The Cytosensor microphysiometer uses silicon chip technology to correlate changes in extracellular acidification rates with quantitative changes in cellular metabolism in response to ligand binding to surface receptors. This functional measure of physiology makes the Cytosensor a valuable tool in drug discovery research by allowing application of the instrument to screening of prospective pharmacologically active agents, characterizations of dose responses and structure-activity relationships, and investigation of mechanisms of action.  相似文献

This paper describes the purification and characterization of microviridin J, a newly discovered metabolite of Microcystis that causes a lethal molting disruption in Daphnia spp., upon ingestion of living cyanobacterial cells. Microviridin J consists of an acetylated chain of 13 amino acids arranged in three rings and two side chains. Unlike other known isoforms of microviridin, microviridin J contains arginine that imparts a unique solution conformation characterized by proximal hydrophobic interactions between Arg and other regions of the molecule. This eventually results in the formation and stabilization of an additional ring system. Microviridin J potently inhibits porcine trypsin, bovine chymotrypsin, and daphnid trypsin-like proteases. The activity against trypsin is most likely due to Arg and its distinctive conformational interactions. Overall, the data presented for microviridin J emphasize once again the ability of cyanobacteria to produce numerous and potent environmental toxins.  相似文献

Current theories of snow metamorphism indicate that sublimating snow crystals have rounded shapes, while growing crystals have shapes that depend on growth rates. At slow growth rates, crystals are rounded. At moderate rates, they have flat faces with rounded edges. At fast growth rates, crystals have flat faces with sharp edges, and they have hollow faces at very fast growth rates. The main growth/sublimation mechanism is thought to be by the homogeneous nucleation of new layers at or near crystal edges. It was also suggested that the equilibrium shape of snow crystals would be temperature dependent: rounded above -10.5 degrees C, and faceted below. To test these paradigms, we have performed SEM investigations of snow samples having undergone metamorphism under natural conditions, and of snow samples subjected to isothermal metamorphism at -4 degrees and -15 degrees C in the laboratory. In general, current theories predicting crystal shapes as a function of growth rates, and of whether crystals are growing or sublimating, are verified. However, the transition in equilibrium shapes from rounded to faceted at -10.5 degrees C is not observed in our isothermal experiments that reveal a predominance of rounded shapes after more than a month of metamorphism at -4 and -15 degrees C. Some small crystals with flat faces that also have sharp angles at -15 degrees C, are observed in our isothermal experiments. These faces are newly formed, and contradict current theory. Several hypotheses are proposed to explain their occurrence. One is that they are due to sublimation at emerging dislocations.  相似文献

Tree killing and saprophytic bark beetles exert important ecological and economic roles in North American spruce forests. Chemical signaling among bark beetles, and responses by associate insects such as predators and competitors, have significant effects on the population dynamics and ecology of this community. Synthetic pheromones of primary (tree killing) and secondary (saprophytic) bark beetle species and blank controls were tested using multiple funnel and lower stem flight traps in white spruce forests in Wisconsin, Michigan, and Minnesota. Six phloeophagous and four predaceous species were collected with significant attraction by the bark beetles Dryocoetes affaber, Dryocoetes autographus, and Polygraphus rufipennis, and the predatory checkered beetles (Coleoptera: Cleridae) Thanasimus dubius and Enoclerus nigrifrons. In general, trap catches to synthetic lures resembled the species composition obtained by felling trees and collecting emerging beetles in a companion study, although several species showed differing trends. Some cross attraction occurred among bark beetles and between bark beetles and predatory beetles. For example, P. rufipennis was abundant in traps baited with Dryocoetes spp. pheromones. Thanasimus dubius and E. nigrifrons were collected in significant numbers in traps baited with the pheromone of the spruce beetle (Dendroctonus rufipennis), frontalin plus -pinene. This is a new observation for E. nigrifrons. Attraction of T. dubius to the pheromones of at least three bark beetle species in the Great Lakes region, as well as to several southern and western species, reflects its role as a habitat specialist and feeding generalist. Several other important predators and competitors commonly obtained in pine forests in this region were not obtained in these spruce stands, either in response to synthetic pheromones of spruce colonizing beetles, or in host material colonized by these beetles. Potential differences in predator prey dynamics between spruce and pine ecosystems in the Great Lakes region are discussed.  相似文献

