It has been mandatory to label five allergenic substances (AS; egg, milk, wheat, buckwheat and peanut) in all processed foods, since April 2002 in Japan. Two kinds of ELISA kits have been provided as screening test kits for the Japanese official method. The kits have many advantages but some disadvantages, i.e., the kits are not necessarily suitable for daily monitoring in food manufacturing plants, because they require various analytical equipments and the use of complicated procedures. To overcome these drawbacks, we have developed other diagnostic kits based on immunochromatography that should enable more rapid and simple screening for food allergens. Then we examined the performance of these immunochromatographic test kits (IC kits) in terms of sensitivity, repeatability and cross-reactivity to AS proteins in 11 kinds of food models with various heating conditions and physical properties. We also examined processed food models including AS protein of constant concentration, using the IC kits and ELISA kits, and compared the results. The IC kits detected AS proteins at 5 microg/g in the extracts from processed food models, and provided highly reproducible results. Cross-reactivity among the AS proteins was not observed. The results obtained using the IC kits showed performance equivalent to that of the ELISA kits we examined in unheating processed food models including AS proteins of constant concentration. The IC kits should be more suitable for daily monitoring in food manufacturing plants.  相似文献

Utilizing the Escherichia coli/pGex vector expression system incorporating a thrombin cleavage site, full-length (residues -6-243) and truncated forms of proapolipoprotein AI (proapoAI), terminating at amino acid residues 222, 210, 150, and 135, were purified to levels of at least 5 mg/L, after thrombin cleavage. Assessed by circular dichroism, the helical contents of L-alpha-dimyristoylphosphatidylcholine-associated forms of human plasma-derived apolipoprotein AI (apoAI) and recombinant proapoAI were comparable, being 69% and 65%, respectively. Circular dichroism measurements of the lipid-associated complexes of the truncated forms showed that between the sequence of residues 150-222 no additional helicity was gained until the carboxyl-terminal sequence was present in the molecule, indicating that the carboxyl terminus of the protein is required for the formation of helix within this central region. While tryptophan residues were more than 86% accessible, as assessed by iodide quenching, in the two truncated forms, proapoAI-6-135 and proapoAI-6-150, for both free and complexed protein, this figure fell to about 50% for full-length recombinant proapoAI, further indicating the influence of the carboxyl terminus on the structure of the whole protein. While cross-linking human plasma apoAI in solution with dithiobis-(succinimidyl propionate) revealed high molecular weight oligomers by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, recombinant proapoAI did not strongly form complexes larger than trimers. None of the truncated proapoAI molecules formed oligomers larger than trimers. The shortest form, proapoAI-6-135, only dimerized. Initial results from lecithin:cholesterol acyltransferase activation (apoAI peptide concentration 0.2 microM) indicated that truncation of the 21 carboxy-terminal amino acids resulted in a drop of approximately 53% in activation and 33 residues a drop of 67% relative to the full-length protein. Overall these results indicate the important influence of the carboxyl terminus on the structure of apoAI.  相似文献

The shapes of the motor domains of kinesin and ncd, which move in opposite directions along microtubules, have been investigated. Using proteins expressed in Escherichia coli, it was found that at high salt (> 200 mM) Drosophila ncd motor domain (R335-K700) and human kinesin motor domain (M1-E349) were both sufficiently monomeric to allow an accurate determination of their radii of gyration (Rg) and their molecular weights. The measured Rg values of the ncd and kinesin motor domains in D2O were 2.06 +/- 0.06 and 2.05 +/- 0.04 nm, respectively, and the molecular weights were consistent with those computed from the amino acid compositions. Fitting of the scattering curves to approximately 3.5 nm resolution showed that the ncd and kinesin motor domains can be described adequately by triaxial ellipsoids having half-axes of 1.42 +/- 0.38, 2.24 +/- 0.44, and 3.65 +/- 0.22 nm, and half-axes of 1.52 +/- 0.23, 2.00 +/- 0.25, and 3.73 +/- 0.10 nm, respectively. Both motor domains are described adequately as somewhat flattened prolate ellipsoids with a maximum dimension of approximately 7.5 nm. Thus, it appears that the overall shapes of these motor domains are not the major determinants of the directionality of their movement along microtubules.  相似文献

This study evaluated the inhibitory effects of thiazolidine derivatives on hepatitis C virus (HCV) protease and other human serine proteases. The inhibition efficacy was tested with a reversed-phase high-performance liquid chromatography (HPLC) assay system using a NS3-NS4A fusion protein as the HCV protease and a synthetic peptide substrate that mimics the NS5A-5B junction. Nine thiazolidine derivatives showed more than 50% inhibition at 50 microg/ml. The most potent derivative was RD4-6250, with 50% inhibition at a concentration of 2.3 microg/ml; this concentration was lower than those of other protease inhibitors reported previously. The most selective derivative was RD4-6205, with 50% inhibition at a concentration of 6.4 microg/ml, a lower concentration than those on other serine proteases (chymotrypsin, trypsin, plasmin, and elastase). These results suggest that the RD4-6205 skeleton is an important structure for inhibitory activity on the HCV protease NS3-NS4A.  相似文献

