In search of an efficient nonviral vector, polyethyleneimine (PEI)-based vectors were examined. In general, the transfection efficiency of nonviral vectors is suppressed by serum. Here we show that PEI based vectors, particularly, the chitosan hexamer-PEI vector, could perform efficient gene transfection into intestinal epithelial cells (IEC-6) in the presence of serum. The conjugation order of the two polymers with a plasmid (first, chitosan hexamer; second, PEI) was found to be an important factor in enhancing transfection efficiency. 相似文献

The science and an overview of the Soft X-ray Spectrometer onboard the STRO-H mission are presented. The SXS consists of X-ray focusing mirrors and a microcalorimeter array and is developed by international collaboration lead by JAXA and NASA with European participation. The detector is a 6×6 format microcalorimeter array operated at a cryogenic temperature of 50 mK and covers a 3′×3′ field of view of the X-ray telescope of 5.6 m focal length. We expect an energy resolution better than 7 eV (FWHM, requirement) with a goal of 4 eV. The effective area of the instrument will be 225 cm2 at 7 keV; by a factor of about two larger than that of the X-ray microcalorimeter on board Suzaku. One of the main scientific objectives of the SXS is to investigate turbulent and/or macroscopic motions of hot gas in clusters of galaxies. 相似文献

In this paper, a touch fault calculation method is proposed based on the symmetric coordinate method. The touch fault in this paper means that a line‐to‐line short circuit fault occurred on parallel transmission lines stringing on a same tower. It is known that the touch fault occurs very rarely such as the galloping phenomena, and it causes an abnormal voltage rise especially on a low‐voltage transmission line. However, a quantitative analysis method of the abnormal voltage has not been proposed. Therefore, a general analysis method of the voltage profile solution for the touch fault is proposed based on the symmetrical coordinate method. The proposed method is able to calculate the precise voltage and current profile solution efficiently by introducing the similar formation for the touch fault solution same as the general fault solution. Demonstrative result to the application in the touch fault calculation is presented in order to verify the practicability of the proposed method. 相似文献

The societal and economic forces driving change in medical education are affecting communities as well as universities. Each of the four authors of this paper is deeply involved in one of the components of their locale's well-developed community-based medical educational system, and each describes how change is influencing his role in that system, whether the role be managing a community hospital, directing a local Area Health Education Center, participating as a family medicine faculty member, or being a community preceptor. They agree on some common themes: (1) that it is good that medical students' education is moving into the community (e.g., this validates the importance of the community hospital to medical education, is an acknowledgment of the importance of generalism, and provides students invaluable learning experiences); (2) that educating medical students in the community is expensive, and more funding and resources are needed so that the area's hospitals, community faculty, preceptors, and support services can be fairly compensated for their commitment; and (3) that their community-based education system can no longer absorb the costs of training more medical students. This is not a criticism of academic medical centers, which are under tremendous financial pressures themselves, but is simply to state the community perspective and to urge fairness in the distribution of resources for medical education. Community institutions and academic medical centers will work individually to create their own integrated health care systems but must work together to create a better, more cost-effective system for educating medical students. 相似文献

Two endo-1,4-beta-glucanase genes, designated celA and celB, from a shoyu koji mold Aspergillus oryzae KBN616, were cloned and characterized. The celA gene comprised 877 bp with two introns. The CelA protein consisted of 239 amino acids and was assigned to the cellulase family H. The celB gene comprised 1248 bp with no introns. The CelB protein consisted of 416 amino acids and was assigned to the cellulase family C. Both genes were overexpressed under the promoter of the A. oryzae taka-amylase A gene for purification and enzymatic characterization of CelA and CelB. CelA had a molecular mass of 31 kDa, a pH optimum of 5.0 and temperature optimum of 55 degrees C, whereas CelB had a molecular mass of 53 kDa, a pH optimum of 4.0 and temperature optimum of 45 degrees C. 相似文献

The mouse ob gene and its human homologue OB have recently been cloned. The mutations in the ob gene are known to be associated with extreme obesity. The relationship between the human OB gene and disease, however, is largely unknown due to the lack of suitable markers within or adjacent to the OB gene. To obtain informative markers, we searched for simple tandem repeat polymorphisms in the genomic sequence of the human OB gene and identified a novel tetranucleotide repeat in the 3' flanking region. Fifteen alleles were detected in this marker with a heterozygosity of 0.85 and polymorphism information content of 0.83, indicating a highly informative nature of this marker. Two-point linkage mapping in two Centre Etude Polymorphisme Humaine (CEPH) reference families suggested that this marker is located in the interval between D7S514 and D7S530, the same interval where the OB gene is located (recombination fractions with D7S514 and D7S530 were 0.026 and 0.034, respectively). Although allele frequency distributions of this marker did not differ between 84 control subjects and 69 NIDDM patients, there was a tendency to higher body weight in control subjects with class I/class I genotype than in those without this genotype (68.8 +/- 11.1 vs 60.8 +/- 10.3 kg, p = 0.05). The highly polymorphic nature of this marker and its location in the OB gene makes this marker useful for linkage studies of the OB gene with a number of phenotypes, such as obesity, non-insulin-dependent diabetes mellitus, hypertension and the insulin resistance syndrome. 相似文献

In this study, gallium oxide (Ga2O3) nanoparticles were synthesized by sonochemical techniques at room temperature. We investigate the ultrasound irradiation conditions and the solvent type to synthesize Ga2O3 nanoparticles. γ-Ga2O3 nanoparticles were synthesized by irradiating gallium metal with ultrasound in hydrazine monohydrate solvent. The irradiation of hydrazine with ultasound suppresses the generation of ·OH, and GaOOH was not formed, and gallium metal directly oxidized. When the synthesized γ-Ga2O3 nanoparticles were heat-treated, a transition to β-Ga2O3 was observed. The heat-treated Ga2O3 nanoparticles showed excellent photocatalytic activity for the decomposition of RhB under UV irradiation. 相似文献

One of the most serious problems in medical science is an increment of Alzheimer's disease. It is known that the patient's brain atrophy is a result of neural cell loss. It is useful for the diagnosis of Alzheimer's disease to measure the volumes of the brain portions and to display them. We can obtain anatomical information from 2D slice images produced by MRI. We propose a computer-aided system for the diagnosis of Alzheimer's disease. The system consists of: (1) extraction of the portions from MRI data, (2) measurement of the volumes of the portions and then displaying them, and (3) user interface for a medical doctor. In this paper we describe procedures for the above. For extraction of the brain portions we propose the method based on standard region growing algorithm and the method of figure decomposition using the distance value. Comparison of the volumes of our extracted portions with volumes manually measured by a physician shows that the error rate, on the average, is 1.74% for 48 MRI data.