Wound healing following implantation is characterized by an acute inflammatory reaction and a subsequent reorganizing phase in which angiogenesis is involved. Endothelial cells (EC) participate in both inflammation and angiogenesis. Thus, the effects on functions of EC exerted by implanted materials could affect the progression of wound healing. The corrosion of metallic implants can cause high concentrations of heavy metal ions in the peri-implant tissues. The purpose of the present study was to test the effects of possible corrosion products on the function and viability of human EC in vitro. Long-term exposure of EC to CoCl2 and NiCl2 (3 days, 0.7 mM) leads to a decrease of cell number and changes in cellular morphology. However, the morphological changes between CoCl2- and NiCl2-treated cells differ significantly. The changed morphology of CoCl2-treated EC and the fragmented DNA pattern indicates apoptosis. Nickel-treated cells demonstrated necrosis. The activity of integrins was tested by an assay of cellular adhesion on collagen-coated surfaces. It was shown that the number of adherent cells significantly decreased upon exposure to CoCl2. Our studies suggest that induction of cell death in EC upon exposure to CoCl2 could be attributed to impaired integrin signaling, which leads to a damaged cytoskeleton and culminates in apoptosis.  相似文献

The impact of group identity on adolescent tobacco, alcohol, and marijuana use was examined through a postal survey. The study included adolescents who identified with 1 subgroup (n = 1425) as well as adolescents who identified with 2 (n = 895) or 3 (n = 339) subgroups. The results showed that identification with the pop, skate/hip-hop, techno, and hippie subgroups was associated with higher risks of substance use, whereas identification with the sporty, quiet, computer nerd, and religious subgroups was associated with lower risks. Perceived group norm mediated the group identity-substance use relationship. Furthermore, identification with multiple groups with corresponding norm increased norm-consistent substance use, whereas identification with multiple groups with opposing norms reduced normative behavior. In this contribution, a novel two-channel acoustic front-end for robust automatic speech recognition in adverse acoustic environments with nonstationary interference and reverberation is proposed. From a MISO system perspective, a statistically optimum source signal extraction scheme based on the multichannel Wiener filter (MWF) is discussed for application in noisy and underdetermined scenarios. For free-field and diffuse noise conditions, this optimum scheme reduces to a Delay & Sum beamformer followed by a single-channel Wiener postfilter. Scenarios with multiple simultaneously interfering sources and background noise are usually modeled by a diffuse noise field. However, in reality, the free-field assumption is very weak because of the reverberant nature of acoustic environments. Therefore, we propose to estimate this simplified MWF solution in each frequency bin separately to cope with reverberation. We show that this approach can very efficiently be realized by the combination of a blocking matrix based on semi-blind source separation (‘directional BSS’), which provides a continuously updated reference of all undesired noise and interference components separated from the desired source and its reflections, and a single-channel Wiener postfilter. Moreover, it is shown, how the obtained reference signal of all undesired components can efficiently be used to realize the Wiener postfilter, and at the same time, generalizes well-known postfilter realizations. The proposed front-end and its integration into an automatic speech recognition (ASR) system are analyzed and evaluated in noisy living-room-like environments according to the PASCAL CHiME challenge. A comparison to a simplified front-end based on a free-field assumption shows that the introduced system substantially improves the speech quality and the recognition performance under the considered adverse conditions.  相似文献

OctoMap: an efficient probabilistic 3D mapping framework based on octrees   总被引:1,自引:0,他引:1  
Three-dimensional models provide a volumetric representation of space which is important for a variety of robotic applications including flying robots and robots that are equipped with manipulators. In this paper, we present an open-source framework to generate volumetric 3D environment models. Our mapping approach is based on octrees and uses probabilistic occupancy estimation. It explicitly represents not only occupied space, but also free and unknown areas. Furthermore, we propose an octree map compression method that keeps the 3D models compact. Our framework is available as an open-source C++ library and has already been successfully applied in several robotics projects. We present a series of experimental results carried out with real robots and on publicly available real-world datasets. The results demonstrate that our approach is able to update the representation efficiently and models the data consistently while keeping the memory requirement at a minimum.  相似文献
