sampling was used to record the location and lying time from 6 h before to 72 h after calving. Individual feed intake was measured after calving. Binomial tests were used to determine if cows in both pen types were more likely to calve in the corner or window side of the pen. Repeated-measures ANOVA were used to determine if cows used the corner more as calving approached and if ill cows spent more time lying or more time in the corner compared with healthy cows in the 72 h after calving. Cows in the uncovered pens were equally likely to calve on both sides of the pen (10 vs. 10), but 79% of cows in the partially covered pens calved on the corner side of the pen (15 vs. 4). Cows in the partially covered pens used the corner side of the pen more in the 1 h before and after calving compared with those housed in the uncovered pens. Ill cows housed in the partially covered pens ate less, tended to spend more time lying down, and spent more time in the corner of the pen compared with healthy cows. These results indicate that periparturient dairy cows seek seclusion to calve and when ill, which suggests that adding a secluded area to maternity and hospital pens may be beneficial.  相似文献

Although early studies suggested that coffee consumption might increase risk of some cancers, more comprehensive epidemiological and experimental data now generally indicate either neutral or beneficial effects. In this review, we summarize the current evidence for associations between breast, prostate, colorectal, and liver cancers and the consumption of coffee, and discuss the experimental evidence for potential chemopreventive mechanisms of coffee and coffee constituents. The epidemiological evidence consistently indicates that coffee protects against liver cancer, and also point toward protective effects for risk of colorectal cancers (with relative risks of 0.50 (95% CI: 0.42–0.59) and 0.83 (95% CI: 0.75–0.92), respectively, in the most recent meta‐analyses). There seems to be no association between the overall risk of breast and prostate cancer and coffee intake. However, for subgroups such as postmenopausal breast cancers, advanced prostate cancers, and breast and prostate cancer survivors, an inverse association with coffee intake is indicated. Potential mechanisms for chemopreventive effects of coffee phytochemicals includes inhibition of oxidative stress and oxidative damage, regulation of DNA repair, phase II enzymatic activity, apoptosis, inflammation, as well as having antiproliferative, antiangiogenetic effects and antimetastatic effects. The experimental evidence for effects of coffee and coffee constituents on each of these processes is discussed.  相似文献

The objective of the present study was to evaluate the influence of two diets containing either Brussels sprouts or inulin/rape seed cake, compared with a standard diet (control) for slaughter pigs on flavour and odour attributes and sensory profile of cooked pork. Three weeks prior to slaughter 24 female pigs were allocated to three diets: (1) a standard grower-finishing diet (control) for slaughter pigs containing barley, wheat and soy-bean meal, (2) the control diet containing 11 energy percent Brussels sprouts and (3) a diet containing 25% inulin and 55% rape seed cake. The odour and flavour of the cooked meat from inulin/rape seed cake-fed pigs differed significantly from the other two diets, showing reduced meat odour, increased pig and acrid odour, increased pig flavour, reduced fresh flavour and total impression. Meat from the Brussels sprouts-fed pigs deviated only slightly from the control-fed pigs.  相似文献

