Scheffersomyces stipitis PJH was mutagenized by random integrative mutagenesis and the integrants were screened for lacking the ability to grow with glutamate as sole carbon source. One of the two isolated mutants was damaged in the COX5 gene, which encodes a subunit of the cytochrome c oxidase. BLAST searches in the genome of Sc. stipitis revealed that only one singular COX5 gene exists in Sc. stipitis, in contrast to Saccharomyces cerevisiae, where two homologous genes are present. Mutant cells had lost the ability to grow with the amino acids glutamate, proline or aspartate and other non-fermentable carbon sources, such as acetic acid and ethanol, as sole carbon sources. Biomass formation of the mutant cells in medium containing glucose or xylose as carbon source was lower compared with the wild-type cells. However, yields and specific ethanol formation of the mutant were much higher, especially under conditions of higher aeration. The mutant cells lacked both cytochrome c oxidase activity and cyanide-sensitive respiration, whereas ADH and PDC activities were distinctly enhanced. SHAM-sensitive respiration was obviously essential for the fermentative metabolism, because SHAM completely abolished growth of the mutant cells with both glucose or xylose as carbon source.  相似文献

The recent EU Commission proposal for promoting the supply of power from renewable energy sources was originally based on a pan-European, harmonised tradable green certificate (TGC) scheme. We suggest, on the basis of a multi-disciplinary analysis, that a pan-EU TGC system is not the way forward for Europe. It is vital that the Commission (and the majority of Member States) avoids implementation of such policy designs put forward by a coalition of vested interests. They should instead look at, and act upon, the available evidence from those countries that have experimented with TGCs (e.g. Flanders, UK and Sweden) and design policies that stand a better chance of meeting the criteria of effectiveness, efficiency and equity. In particular, the policies must enable EU to meet the immense innovation/industrialisation challenge by inducing the development of a capital goods industry that can, eventually, diffuse a broad range of technologies that use renewable energy sources. Only then we can acquire an ability to implement an industrial revolution in the energy system in a way that broadly meets the criteria of effectiveness and dynamic efficiency.  相似文献

The expansion of renewable energy is a central element of the German Federal Government's climate and energy policy. The target for 2020 is to produce 30% of the electricity from renewable energies. Wind power has been selected to be a major contributor to this change. Replacing old wind turbines by modern ones and building new turbines on land will be crucial in meeting this target. However, the expansion of onshore wind power is not universally accepted. In several regions of Germany residents are protesting against setting up new wind turbines. To determine the negative effects two choice experiments were applied in Westsachsen and Nordhessen, Germany. In both regions the externalities of wind power generation until 2020 based on today's state of technology were measured. The results show that negative landscape externalities would result from expanding wind power generation. Using latent class models three different groups of respondents experiencing different degrees of externalities were identified.  相似文献

In groupware tools, the activation of a function may affect other users who might have conflicting interests. We developed technical mechanisms to support users in resolving them. Contrary to current implementations of groupware tools, these mechanisms strengthen the position of the users who are affected by the activation of said functions. Supporting the visibility of a function's activation, and providing a channel for communication or means to intervene against the function's activation are approaches which constitute a framework to implement these mechanisms. We conclude with showing implementation examples of the framework and their evaluation in the POLITeam project.  相似文献

4-Nonylphenols (NPs) are common products of biodegradation of a widely used group of nonionic surfactants, the nonylphenol ethoxylates (NPEs). These compounds are known to be persistent, toxic, and estrogen active. There is a worldwide scientific and public discussion on the potential consequences of human long term dietary exposure to such endocrine disrupters. Despite numerous determinations of NPs in environmental samples no systematical reports exist relating to concentrations of NPs in food. We analyzed NPs in 60 different foodstuff commercially available in Germany. The results indicate that NPs are ubiquitous in food. The concentrations of NPs on a fresh weight basis varied between 0.1 and 19.4 microg/kg regardless of the fat content of the foodstuff. Based on data on German food consumption rates and these first analyses of NPs in food, the daily intake for an adult was calculated to be 7.5 microg/day NPs. For infants exclusively fed with breast milk or infant formulas daily intakes of 0.2 microg/day and 1.4 microg/day NPs, respectively, can be estimated.  相似文献

A fast and convenient method for on-line monitoring of the extraction of heavy metals from solid (environmental) matrixes was developed. By the incorporation of microcartridges filled with dried and pulverized solid samples into the conduits of a flow system and appropriate selection of the liquid flowing through the cartridge, information about the degree of leaching and in particular of the kinetics of the leaching process are obtained. The method was optimized for determination of different metals of environmental concern using in-line detection by FAAS and ICPMS.  相似文献

The aim of this study was to assess the potential impacts of future climate change on the hydrological response in the upper reach of the Spree River catchment using the Soil and Water Assessment Tool (SWAT). The model was calibrated for ten years (1997–2006) and validated with the data from four years (2007–2010) using average monthly stream flow. The impact of future climate change on precipitation, temperature, evapotranspiration and stream flow was then investigated from two different downscaled climate models (CLM and WETTREG2010) under SRES A1B scenarios for two future periods (2021–2030 and 2041–2050). Besides that, sensitivity analysis was carried out with and without observations, to test robustness of the sensitivity algorithm used in the model. Results of the determination coefficient R2 and Nasch-Sutcliff efficiency ENC were 0.81 and 0.80, respectively, during the calibration; 0.71 and 0.70, respectively, during the validation. Although some parameters were changed their sensitiveness ranking when the model run with observations, the SWAT model was, however, able to predict the top influential parameters without observations. According to 12 realizations from the two downscaled climate models, annual stream flow from 2021–2030 (2041–2050) is predicted to decrease by 39 % (43 %). This corresponds to an increase in annual evapotranspiration from 2021–2030 (2041–2050) of 36 % (38 %). The upper reach of the Spree River catchment will likely experience a significant decrease in stream flow due to the increasing in the evapotranspiration rates. This study could be of use for providing insight into the availability of future stream flow, and to provide a planning tool for this area.  相似文献

Service-oriented architectures (SOA) have been successfully adapted by agile businesses to support dynamic outsourcing of business processes and the maintenance of business ecosystems. Still, businesses need to comply with applicable laws and regulations. Abstract service interfaces, distributed ownership and cross-domain operations introduce new challenges for the implementation of compliance controls and the assessment of their effectiveness. In this paper, we analyze the challenges for automated support of the enforcement and evaluation of IT security controls in a SOA. We introduce these challenges by means of an example control, and outline a methodology and a high-level architecture that supports the phases of the control lifecycle through dedicated components for observation, evaluation, decision support and reaction. The approach is model-based and features policy-driven controls. A monitoring infrastructure assesses observations in terms of key indicators and interprets them in business terms. Reaction is supported through components that implement both automated enforcement and the provision of feedback by a human user. The resulting architecture essentially is a decoupled security architecture for SOA with enhanced analysis capabilities and will be detailed and implemented in the MASTER project.  相似文献

SiC-B4C composites with various values of SiC-to-B4C ratio and grain size were fabricated by pressureless sintering. This paper presents the results of current investigations of this composite material. This includes the parameters of manufacture (shrinkage, density, and open porosity), thermoelectric properties (electrical and thermal conductivity, and thermopower), and material characterization (x-ray diffraction, scanning electron microscopy, oxidation resistance, and thermal expansion). The results indicate high potential of this composite as an alternative material for thermoelectric applications at high temperatures. The Seebeck coefficient of the composite was higher than that of the single-component materials B4C and SiC and reached 400 μV/K at 500°C.  相似文献
