Continuous hydrolysis of olive oil byCandida cylindracea’s lipase was studied in a microporous hydrophobic membrane bioreactor. Olive oil and buffer solution, fed continuously through two compartments partitioned by membrane, caused reaction at the interface of lipase-adsorbed membrane and buffer solution. Fatty acid was obtained in a single phase without being mixed with components of other phases. At all mean residence times, countercurrent flow mode was superior to cocurrent one. The lipase was adsorbed onto the membrane, and its adsorption was suggested to be partially specific from the experiments with enzymes having various levels of purity. The percent hydrolysis depended hyperbolically on the interfacial enzyme concentration. The hydrolysis seemed to be limited by diffusion of fat or fatty acid through the micropores of the membrane at higher interfacial enzyme concentrations. The lipase was stabilized significantly by glycerol added to the buffer solution. Satisfactory performance of the membrane bioreactor was obtained in a longterm continuous operation which lasted for 24 days by feeding buffer-glycerol (18.0%) solution over the adsorbed lipase. The operational half-life of the adsorbed enzyme was 15 days at 40 C.  相似文献

The solid-solution compounds of Gd4(Al1 – x Ga x )2O9 (x = 0.0–1.0) were prepared at 1600°C for 5 h in air. The unit cell volume of the compounds increased from 0.853 to 0.878 nm3 with x. Phase transitions having a temperature hysteresis were observed from 1100° to 1400°C by calorimetry and dilatometry. The transition temperature increased with x. The volume of the high-temperature phase was 0.5% smaller than that of the low-temperature phase at the transition temperature. The volume changes were independent of x. The hysteresis width observed by the dilatometry was about 300°C for the Gd4Al2O9 ceramics (grain size: about 1 m) and decreased to 50°C for the Gd4(Al0.2Ga0.8)2O9 ceramics (grain size: over 10 m). Gd4Ga2O9 was unstable at low temperature and decomposed to Gd3GaO6 and Gd3Ga5O12 during the thermal analyses.  相似文献

Lipopeptides carrying a farnesyl thioether or a palmitic acid thioester and a farnesyl thioether were prepared from S-farnesyl cysteine methyl ester by N-terminal extension of the peptide chain employing the base labile Fmoc blocking group of the palladium(0) sensitive Aloc urethane. By means of this technique a lipohexapeptide representing the completely functionalized, i.e. palmitoylated and farnesylated C-terminus of the human N-Ras protein, was prepared. If acid labile blocking functions like the Boc group were used, upon deprotection an undesired addition of the acid to the double bonds of the farnesyl residue occurred. Therefore, acid labile blocking groups should not be employed in the synthesis of farnesylated lipopeptides. The lipopeptide methyl esters which carry only a farnesyl group do not inhibit protein farnesyl transferase, whereas palmitoylated peptides are weak inhibitors of this enzyme.  相似文献

This paper presents a new nonintrusive appliance load monitoring technique based on integer programming. Nonintrusive appliance load monitoring is the problem of identifying the operating conditions of electric appliances in a house by observing only the overall load current and voltage. Since the overall load current is expressed as a superposition of the currents of the operating appliances, the monitoring problem can be formulated as an integer quadratic programming problem by expressing the operating conditions as integer variables. This problem is solvable with a sufficiently small computational burden thanks to the recent development of commercial software. The proposed method does not require relearning even when a new appliance is installed in the house. Furthermore, the proposed formulation is applicable to cases in which some appliance has multiple modes, and cases in which some appliances of the same type are operating simultaneously. We have developed an innovative power generation scheduling method using quadratic programming (QP). The advantage of using our method is that it simultaneously solves unit commitment and economic load dispatch. We relax the binary variables of the unit state into continuous variables to apply QP to this problem. We also add a penalty term to converge the value of those variables to 0 or 1 to the objective function: the sum of the fuel costs and the start‐up costs. This penalty term depends on the per‐unit fuel cost. The possibility of its variable converging to zero increases as the cost increases. This method was applied to a test system of daily generation scheduling that consisted of 29 thermal units, two pumped‐storage units, four cascaded hydro units, and one transmission. The schedule satisfied all constraints, that is, load‐power balance, operation reserve, power flow, minimum up/down‐times, and fuel consumption. This paper describes the phenomenon of internal pressure‐rise due to arcs ignited between metal rod electrodes in air. First, the internal pressure‐rises in a closed chamber due to arcs were measured at currents within the range of 1 to 12.5 kA. The rod electrodes were made of copper, iron, and aluminum, and were 20 mm in diameter. The measurements showed that the maximum pressure‐rise distinctly differed with each electrode material at the same total arc energy, namely, the maximum pressure‐rise was the highest in the case of the aluminum electrode and was the lowest in the case of the iron electrode. Next, to quantitatively verify this difference, the internal pressure‐rise was estimated, taking into account the energy balance in the closed chamber on each electrode material. It was found that the estimated pressure‐rise approximately agreed with the above experimental results by taking into consideration oxidation reactions of the electrodes, melting and vaporization of the electrodes, and radiation loss of the arcs under certain realistic assumptions. © 2010 Wiley Periodicals, Inc. We examined the issue of reliability of InP-based high-electron mobility transistors (HEMTs), focusing on the increase of drain resistance Rd. In investigations of the mechanism of Rd increase, we took note of contaminant incorporation and of the relations between the device lifetime and the strength of the channel electric field. In the fabrication process, reducing contaminants, especially fluorine, significantly suppressed the increase of source and drain resistances. Cross-sectional views of the gate of improved devices, which had a long lifetime, confirmed an almost contaminant-free surface around the gate. In acceleration tests, the most negative impact on drain resistance stability among several bias conditions was found when the current density was high and the channel electric field was large at the same time. The dependence of drain-gate electric field strength E showed that the device lifetimes of HEMTs determined from Rd increase obeyed exp(1/E), which means that impact ionization was the main cause of degradation. We elucidate that the interactions of hot carriers with contaminants around the gate are the main causes of the Rd increase in HEMTs. Suppression of device degradation was achieved by optimizing the fabrication process around the gate. In this way, device lifetime was remarkably enhanced.  相似文献

This paper describes experimental and analytic studies on pressure rise and propagation phenomena due to high current arcs in a container with opened pipes. The shape of the experimental container was chosen taking into account underground common ducts. First, arcs were ignited in the container with varying diameter D and length L of the opened pipes and at arc currents of 4 kA to 12.5 kA and an arc duration of 0.1 s. Based on measured waveforms, the maximum pressure rise and the pressure oscillation frequency were obtained. It was found that there is a tendency for to increase with decreasing D and increasing L, and that D has a much greater impact on . In addition, the tendencies of with changing D and L can be approximately explained by Helmholtz resonance. Second, a CFD model for the container with opened pipe was developed based on the experimental results, and simulations were performed. The simulation results were found to correspond to the experimental pressure rise.  相似文献
