Tissue engineering strategies promote bone regeneration for large bone defects by stimulating the osteogenesis route via intramembranous ossification in engineered grafts, which upon implantation are frequently constrained by insufficient integration and functional anastomosis of vasculature from the host tissue. In this study, we developed a hybrid biomaterial incorporating decellularized cartilage extracellular matrix (CD-ECM) as a template and silk fibroin (SF) as a carrier to assess the bone regeneration capacity of bone marrow-derived mesenchymal stem cells (hBMSC’s) via the endochondral ossification (ECO) route. hBMSC’s were primed two weeks for chondrogenesis, followed by six weeks for hypertrophy onto hybrid CD-ECM/SF or SF alone scaffolds and evaluated for the mineralized matrix formation in vitro. Calcium deposition biochemically determined increased significantly from 4-8 weeks in both SF and CD-ECM/SF constructs, and retention of sGAG’s were observed only in CD-ECM/SF constructs. SEM/EDX revealed calcium and phosphate crystal localization by hBMSC’s under all conditions. Compressive modulus reached a maximum of 40 KPa after eight weeks of hypertrophic induction. μCT scanning at eight weeks indicated a cloud of denser minerals in groups after hypertrophic induction in CD-ECM/SF constructs than SF constructs. Gene expression by RT-qPCR revealed that hBMSC’s expressed hypertrophic markers VEGF, COL10, RUNX2, but the absence of early hypertrophic marker ChM1 and later hypertrophic marker TSBS1 and the presence of osteogenic markers ALPL, IBSP, OSX under all conditions. Our data indicate a new method to prime hBMSC’S into the late hypertrophic stage in vitro in mechanically stable constructs for ECO-mediated bone tissue regeneration.  相似文献

High‐pressure forming of metal sheets is an innovative forming technology for the production of complex components and offers high potentials to improve the properties and qualities of sheet metal parts. This report describes investigations of a newly developed active‐elastic tool system referred to as ACTEC system. Unlike the use of a comparable semi‐rigid tool system, the ACTEC system shows improvements with respect to the material flow in the flange area and reduced sheet thinning in critical corner regions of the workpiece. In addition, the clamping forces respectively sealing forces necessary to avoid leakage in the tool system during the forming process can be reduced. Moreover, the specific design of the ACTEC‐system as well as current experimental examinations are presented and discussed.  相似文献

We present a solution for the real-time simulation of artificial environments containing cognitive and hierarchically organized agents at constant rendering framerates. We introduce a level-of-detail concept to behavioral modeling, where agents populating the world can be both reactive and proactive. The disposable time per rendered frame for behavioral simulation is variable and determines the complexity of the presented behavior. A special scheduling algorithm distributes this time to the agents depending on their level-of-detail such that visible and nearby agents get more time than invisible or distant agents. This allows for smooth transitions between reactive and proactive behavior. The time available per agent influences the proactive behavior, which becomes more sophisticated because it can spend time anticipating future situations. Additionally, we exploit the use of hierarchies within groups of agents that allow for different levels of control. We show that our approach is well-suited for simulating environments with up to several hundred agents with reasonable response times and the behavior adapts to the current viewpoint.  相似文献

Peer selection for query routing is a core task in peer-to-peer networks. Unstructured peer-to-peer systems (like Gnutella) ignore this problem, leading to an abundance of network traffic. Structured peer-to-peer systems (like Chord) enforce a particular, global way of distributing data among the peers in order to solve this problem, but then encounter problems of network volatility and conflicts with the autonomy of the peer data management. In this paper, we propose a new mechanism, INGA, which is based on the observation that query routing in social networks is made possible by locally available knowledge about the expertise of neighbors and a semantics-based peer selection function. We validate INGA by simulation experiments with different data sets. We compare INGA with competing peer selection mechanisms on resulting parameters like recall, message gain or number of messages produced  相似文献

