Thermal degradation and generation of objectionable compounds such as hydroxymethylfurfural have been observed in hot pressurised water extraction of polyphenols. Addition of small amounts of ethanol can reduce process temperatures and retain extraction efficiencies. However, ethanol may reduce the recovery of polyphenols in the subsequent purification stage. The overall effect of increasing amounts of ethanol (0–15%) and reduced temperatures (90, 75, 60 °C) in the extraction stage, as well as increasing amounts of ethanol in the desorption eluent (60–80%) were analysed in a combined hot pressurised liquid extraction–resin purification grape pomace polyphenols process for the first time. Without decreasing the polyphenols recovery (~24 mg GAE g?1), ethanol addition (15%) reduced the extraction temperature (from 130 to 90 °C) which not only decreased the reducing sugar extraction (~25%) but also avoided the hydroxymethylfurfural generation in the extraction stage. Additionally, the combined hot pressurised liquid extraction–resin purification process preserved the proanthocyanidin oligomeric distribution and was selective to polyphenols. The operating conditions found for the combined process, efficiently produced purified polyphenols extracts (11 mg GAE g?1), free of hydroxymethylfurfural and reducing sugars.  相似文献

Zinc is a necessary component in the action and structural stability of many enzymes. Some of them are well characterized, but in others, Zn stoichiometry and its association is not known. PIXE has been proven to be a suitable technique for analyzing metallic proteins embedded in electrophoresis gels. In this study, PIXE has been used to investigate the Zn content of enzymes that are known to carry Zn atoms. These include the carbonic anhydrase, an enzyme well characterized by other methods and the cytoplasmic pyrophosphatase of Rhodospirillum rubrum that is known to require Zn to be stable but not how many metal ions are involved or how they are bound to the enzyme. Native proteins have been purified by polyacrylamide gel electrophoresis and direct identification and quantification of Zn in the gel bands was performed with an external proton beam of 3.7 MeV energy.  相似文献

The aim of this paper is to illustrate the similarities and variations between friction and wear data acquired with two different types of laboratory tribological test equipment that not only operate in the gap between macro‐ and nano‐testing, but also provide test parameters that allow an overlap across the whole scale from macro‐ to nanoscale friction and wear tests. Comparative wear tests were conducted under identical test conditions in the overlap region between macro‐ and micro‐regions using an existing piece of laboratory fretting test equipment and a novel one. The experimental test data obtained with these two experimental set‐ups are discussed.  相似文献

The effect of two different inoculum pretreatments, thermal and cell wash-out (A1 and A2, respectively) on the performance of anaerobic fluidized bed reactors for hydrogen production was determined. The reactors were operated for 112 days under the same operational conditions using glucose as substrate at increasing organic loading rates and decreasing hydraulic retention times. Both treatments were effective avoiding methanogenesis. Reactor A2 showed better performance and stability than reactor A1 in each one of the different operational conditions. Cell wash-out treatment produced higher hydrogen volumetric production rates and yields than thermal treatment (7 L H2/L-d, 3.5 mol H2/mol hexose, respectively). DGGE analysis revealed that the microbial communities developed were affected by the inoculum treatment. Organisms from the genera Clostridium and Lactobacillus predominated in both reactors, with their relative abundances linked to hydrogen production. Resilience was observed in both reactors after a period of starvation.  相似文献

The present work studies the effect of reinforcing additives of submicron-size silicon carbide and thermal treatment on fretting wear of nickel-phosphorus (NiP) electroplated coatings. The tests are conducted under a 500-μm shear of the friction contact. The tribotests show that all coatings under study undergo abrasive-oxidative wear. Thermal treatment is found to reduce the friction coefficient and wear rate of the coatings under fretting, whereas the increased content of the SiC additive leads to increased friction coefficient and wear rate. The annealed NiP coatings have a lower wear rate compared to the composite NiP-SiC coatings.  相似文献

Alumina dispersed FeCuAl-based nanostructured cermet coatings were deposited from nanostructured powders by atmospheric plasma spraying on low carbon steel substrates. Nanostructuring was retained in the deposited coatings which exhibit up to four distinctive phases as revealed by electron microscopy. In this study, the friction behavior of the distinctive phases at nano-normal load scale was investigated alongside their contribution to the overall friction behavior at macro-normal load scale. Friction behavior at nano-normal load scale was investigated by lateral force microscopy, whereas conventional tribometers were used for investigations at micro and macro-normal loads. It appeared that, the friction measured at nano-normal loads on individual phases is dictated by both composition and hardness of the corresponding phases, and thus influences the overall friction behavior of the coatings at macro-normal loads. Moreover, the coefficient of friction at macro-normal loads differs from the one at nano-normal loads, and deviates from Amonton’s friction law.  相似文献

An important element in melanoma vaccine construction is to identify peptides from melanoma-associated Ags that have immunogenic potential in humans and are recognized by CD8+ T cells in vivo. To identify such peptides, we evaluated HLA-A*02+ melanoma patients immunized to a polyvalent vaccine containing multiple Ags, including MAGE-3, Melan-A/MART-1, gp100, tyrosinase, melanocortin receptor (MC1R), and dopachrome tautomerase (TRP-2). Using a filter spot assay, we measured peripheral blood CD8+ T cell responses, before and after immunization, to a panel of 45 HLA-A*0201-restricted peptides derived from these Ags. The peptides were selected for immunogenic potential based on their strong binding affinity in vitro to HLA-A*0201. Vaccine treatment induced peptide-specific CD8+ T cell responses to 22 (47.8%) of the peptides. The most striking finding was the HLA-independent heterogeneity of responses to both peptides and Ags. All responding patients reacted to different combination of peptides and Ags even though the responding patients were all A*0201+ and the peptides were all A*0201-restricted. From 9 to 27% of patients developed a CD8+ T cell response to at least one peptide from each Ag, but no more than 3 (14%) reacted to the same peptide from the same Ag. This heterogeneity of responses to individual peptides and Ags in patients with the same haplotype points to the need to construct vaccines of multiple peptides or Ags to maximize the proportion of responding patients.  相似文献