Summary Mechanochemical block copolymerization in heterogeneous systems of the solid poly(vinyl chloride)-styrene-sodium dodecyl sulfate aqueous solutions has been studied by ultrasonic irradiation at 65 °C. The block copolymerization of styrene was initiated by free radicals produced from the poly(vinyl chloride) particles by ultrasonic waves. The initial rate of the block copolymerization Rp was given by Rp α [Styrene] [Sodium dodecyl sulfate]1/2. Both copolymer and homopolymer were obtained. For example, when 2.506 g of the poly(vinyl chloride) particles, 24.23 g of styrene, and 54.00 g of sodium dodecyl sulfate aqueous solution (0.500 wt%) were added in the reaction system, the weight proportions of the block copolymer and polystyrene after 60 min were approximately 50 and 20%.  相似文献

The permeability of an asymmetrically oxidized poly(L-methionine) film to oxygen dissolved in water was examined. It was recognized that the permeability along an increasing gradient of oxidized methionine composition is higher than the gradient is reversed. A qualitative analysis of this result was presented in terms of the oxygen solubilities into the film and water absorbed in the film.  相似文献

Thermal cracking of tar-sand bitumens has been carried out using a two-stage pyrolysis reactor with temperature zones of 440°C and 750–800°C, respectively. Feedstocks were pyrolysed in the first stage into cracked oils, which were carried to the second stage for subsequent pyrolysis. Only 12–14 wt% of ethylene was obtained from tar-sand bitumens at the residence time of 1.2 s in the second stage, although 27 and 16 wt% were obtained from Taching and Iranian heavy vacuum residues, respectively. The tar-sand bitumens contain shorter paraffinic straight-chains and have more branched molecules than the vacuum residues of petroleum. A straight-chain paraffin index is proposed, with which a good correlation was obtained between ethylene yield and the fraction of straight-chain paraffin carbons in the heavy oil.  相似文献

Polyacetal copolymers were prepared by cationic ring‐opening copolymerizations of 1,3,5‐trioxane (TOX) with 1,3‐dioxolane (DOX), and polyacetal terpolymers were prepared by terpolymerizations of TOX, DOX, and 2‐ethylhexyl glycidyl ether (EHGE). Polyacetal polymers with three different structures such as polyacetal homopolymers, polyacetal copolymers, and polyacetal terpolymers were compared in the mechanical properties and the creep characteristics, and discussed from the view point of the polymer structure. The polyacetal copolymers and the polyacetal terpolymers were determined by 1H‐MNR measurement. About 80 mol % of DOX and EHGE amounts in feed were incorporated randomly into the each polymer. From the plots of the degree of crystallinity (Xc) versus the tensile strength, the tensile strength and crystallintiy of the polyacetal homopoymers are higher than those of the polyacetal copolymers and the polyacetal terpolymers. However, the tensile strength does not decrease linearly with a decrease in the crystallinity among the polyacetal polymers with three different structures, the polyacetal homopolymer, the polyacetal copolymers, and the polyacetal terpolymers. Creep rupture was characterized by the activation volume, υc, value in Zhurkov's equation, which can be estimated from the slope in the plots of load versus log (rupture time) at 80°C. The polyacetal polymers with higher molecular weight have larger values of the activation volume than those with lower molecular weight. When the activation volume values are compared among the polyacetal polymers with the same molecular weights, they increase in the following order: the polyacetal homopolymers < the polyacetal copolymers < polyacetal terpolymers. © 2010 Wiley Periodicals, Inc. The glass formation region, crystalline phases, second harmonic (SH) generation, and Nd:yttrium aluminum garnet (YAG) laser-induced crystallization in the Sm2O3–Bi2O3–B2O3 system were clarified. The crystalline phases of Bi4B2O9, Bi3B5O12, BiBO3, Sm x Bi1− x BO3, and SmB3O6 were formed through the usual crystallization in an electric furnace. The crystallized glasses consisting of BiBO3 and Sm x Bi1− x BO3 showed SH generations. The formation of the nonlinear optical BiB3O6 phase was not confirmed. The formation (writing) region of crystal lines consisting of Sm x Bi1− x BO3 by YAG laser irradiation was determined, in which Sm2O3 contents were∼10 mol%. The present study demonstrates that Sm2O3–Bi2O3–B2O3 glasses are promising materials for optical functional applications.  相似文献

The light scattering from the spherulites of polyethylene terephthalate grown near the glass transition temperature has been investigated. The Hv scattering profiles can be reproduced by the sum of the ideal spherulite scattering with the distribution of spherulite radius and the isotropic scattering from randomly oriented crystallites. The ratio of optical anisotropies in the isotropic scattering to the ideal spherulite scattering is obtained by the method established to eliminate the effects of the number density of spherulites and the coefficient depending on the experimental conditions. It is found that the anisotropy ratio is almost independent of the crystallization time and of temperature above 106 °C, while it is larger at a crystallization temperature of 103 °C. The spherulitic structure is discussed in terms of the anisotropy ratio.  相似文献