The richness and expressive power of geometric constraints causes unintended ambiguities and inconsistencies during their solution or realization. For example, geometric constraint problems may turn out to be overconstrained requiring the user to delete one or more of the input constraints, and the solutions must then be dynamically updated. Without proper guidance by the constraint solver, the user must have profound insight into the mathematical nature of constraint systems and understand the internals of the solver algorithm. But a general user is most likely unfamiliar with those problems, so that the required interaction with the constraint solver may well be beyond the user's ability. In this paper, we present strategies and techniques to empower the user to deal effectively with the overconstraint problem while not requiring him or her to become an expert in the mathematics of constraint solving.We formulate this problem as a series of formal requirements that gel with other essentials of constraint solvers. We then give algorithmic solutions that are both general and efficient (running time typically linear in the number of relevant constraints).  相似文献

Estimation of Surface Flow and Net Heat Flux from Infrared Image Sequences   总被引:1,自引:0,他引:1  
The study of dynamical processes at the sea surface interface using infrared image sequence analysis has gained tremendous popularity in recent years. Heat is transferred by similar transport mechanisms as gases relevant to global climatic changes. These similarities lead to the use of infrared cameras to remotely visualize and quantitatively estimate parameters of the underlying processes. Relevant parameters that provide important evidence about the models of air-sea gas transfer are the temperature difference across the thermal sub layer, the probability density function of surface renewal and the flow field at the surface. Being a driving force in air sea interactions, it is of equal importance to measure heat fluxes. In this paper we will present algorithms to measure the above parameters of air-sea gas transfer during night-time and show how to combine physical modeling and quantitative digital image processing algorithms to identify transport models. The image processing routines rely on an extension of optical flow computations to incorporate brightness changes in a total least squares (TLS) framework. Statistical methods are employed to support a model of gas transfer and estimate its parameters. Measurements in a laboratory environment were conducted and results verified with ground truth data gained from traditional measurement techniques.  相似文献

For almost two decades, layer manufacturing techniques have assisted the product development at various stages from the operational point of view. Laser sintering is a powder-based layer manufacturing technology. Since the beginning of the 1990s, laser sintering of ceramic materials has been investigated academically. Since laser sintered bodies are porous correlating with a comparatively low strength, industrial use is still limited. This paper provides a knowledge base for laser sintering of ceramic suspensions. Therefore a model based on system analysis is generated, which is sub-divided into a drying and a process model. Furthermore experimental investigations with different high-performance oxide ceramic slurries (e.g., zirconia) are conducted in order to approve the estimations of the model.  相似文献

Scratch and indentation tests were performed on a polyoxymethylene homopolymer that was molded using three different mold temperatures. The different microstructures that developed during processing were studied by examining two samples for each mold temperature at different depths. During the scratch tests, the normal and the tangential force were recorded. The tests on the “as-molded” surface, in comparison to those that were polished until 1 mm of the material was removed, showed extrema of the normal force at a scratch depth that correspond to the interface of different regimes in the microstructure. The first extremum was considered to be caused by the breaking of the skin layer of the polymer (as a result of the rapid cooling process in the mold). Further extrema correspond to the permeation of the transition zone of the indenter into the spherulitic core of the sample. Indentation tests that were made on the same samples showed that the skin had a slightly lower hardness than the bulk of the polymer and that the difference in hardness decreases with increasing mold temperature. © 1997 John Wiley & Sons, Inc. The performance of organic light‐emitting devices (OLEDs) is strongly influenced by the electronic properties of the employed materials. In order to determine the effect of these materials' parameters, several different hole‐transporting 1,3,5‐tris(4‐diphenylaminophenyl)benzenes (TDAPBs) were synthesised. These TDAPBs contained different substituents, different numbers of substituents and different positions of theses substituents. For the evaluation of the electronic properties, cyclic voltammetry was employed in order to determine the HOMO values, and time‐of‐flight (TOF) measurements to obtain the hole mobilities. OLEDs were prepared consisting of the TDAPBs blended in a polymer matrix, and of Alq3 as electron‐conducting and light‐emitting layer. These devices were investigated regarding their current density/voltage characteristics, efficiencies, onset voltages for electroluminescence, and lifetimes. For hole‐transporting blend systems an exponential relationship between the current density and the HOMO levels of the TDAPBs was found. However, even though the HOMO values cover a range from −5.09 to −5.35 eV, no effects on the performance of the OLEDs were detected for electroluminescent two‐layer systems.
