Objective: Innovation in material science has made it possible to fabricate a pharmaceutical material of modifiable characteristics and utility, in delivering therapeutics at a sustained/controlled rate. The objective of this study is to design and optimize the controlled release transdermal films of S-Amlodipine besylate by intercalating hydrophilic and hydrophobic polymers.
Methods: 3(2) factorial design and response surface methodology was utilized to prepare formulations by intercalating the varied concentration of polymers(A) and penetration enhancer(B) in solvent. The effect of these independent factors on drug release and flux was investigated to substantiate the ex-vivo, stability and histological findings of the study.
Results: FTIR, DSC revealed the compatibility of drug with polymers; however, the semicrystallinity in drug was observed under PXRD. SEM micrographs showed homogeneous dispersion and entanglement of drug throughout the matrix. Results from the permeation study suggested the significant effect of factors on the ex vivo permeation of drug. It was observed that drug release was found to be increased with an increase in hydrophilic polymer concentration and PE. The formulations having polymers (EC:PVPK-30) at 7:3 showed maximum drug release with highest flux (102.60?±?1.12?µg/cm2/h) and permeability coefficient (32.78?±?1.38?cm/h). Significant effect of PE on lipid and protein framework of the skin was also observed which is responsible for increased permeation. The optimized formulation was found to be stable and showed no-sign of localized reactions, indicating safety and compatibility with the skin.
Conclusion: Thus, results indicated that the prepared intercalated transdermal matrix can be a promising nonoral carrier to deliver effective amounts of drug. 相似文献
The present study concentrates on design, commissioning and calibration of a uniaxial laminar soil box suitable for use on a low base-shear capacity shake table available at IIT Kanpur, India. The box is designed to simulate the behavior of soil deposits subjected to earthquake motions, with minimal boundary effects due to reflection of waves at the boundary. The 1.1 m × 1.6 m × 0.765 m box is comprised of a series of individual lamina supported independently on multiple roller bearings guided through a guide channel. The outer frame connected to the guide rods is designed in such as way that it can transfer the self weight of each lamina out of the shake table. A series of free-field tests are carried out on dry Ganga sand sample to calibrate the box. Dynamic response parameters, such as acceleration, displacement, stress-strain behavior, strain-dependant modulus and damping ratio of the sand at various depth are investigated. Large strain and subsequent increased inelasticity is observed towards the top of the sand bed. The experimental results are further compared with equivalent-linear SHAKE analysis and nonlinear finite element ground response analysis of the free-field soil using OpenSees for assessing the performance of the laminar box. 相似文献
The aim of this study was to compare physico-chemical and biological properties of hydroxyapatite (HA) and hardystonite (HS) based composite scaffolds. Hardystonite (Ca2ZnSi2O7) powders were synthesized by a sol–gel method while polycaprolactone–hardystonite (PCL–HS) and polycaprolactone–hydroxyapatite (PCL–HA) were fabricated in nanofibrous form by electrospinning. The physico-chemical and biological properties such as tensile strength, cell proliferation, cell infiltration and alkaline phosphatase activity were determined on both kinds of scaffolds. We found that PCL–HS scaffolds had better mechanical strength compared to PCL–HA scaffolds. Addition of HA and HS particles to PCL did not show any inhibitory effect on blood biocompatibility of scaffolds when assessed by hemolysis assay. The in vitro cellular behavior was evaluated by growing murine adipose-tissue-derived stem cells (mE-ASCs) over the scaffolds. Enhanced cell proliferation and improved cellular infiltrations on PCL–HS scaffolds were observed when compared to HA containing scaffolds. PCL–HS scaffolds exhibited a significant increase in alkaline phosphatase (ALP) activity and better mineralization of the matrix in comparison to PCL–HA scaffolds. These results clearly demonstrate the stimulatory role of Zn and Si present in HS based composite scaffolds, suggesting their potential application for bone tissue engineering. 相似文献

The upcoming energy sources of Jatropha curcas and Ricinus communis promise to mitigate the energy crisis and environmental pollution belonging to the Euphorbiaceae family. Both of them have been researched in terms of availability, cost, and biochemical parameters. The seed oils of various jatropha and castor biotypes were screened out and evaluated for their physiochemical parameters viz. oil content (34–49%), biodiesel yield (30–81%), density (0.875–0.971 g/cm3), viscosity (0.6032–2.004 mm3/s), iodine value (75–450.45 mg/g), free fatty acid value (0.986–3.400 mg/g), saponification value (59.29–93.79 mg/g), flash point (133–218°C), fire point (163–262°C), fatty acid composition, and ash content (0.065–0.398%), and were estimated for comparison between jatropha and castor biotypes. Various combinations of auxins with cytokinins were used for a regeneration study. The best shoot regeneration (70%) was observed in MS medium supplemented with NAA (0.5 ppm) and BAP (2.5 ppm). Root induction (95%) was successfully obtained in plane MS. Acclimatization and hardening was quite successful with a survival rate of 70%. 相似文献
