Reconstitution of Monocyte Subsets and PD-L1 Expression but Not T Cell PD-1 Expression in Obstructive Sleep Apnea Patients upon PAP Therapy

Obstructive sleep apnea (OSA) is characterized by nocturnal breathing intermissions resulting in oxidative stress and eventually, a low-grade systemic inflammation. The study aimed to investigate the impact of positive airway pressure (PAP) therapy on the inflammatory milieu as measured by monocyte and T cell phenotypic alterations. Participants were assessed for their OSA severity before PAP therapy and about six months later, including patient-reported outcome and therapy usage by telemetry readout. The distributions of the CD14/CD16-characterized monocyte subsets as well as the CD4/CD8-characterized effector T cell subsets with regard to their PD-1 and PD-L1 expression were analyzed by flow cytometry from blood samples. Data of 25 patients revealed a significant reconstitution of the monocyte subset distribution and a decrease in PD-L1 expression on pan-monocytes and CD8⁺ T cells without an association to initial AHI and overweight. The PD-1 expression was still increased on T cell subsets, especially on CD4⁺ TH17/22 cells. We conclude that PAP therapy might have a rapid effect on the monocyte phenotype and overall PD-L1 expression levels. However, T cell immune alterations especially on TH17/22 cells persist longer, indicating an ongoing disturbance of the adaptive immune system.     

EuroPriSe Trust Certification

Trust marks are well known arbiters of trust. To be successful, sustainable and trustworthy the underlying certification scheme ought to implement specific requirements including transparent criteria and procedures verifying the actual trustworthiness of the recipient by an independent certification authority. The concept of an explicit trust certification scheme is set forth in this article using the example of EuroPriSe, the European Privacy Seal for IT products and IT-based services.     

The development of language and abstract concepts: The case of natural number.

These experiments probed the natural number words and concepts of 3-year-old children who can recite number words to ten but who can comprehend only one or two. Children correctly judged that a set labeled eight retains this label if it is unchanged, that it is not also four, and that eight is more than two. In contrast, children failed to judge that a set of 8 objects is better labeled by eight than by four, that eight is more than four, that eight continues to apply to a set whose members are rearranged, or that eight ceases to apply if the set is increased by 1, doubled, or halved. The latter errors contrast with children's correct application of words for the smallest numbers. These findings suggest that children interpret number words by relating them to 2 distinct preverbal systems that capture only limited numerical information. In discussing problems of genetic engineering animal ethicists frequently use the concept of animal integrity besides the concept of dignity. Integrity in general refers to the physical aspect of bodily intactness. But beyond that, the integrity of an animal comprises also a dynamic and an active-directed aspect. Within the framework of the integrity approach no fundamental difference can be found between genetic modification of farm animals by means of genetic engineering or selective breeding. Nevertheless genetic engineering is of particular importance for animal ethics because it evokes strong public reactions that may lead to a change in our attitude towards animals in our care. In the following text I will argue for this thesis using the example of genetic reduction of an animal’s essential capacities.     

Home Dissolution: What Happens After Separation?

Building a home and creating a family are highly interconnected processes. So what happens to the home when people separate or divorce? This paper addresses this question using both a quantitative and a qualitative approach. Based on an extensive database with socio-economic background data of 42 000 separated Danish couples in 2002, the housing careers of both partners were followed to see how the housing situation of different types of people was affected by the separation. In the qualitative approach, nine interviews with couples who had parted shed light on the emotions and practical problems of dissolving a home: how to decide who should stay? Who should move? What was it like to stay alone in, or to leave, the matrimonial home?     

The Energy Concept Adviser—A tool to improve energy efficiency in educational buildings

Educational buildings such as kindergartens, schools and universities display many similar design, operation and maintenance features in most countries. The two most noteworthy similarities amongst these building types are the high energy consumption and the necessity for retrofitting many buildings within this sector. However, studies have shown that during retrofit, energy saving measures are only rarely applied, because the decision-makers lack knowledge of investments and the efficiency of potential energy saving measures. The main goal of the International Energy Agency ECBCS Annex 36 is to provide the educational building decision-makers with sufficient data, information and tools to improve their learning and teaching environments by improving the energy efficiency of their buildings.     

Pessimistic cost-sensitive active learning of decision trees for profit maximizing targeting campaigns

In business applications such as direct marketing, decision-makers are required to choose the action which best maximizes a utility function. Cost-sensitive learning methods can help them achieve this goal. In this paper, we introduce Pessimistic Active Learning (PAL). PAL employs a novel pessimistic measure, which relies on confidence intervals and is used to balance the exploration/exploitation trade-off. In order to acquire an initial sample of labeled data, PAL applies orthogonal arrays of fractional factorial design. PAL was tested on ten datasets using a decision tree inducer. A comparison of these results to those of other methods indicates PAL’s superiority.
