This paper offers a set of sociotechnical principles to guide system design, and some consideration of the role of principles of this kind. The principles extend earlier formulations by Cherns (1976, Human Relations, 29, 783-792; 1987, Human Relations, 40, 153-162). They are intended to apply to the design of new systems, including those incorporating new information technologies and a range of modern management practices and ways of working. They attempt to provide a more integrated perspective than is apparent in existing formulations. The principles are of three broad types: meta, content and process, though they are highly interrelated. They are for use by system managers, users and designers, and by technologists and social scientists. They offer ideas for debate and provide devices through which detailed design discussions can be elaborated. The principles are most likely to be effective if they are relatively freestanding, but supported by relevant methods and tools. The principles are necessary but not sufficient to make a substantial contribution to design practice. 相似文献

Responses of registered Jersey cows to various levels of inbreeding were examined with pedigree data supplied by the American Jersey Cattle Association and test-day production data from 1970 through 1998 obtained from the Animal Breeding Center at Cornell University. Rate of increase in level of inbreeding is accelerating with time, making it more difficult for producers to make matings that avoid the potentially deleterious effects of inbreeding. Production losses caused by inbreeding were significant and curvilinear for all traits studied except somatic cell linear score, with the greatest losses at higher levels of inbreeding. Inbreeding was found to have the greatest effect on production at early ages and early in lactation. Early onset of the deleterious effects of inbreeding resulted in larger net present value losses than if effects of inbreeding occurred later in the life of an animal. Losses were probably enhanced because of the need to freshen animals as early as possible to maximize net present value returns. Survival decreased as level of inbreeding increased and was likely to have a greater negative impact on the financial health of the dairy enterprise than production losses. 相似文献

Reliable methods are required for the detection and enumeration of potentially injured E. coli O157 in food in the presence of outnumbering competing bacteria. Selective agents can prevent or inhibit the recovery and subsequent multiplication of injured cells and direct inoculation, either into selective enrichment broths or onto selective agar plates is still used in many methods for E. coli O157 detection and enumeration. When compared with tryptone soya agar (TSA), sorbitol MacConkey agar (SMAC) was shown to underestimate the concentration of viable E. coli O157:H7 subjected to low pH and high NaCl concentration. Using a resuscitation stage on TSA followed by membrane transfer to SMAC improved recovery to levels obtained on TSA. The membrane method was used to monitor the numbers of artificially contaminated E. coli O157:H7 during the fermentation of a meat product and demonstrated better survival when compared to counts on SMAC. Six rapid methods for the detection of E. coli O157 in food (BAX E. coli O157, Reveal 8 E. coli O157-H7 screening test, VIP EHEC, VIDAS E. coli O157 (ECO), EHEC-Tek and Tecra E. coli O157 visual immunoassay), were evaluated using beetburgers, parsley and fermented meat artificially contaminated with injured cells. Methods using direct selective enrichment, with or without an elevated incubation temperature gave false-negative results. A flow injection (FI) system with a microcolumn of anion exchanger has been used to effect rapid on-line separation of bromate and bromide prior to quantitation by ICP mass spectrometry. Basic performance studies are described including the effect of key FI parameters, i.e., sample injection volume, carrier stream flow rate, and eluent concentration on system response. The new approach permitted ultratrace determinations of bromate in drinking waters, the main benefits being low limit of detection (0.13 microg/L based on a 500-microL sample injection), rapid analysis time (10 min/sample), and good precision (2.8% at the 5 microg/L level). Isotopic analysis of uranium in tree bark by ICP mass spectrometry is proposed as a new measurement strategy for monitoring airborne contamination and for discrimination of nuclear and nonnuclear emission sources. A quadrupole-based ICP mass spectrometer equipped with a microconcentric nebulizer and membrane desolvator was used to provide high-sensitivity measurement. The limit of detection for uranium (238U) was 0.004 ng L(-1). Measurement precision (235U/238U) was between 0.2 and 0.5% RSD for isotopic SRMs (U005 and U015; concentration, 1 microg L(-1)) and ranged from 0.4 to 3.1% RSD for tree bark extracts (U concentration, 0.03-0.08 microg L(-1)). Bark samples collected from the Peak District National Park in Derbyshire (U.K.) exhibited a natural 235U/238U isotope ratio value (0.0072) whereas samples from Sellafield, West Cumbria (U.K.) showed depletion in 235U (235U/238U = 0.0053-0.0064). 相似文献

A macrophage plays an important role in mediating the inflammatory response. Cytokines released by activated macrophages contribute to inflammation in glomerulonephritis (GN). Clodronate, a biphosphonate, causes macrophage depletion when administered in an encapsulated form in liposomes. We used albumin as the polymer matrix to microencapsulate clodronate to the microspheres (MS) in the 1-micron size range. The purpose of this study was to (a) determine macrophage depletion by clodronate MS, (b) determine the effect of clodronate MS on endotoxin-induced cytokine release in vitro, and (c) assess the effect of clodronate MS on macrophage infiltration in experimental antiglomerular basement membrane nephritis. Macrophage depletion by clodronate MS was assessed by staining for the EDI marker. The results indicate greater than 95% depletion of macrophages from the spleen, liver, kidney, and blood. In the whole blood model, clodronate MS attenuated endotoxin-induced tumor necrosis factor alpha (TNF-alpha) and interleukin-1 beta (IL-1 beta) release, and the attenuation by the microencapsulated form of clodronate was also more effective than the free (solution) form of clodronate. Macrophage infiltration into the glomerulus in experimental GN was also blocked very effectively by pretreatment with clodronate MS. The continuing need for in-situ measurements of physical properties of wastes contained within many high level radioactive waste tanks within the Hanford Site has initiated experimental and theoretical investigations of candidate measurement methods. This paper describes experiments performed with acoustic waveguide sensors. This technology has potential application at the Hanford Site for in-situ measurements of density, viscosity, and temperature of liquid wastes. Waveguides of both circular and rectangular geometry were used in these studies for determination of the densities and viscosities of various fluids. The flight time of a torsional pulse through the sensing region of the waveguide forms the measured quantity. The flight time depends on the velocity of the wave through the sensing region of the waveguide, and this velocity in turn depends upon the properties of the fluid in contact with the waveguide. We performed experiments with 15 different fluids, most of which were single-phase Newtonian fluids. However, three of the fluids were particle-liquid mixtures, and one of these Newtonian in behavior. Most of the wastes held in Hanford tanks contain high solids content. The results of our experiments showed that acoustic waveguides were well suited for measurements in most Newtonian fluids, in agreement with earlier research presented in the literature. However, results for two-phase Newtonian fluids containing particles indicate that, in our case, the waveguides responded primarily to the background fluid rather than the mixture. Very poor results were obtained with the non-Newtonian fluid. In addition, there was a class of fluids, which serve the community as viscosity standards, for which viscosities determined with torsional waveguides were in disagreement with viscosities obtained with standard viscometers. 相似文献