BACKGROUND: Noni (Morinda citrifolia L.) leaves have a documented history of food use. However, previous safety and antinutrient studies are absent. The current investigation was conducted to assess the utility of noni leaves as food. RESULTS: No evidence of toxicity or differences in weight gain were observed in acute, subacute, and subchronic oral toxicity tests of ethanol–water (1:1 v/v) and hot‐water extracts of noni leaves in mice at doses of 2000, 200, and 20 mg kg?1 body weight, respectively. Acute systemic anaphylaxis tests of the ethanol–water (4:1 v/v) and hot‐water extracts were negative. Further, leaf proteins were readily digested in simulated gastric fluid. Tannic acid concentrations in frozen and dried leaf were 1.6 and 25.8 g kg?1, respectively. Phytic acid was not detected in the raw leaf (<1 g kg?1). The average oxalic acid content was 1 g kg?1 and was fairly uniform among 22 leaf samples from 11 islands throughout French Polynesia. Similarly, campesterol, stigmasterol, and β‐sitosterol content did not vary widely, suggesting low inter‐island content variability. CONCLUSIONS: The apparent lack of toxicity of the leaves and the hardiness of the plant make it ideal for further agricultural development, especially where sustained growth of other food crops is difficult. BACKGROUND: During recent years, increases in liquid loss and soft texture in cold smoked salmon have been reported, possibly due to increased lipid content and a change in muscle structure. These problems can lead to downgrading of the product. This study aimed to investigate the influence of raw material composition and chill storage on quality parameters of cold smoked salmon. RESULTS: Initial smoked fish had a higher liquid‐holding capacity (LHC) than samples stored for 20 days. Large fish lost more liquid than small fish. The difference in LHC was consistent with a change in water distribution, which could indicate denaturation of muscle protein. Studies of the microstructure showed the influence of both processing and chill storage. An indication of lipid released from the cells was seen after 20 days of chill storage, which could be related to the reduced LHC at that time. CONCLUSION: Both raw material composition and chill storage affected the quality parameters of smoked salmon. This study has improved knowledge about the relationships between muscle structure, liquid‐holding properties and water distribution in smoked salmon. The effect of addition of rapeseed oil (canola), CuSO(4) and vitamin E (all-rac-α-tocopheryl acetate) to pig diets on pork meat quality (lipid oxidation, colour and drip loss) was studied. Pigs were reared on ten different diets, either a control diet (no supplementation of rapeseed oil, CuSO(4) or vitamin E) or 6% rapeseed oil diets supplemented with CuSO(4) (0, 35 or 175mg/kg) and vitamin E (0, 100 or 200mg all-rac-α-tocopheryl acetate/kg). The natural content of vitamin E originating from feed ingredients amounted to 9-23mg vitamin E (α-tocopherol) per kg feed. Muscle vitamin E levels reflected the dietary intake and pigs fed the control diet had significantly lower levels than pigs fed rapeseed oil diets. The quality of fresh pork chops packed in air or in 80% O(2):20% CO(2) was followed during chill storage for 8 and 13 days, respectively. Colour, as measured by tristimulus colorimetry of pork chops packed in 80% oxygen atmosphere, was significantly improved with respect to redness when compared to chops packed in air, regardless of dietary treatment. The low vitamin E content in pigs fed the control feed significantly decreased a values and the oxidative stability of pork chops during chill storage compared to the other feeding groups. Packing of chops in a high-oxygen atmosphere increased lipid oxidation, especially in chops with low levels of vitamin E. Supplementation of rapeseed oil diets with 100 or 200mg vitamin E significantly decreased lipid oxidation of chill stored chops. Supplementation with CuSO(4) did not influence meat quality attributes (drip loss, colour stability and lipid oxidation) for any of the storage conditions.  相似文献

Dmrt1 and amh are genes involved in vertebrate sex differentiation. In this study, we cloned dmrt1 and amh cDNAs in zebrafish (Danio rerio) and investigated the effects of exposure to 17a-ethinylestradiol (EE2), during early life on their patterns of expression and impact on the subsequent gonadal phenotype. Expression of both amh and dmrt1 in embryos was detected as early as at 1 day post fertilization (dpf) and enhanced expression of amh from 25 dpf was associated with the period of early gonadal differentiation. Sex-dependent differences in enhanced green fluorescent protein transgene expression driven by the promoter of the germ cell-specific vas gene were exploited to show that at 28dpf and 56dpf both amh and dmrt1 mRNA were overexpressed in males compared with females. Exposure during early life to environmentally relevant concentrations of EE2 had a suppressive effect on the expression of both amh and dmrt1 mRNAs and this was associated with a cessation/retardation in male gonadal sex development. Our findings indicate that estrogen-induced suppression in expression of dmrt1 and amh during early life correlate with subsequent disruptive effects on the sexual phenotype in males.  相似文献
